SEPT10Expression in Chronic Lymphocytic Leukemia. Correlation with Clinical and Biological Prognostic Factors

Chronic lymphocytic leukemia (CLL) is characterized by a highly variable clinical course. Microarray studies allowed highlight genes differentially expressed in this pathology. In this study, we have evaluated the prognostic significance of SEPT10 expression in CLL patients. Results were correlated with immunoglobulin heavy-chain variable (IGHV) genes mutational status, genomic rearrangements and clinical parameters. SEPT10 mRNA levels were determined by quantitative real-time PCR in 70 newly diagnosed CLL patients consecutively referred to our Institution. A wide heterogeneity for SEPT10 expression was found. Gene upregulation was observed in 18.5% of cases. The univariate analysis showed a positive association between gen expression and platelet count (p 0.0001) and a negative correlation with hemoglobin levels (p = 0.0094). Although no significant differences were observed, mean treatment free survival was shorter in patients with high expression (31 months) with respect to those with low mRNA levels (72 months). Cases with abnormal karyotypes had increased expression compared to those with normal karyotypes and no association between gene expression and FISH (fluorescence in situ hybridization) risk groups and IGHV mutational status was found. Cases using IGHV3-23 gene rearrangement had low SEPT10 expression. Our results showed an association between SEPT10 expression and features of adverse outcome but without independent prognostic value. The study of SEPT10 expression may be important for a better understanding of disease heterogeneity, adding further information to those provided by established prognostic factors.

Quorum sensing(QS) is cell communication that is widely used by bacterial pathogens to coordinate the expression of several collective traits, including the production of multiple virulence factors, biofilm formation, and swarming motility once a population threshold is reached. Several lines of evidence indicate that QS enhances virulence of bacterial pathogens in animal models as well as in human infections; however, its relative importance for bacterial pathogenesis is still incomplete. In this review, we discuss the present evidence from in vitro and in vivo experiments in animal models, as well as from clinical studies, that link QS systems with human infections. We focus on two major QS bacterial models, the opportunistic Gram negative bacteria Pseudomonas aeruginosa and the Gram positive Staphylococcus aureus, which are also two of the main agents responsible of nosocomial and wound infections. In addition, QS communication systems in other bacterial, eukaryotic pathogens, and even immune and cancer cells are also reviewed, and finally, the new approaches proposed to combat bacterial infections by the attenuation of their QS communication systems and virulence are also discussed.

Influence of GSTT1, GSTM1 and GSTP1 Polymorphisms on the Development of Breast Cancer

Introduction: Every year, more than 10 million women are diagnosed worldwide with breast cancer. Breast cancer remains the leading cause of death from malignancy in women. In Mexico about 12 Mexican women die every day from this disease. Glutathione S-transferase (GST) family consists of a group of isoenzymes involved in Phase II detoxification of xenobiotics by glutathione conjugation. There have been reports showing GST polymorphisms as a risk factor for developing cancer. Objective: Determine if the frequencies of polymorphisms of GSTT1, GSTM1 and GSTP1 are associated with breast cancer development. Materials and Methods: We studied 22 women with breast cancer and a control group of 30 women. DNA was extracted from blood. Polymorphisms of T1 and M1 were identified by multiplex PCR. The identification of GSTP1b (exon 5) and GSTP1c (exon 6), was performed separately by RFLP-PCR, using BsmAI and AciI enzymes respectively. Results: Frequencies of polymorphisms were as follows: for breast cancer group 33% null GSTT1 vs 40% null in the control, 32% null GSTM1 vs 63% of control;63% heterozygote GSTP1b P1a/P1b and 18% homozygote P1b/P1b, vs 53% heterozygote and 20% homozygote in the control, 41% heterozygote P1a/P1c GSTP1c and 0% homozygote vs 10% heterozygote P1c/P1c and 0% homozygote control. Significant differences were found between GSTM1 and GSTP1c polymorphisms with GSTM1 null percentage higher in the control group and a higher frequency of P1c heterozygotes in the cancer group. Conclusion: As this is a pilot study where the results showed no association with breast cancer, further investigation is required on the involvement of GST genes in neoplasm development.

