Immunohistochemical analysis of the Wnt/β-catenin signaling pathway in pancreatic neuroendocrine neoplasms

AIM: To investigate the role of the Wnt/β-catenin pathway in pancreatic neuroendocrine neoplasms(PanN ENs). METHODS: Tissue microarrays containing 88 PanN ENs were immunohistochemically labeled with antibodies to β-catenin, E-cadherin, adenomatous polyposis coli(APC), chromogranin and synaptophysin. One case had only metastatic tumors resected, whereas others(n = 87) received pancreatectomy with or without partial hepatectomy. Pathology slides, demographic, clinicopathologic, and follow up data were reviewed. Patients' demographics, clinicopathologic features, and immunohistochemical results from 87 primary tumors were compared between patients with low stage(stage Ⅰ/Ⅱ) and high stage(stage Ⅲ/Ⅳ) tumors. In addition, correlation of immunohistochemical results from primary tumors with disease-specific survival(DSS) was evaluated. RESULTS: Strong membranous β-catenin staining in the primary tumor was observed in all 13 stage Ⅲ/Ⅳ Pan NENs as compared to 47%(35/74) of stage Ⅰ/Ⅱtumors(P branous β-catenin staining was unassociated with tumor grade or DSS. Decreased membranous β-catenin staining was associated with decreased membranous E-cadherin labeling. Nuclear β-catenin staining was seen in 15%(2/13) of stage Ⅲ/Ⅳ Pan NENs as compared to 0%(0/74) of stage Ⅰ/Ⅱ tumors(P = 0.02). The case with metastasectomy only also showed nuclear β-catenin staining. Two of the three cases with nuclear β-catenin staining were familial adenomatous polyposis(FAP) patients. Lack of APC expression was seen in 70%(57/81) of the cases, including the 3 cases with nuclear β-catenin staining. Expression of E-cadherin and APC in primary tumor was not correlated with tumor grade, tumor stage, or disease specific survival. CONCLUSION: The Wnt/β-catenin pathway was altered in some PanN ENs, but did not Impact DSS. PanN ENs in FAP patients demonstrated nuclear β-catenin accumulation and loss of APC.

Estimated binding Energies of Drug-Like and Nondrug-Like Molecules in the Active Site of HIV-1 Integrase, ***, and Two Mutant Models: Y143R and N155H

Lipinski’s “Rule of Five” was introduced for predicting oral bioavailability to describe drug-like molecules. For the purpose of this research the rules were used to separate potential inhibitors of HIV-1 integrase (***) into two groups: drug-like and nondrug-like. If one of Lipinski’s “Rule of Five” was not followed the potential inhibitor was classified as nondrug-like. Thirty molecules were identified from the literature, twenty-four drug-like and six nondrug-like, that were docked into the active site of *** (considered the non-mutated protein) and two mutant models, Y143R and N155H. These are two of the mutations that have led to increased resistance to HIV-1 integrase drugs such as raltegravir and elvitegravir. The computational software, ICM-Pro (Molsoft L.L.C.), was used to determine the estimated binding energy (EbE) of the drug/protein complex. It was found that the nondrug-like molecules generally had a more negative EbE, that is, tighter binding with 1bIS. pdb, though there were several exceptions in the drug-like group. With the protein mutant model Y143R, the majority of drug-like (58%) and nondrug-like molecules (67%) had tighter binding. However, for the mutant model N155H, there was the same percent (46%) of drug-like molecules with tighter binding with the mutant model as with ***. The drug-like molecules were used when there was a ≥1 kcal/mole difference between *** and either of the two mutant models to suggest a pharmacophore with structural characteristics for an HIV-1 integrase inhibitor.

Iron and Folate Contents of Tajik Legumes

Seven varieties of Tajik legumes and two Tajik snack type ready-to-eat (RTE) whole/split chickpeas were analyzed for iron in raw and cooked legumes and for naturally occurring folate content in cooked legumes. Iron was measured according to AACC method 40 - 41b. Folate contents were determined by microbiological (Lactobacillus casei subsp. Rhamnosus ATCC 7469) and high-performance liquid chromatography analysis utilizing a tri-enzyme treatment (pro-tease, α-amylase and conjugase). Folate derivatives of tetrahydrofolate, 5-formyl-tetrahydrofolate and 5-methyl- -tetrahydrofolate were identified and quantified. Iron content for Tajik legumes ranged from 5.52 to 13.27 mg/100 g for raw;2.81 to 4.12 mg/100 g for cooked and 4.37 and 4.76 mg/100 g for RTE chickpeas. The total folate content of cooked legumes ranged from 53 to 81 μg/100 g for beans;133 to 203 μg/100 g for peas, and from 39 to 22 μg/100 g for small and large lentils, respectively. The predominant form of folate in legumes was tetrahydrofolate, followed by 5-formyl-tetrahydrofolate and 5-methyl-tetrahydrofolate.