Biodegradable magnesium alloys are promising candidates for temporary fracture fixation devices in orthopedics;nevertheless,its fast degradation rate at the initial stage after implantation remains as one of the main challenges to be ***_(2)-based coatings to reduce the degradation rate of the Mg-implants are an attractive solution since they show high biocompatibility and *** this work,the degradation,cytotoxicity,and antibacterial performance of ZrO_(2)thin films deposited by magnetron sputtering on a Mg-Zn-Ca alloy was ***-term degradation of ZrO_(2)-coated and uncoated samples was assessed considering electrochemical techniques and H_(2)evolution(gas chromatography).Additionally,long term degradation was assessed by mass-loss *** results showed that a 380 nm ZrO_(2)coating reduces the degradation rate and H_(2)evolution of the alloy during the initial 3 days after immersion but allows the degradation of the bare alloy for the *** ZrO_(2)coating does not compromise the biocompatibility of the alloy and permits better cell adhesion and proliferation of mesenchymal stem cells directly on its surface,in comparison to the bare ***,the ZrO_(2)coating prevents the adhesion and biofilm formation of ***.

Comparison of Nine Membrane Pairs for Electrodialytic Removal of Nitrate Ions

Electrodialysis (ED) of NaNO3 solutions was performed to stress the role of the membrane pair as a unit i.e. the behavior as a whole of Anion Exchange Membrane (AEM) and Cation Exchange Membrane (CEM), in the process at room temperature. The membrane pair was also tested in the reclamation of nitric acid and sodium hydroxide. It was found that the membrane pair ranking in Limiting Current Density does not agree with that of ions leakage and both rankings are not the same as for Current Efficiency or Specific Power Consumption, each parameter for a specific membrane pair depends on the associated membrane and at a lesser degree on the flow rate. Results show that one cannot select a membrane or a membrane pair only by its limiting current density i.e. it is necessary to evaluate several parameters of the process by characterizing the whole membrane pair and to decide the parameter of interest, i.e. denitrification speed, ions leakage, proton back migration, current efficiency or specific power consumption.

Differences in Browning Index and CIELAB Coordinates of the Two Grape Drying Processes, Traditional Sun-Drying and Chamber-Drying and during the Ageing of Pedro Ximenez Sweet Wine

The color parameters during the Pedro Ximenez grape raisining, as well as the sweet wine aging process from the Montilla-Moriles grapes （Andalusia, Southern Spain）, have been studied. Drying process of grapes was carried out by means of the traditional sun-drying method and an alternative chamber-drying method under 50℃. Chamber-drying allows shorter drying time and select grapes at a higher ripening degree. During raisining grape musts decreased in hab （hue angle） and increased in Cab （chroma）. In comparative terms, the final values of hue were virtually identical in both types of drying, although differences were found in the final values of chromaticity, being lower in the chamber-drying method. Changes in the color parameters during aging were compared in commercial wines with different aging systems and without aging. Likewise, as a reference of traditional wine aging system, the color changes in wines with four aging degrees were also studied. Regarding to the commercial wines studied, it can be pointed out the wine aged without blends for 4 years significantly differed in the values of hab and C＊ab of the remaining wines, which show more similar values among them and in the data obtained for the wines aged by the traditional aging system.

Evolutionary genetics of invasive species has been unexplored in Argentina. Invasive alien species (IAS) have a wide geographical distribution, characteristic life cycles and great ability to adapt, establish and spread in a new environment. Recent advances in novel molecular technologies, the use of higher resolution genetic markers, and the research development on genetic variation of invasive species consolidated the importance of genetic aspects in the invasion process. Undoubtedly, the growing concern for the disturbances generated by invasive species on biodiversity and functioning of ecosystems was also determinant for the inclusion of the Invasion Biology within the broad field of Evolutionary Biology including relevant examples that address the evolutionary genetic aspects of biological invasions. Recent studies suggest that the invasion success of many species depends on their ability to respond to natural selection. Although the number of invasive species registered in Argentina far exceeds 600, little research has been done on invasive mammal species and only five of them were hitherto genetically analyzed. Presuming that invasion genetics is incorporated into the agenda of control and management organizations, it would allow integrating the ecological, genetic, and evolutionary biology aspects for knowledge of invasive species widely distributed in Argentina. The objective of this article is to highlight the importance of evaluating the genetic structure of invasive species for their management and to inform about the invasive species of mammals that were introduced in Argentina and have been or are being analyzed genetically.