Sexual Satisfaction Is Reduced in the Female Patient and Sexually Intimate Partners Following Cancer Therapy

As cancer survival has continued to improve, cancer patients and their sexually intimate partner (SIP) are confronted with a number of issues including sexual function and overall sexual health. Our study objective was to assess changes in sexual function in women undergoing cancer treatment and their SIP, and attempt to identify areas of needed support and improvement. In this questionnaire-based observational study, females (n = 11) completed a Female Sexual Function Index (FSFI) and for SIP’s (n = 11), a brief Sexual Function Inventory (bSFI). Level of satisfaction prior to and within 3 months following treatment with surgery, chemotherapy and/or radiation was compared. Mean pre-and post-treatment total (30.7 ± 2.7 vs. 23.2 ± 3.7, p brication (5.5 ± 0.4 vs. 4.5 ± 0.8, p = 0.006), orgasm (5.0 ± 0.6 vs. 3.8 ± 0.7, p = 0.001), satisfaction (5.3 ± 0.6 vs. 3.5 ± 0.9, p bSFI domain scores for level of satisfaction were significantly lower following partner treatment of SIPs (3.5 ± 5.2 vs. 2.5 ± 5.2, p = 0.004). Overall, female sexual dysfunction was identified in 9% of patients prior to cancer treatment and 91% (n = 11, p be overlooked.

Legacy Effects of Explosive Contamination on Vegetative Communities

Natural selection processes are constantly influencing vegetation community composition. In the presence of anthropogenic contaminants additional forces act as filters controlling persistence of naturally occurring species. Classical species diversity and richness metrics can miss subtle changes under disturbance regimes while species composition and functional characteristics may be able to detect them. Our study was designed to investigate legacy impacts of explosives contaminated soils in an experimental minefield on vegetative communities using ecological metrics. As hypothesized, species diversity and richness showed no change in the presence of explosive compounds while species composition provided clear separation of groups and functional trait dominance was also observed to change. Overall, presence of anthropogenic contaminates have led to community composition and functional shifts for vegetation after initial contamination. The responses in species composition and functional diversity/richness were a result of new tolerant species filling open niches in contaminated plots. More work is needed to confirm this in varied systems and in the presence of diverse contaminants.

TGF-β and Foxp3 expressions are crucial for the induction and functional activity of CD4＋Foxp3＋ regulatory T （iTreg） cells. Here, we demonstrate that although TGF-β-primed CD8＋ cells display much lower Foxp3 expression, their suppressive capacity is equivalent to that of CD4＋ iTreg cells, and both Foxp3- and Foxp3＋ CD8＋ subsets have suppressive activities in vitro and in vivo. CD8＋Foxp3- iTreg cells produce little IFN-γ but almost no IL-2, and display a typical anergic phenotype. Among phenotypic markers expressed in CD8＋Foxp3- cells, we identify CD103 expression particularly crucial for the generation and function of this subset. Moreover, IL-IO and TGF-β signals rather than cytotoxicity mediate the suppressive effect of this novel Treg population. Therefore, TGF-β can induce both CD8＋Foxp3- and CD8＋Foxp3＋ iTreg subsets, which may represent the unique immunoregulatory means to treat autoimmune and inflammatory diseases.

Effectiveness of the flipped classroom on the development of self-directed learning in nursing education:a meta-analysis

Objectives: To examine the best practice evidence of the effectiveness of the flipped classroom（FC） as a burgeoning teaching model on the development of self-directed learning in nursing *** sources: The relevant randomized controlled trial（RCT） and non-RCT comparative studies were searched from multiple electronic databases including PubMed, Embase, Web of Science, Cumulative Index to Nursing and Allied Health Literature（CINAHL）, Cochrane Central Register of Controlled Trials（CENTRAL）, Wanfang Data, China National Knowledge Infrastructure（CNKI）, and Chinese Science and Technology Periodical Database（VIP） from inception to June *** methods: The data were independently assessed and extracted for eligibility by two reviewers. The quality of included studies was assessed by another two reviewers using a standardized form and evaluated by using the Cochrane Collaboration’s risk of bias tool. The self-directed learning scores（continuous outcomes） were analyzed by using the 95% confidence intervals（Cls） with the standard deviation average（SMD） or weighted mean difference（WMD）. The heterogeneity was assessed using Cochran’s I;***: A total of 12 studies, which encompassed 1440 nursing students（intervention group = 685, control group = 755）, were eligible for inclusion in this review. Of 12 included studies, the quality level of one included study was A and of the others was b. The pooled effect size showed that compared with traditional teaching models, the FC could improve nursing students’ selfdirected learning skill, as measured by the Self-Directed Learning Readiness Scale（SDLRS）, Self-Directed Learning Readiness Scale for Nursing Education（SDLRSNE）, Self-Regulated Learning Scale（SRL）, Autonomous Learning Competencies scale（ALC）, and Competencies of Autonomous Learning of Nursing Students（CALNS）.