MONITORING OF FILARIAL ANTIGENS IN JIRDS AND PATIENTS AFTER TREATMENT

We developed a sandwich ELISA with monoclonal antibodies to monitor filarial antigens in animals and patients after infection and treatment. Levels of antimicrofilarial antibodies and parasite antigens were measured periodically in 40 B. malayi infected jirds. In all animals L3HC11 antigen was detected earlier than Mf ES34 antigen, while antimicrofilarial antibodies appeared much more slowly. These serologic changes preceeded the onset of patent infections. After 3 courses of treatment with DEC and N170, the levels of parasite antigen in sera and of Mf in peritoneal cavities were monitored in 23 infected jirds. In 8 jirds Mf became negative, no adult worms were found in 7 jirds and a single degenerating female worm was present in 1 jird. ES34 and HC11 were undetectable in 8/8 and 6/8 necropsy sera. Mf persisted in 11 animals, 9 jirds were necropsied, 8 contained adult worms. Detectable levels of ES34 or HC11 antigen were present in 7/9 and 8/9 from these animals. In sham-treatment, few changes were noted in control animals. Thus, parasitological findings at necropsy are correlated with the results of antigen detection assay. We analyzed serial serum samples from 32 bancroftian microfilaremia collected 1-42 months after DEC therapy. Mf resolved rapidly in all treated individuals. ES34 disappeared faster than HC11, 3 months after treatment. Levels of ES34 and HC11 antigens remained detectable or rising after treatment in 8 and 10 individuals. Four patients' Mf recurred 20-42 months after treatment. These findings show that the remaining or a rise in serum levels of antigen after therapy predicts recurrent microfilaremia in patients and additional treatment is needed.

Simultaneous Growth of Chaetoceros muelleriand Bacteria in Batch Cultures

The relationship between bacterial load and the microalga Chaetoceros muelleri was analyzed in a scale-up experiment. The microalga was grown during five days in a 0.4-L Erlenmeyer flask, 2-L Fernbach flask, 18-L Carboy and 400-L column, during which the cell density of C. muelleri, the abundance of heterotrophic bacteria, Vibrio spp., and total bacteria were determined. The highest specific growth rates (μ) of C. muelleri occurred during the first day of culture (0.88 to 2.29 d-1). Highest cell density was recorded on the fifth day at the 2-L (7.62 × 106 cells·mL-1) and 18-L (6.32 × 106 cells·mL-1), coinciding with the maximum counts of heterotrophic bacteria (16.55 × 105 and >30 × 105 CFU·mL-1, respectively). There was a high correlation (0.80, 0.75, 0.85;p?5 cells·mL-1) during the five days occurred at 18-L Carboy. The concentration of total bacteria at all levels was always higher than that of heterotrophic bacteria. The average ratio of heterotrophic to total bacteria was higher in the 2-L (0.0108) and 18-L (0.0172) cultures. The high biomass of C. muelleri and the presence of Vibrio spp. at the 18-L and 400-L levels indicate that it is necessary to establish programs to prevent diseases and economic losses caused by pathogenic bacteria in penaeid shrimp farming.

Protective Effect of Phenolic-Rich Extracts from Different Parts of Opuntia joconostleFruit against Carbon Tetrachloride-Induced Oxidative Stress in Mice

Opuntia joconostle fruit is a rich source of biocompounds such as polyphenols including gallic, vanilic, 4-hidroxybenzoic, cafeic, and syringic acids, catechin, epicatechin, rutin, and vanillin, besides betalains. The objective of this research was to evaluate the effect of supplementation polyphenols-rich extracts from different parts of Opuntia joconostle against carbon tetrachloride-induced oxidative stress in a mouse model. The animals were treated orally with polyphenols-rich extracts at 50, 100 and 200 mg/kg BW for 30 consecutive days. On day 30th the mice received carbon tetrachloride (CCl4) as hepatoxic agent. Biochemical evaluations were carried out 24 h after induction of the oxidative stress. Data showed that methanolic extracts from different parts of Opuntia joconostle exerting protective effect against the CCl4-induced oxidative stress in mice. Histology examination revealed that the damage decreased in groups treated with polyphenols-rich extracts compared to the group that did not receive any treatment. Opuntia joconostle fruit contains many phenolic compounds, flavonoids and betalains. The protective effect of extracts may be related to the phenolic composition and also by a counteraction with other compounds, such as betalains and flavonoids that increase their antioxidant effect.
