Glycosylation-related gene expression in HT29-MTX-E12 cells upon infection by Helicobacter pylori

AIM To identify glycosylation-related genes in the HT29 derivative cell line, HT29-MTX-E12, showing differential expression on infection with Helicobacter pylori(H. pylori).METHODS Polarised HT29-MTX-E12 cells were infected for 24 h with H. pylori strain 26695. After infection RNA was isolated from both infected and non-infected host cells. Sufficient infections were carried out to provide triplicate samples for microarray analysis and for q RTPCR analysis. RNA was isolated and hybridised to Affymetrix arrays. Analysis of microarray data identified genes significantly differentially expressed upon infection. Genes were grouped into gene ontology functional categories. Selected genes associated with host glycan structure(glycosyltransferases, hydrolases, lectins, mucins) were validated by real-time q RT-PCR *** Infection of host cells was confirmed by the isolation of live bacteria after 24 h incubation and by PCR amplification of bacteria-specific genes from the host cell RNA. H. pylori do not survive incubation under the adopted culture conditions unless they associate with the adherent mucus layer of the host cell. Microarray analysis identified a total of 276 genes that were significantly differentially expressed(P < 0.05) upon H. pylori infection and where the fold change in expression was greater than 2. Six of these genes are involved in glycosylation-related processes. Real-time q RT-PCR demonstrated significant downregulation(1.8-fold, P < 0.05) of the mucin MUC20. REG4 was heavily expressed and significantly downregulated(3.1-fold, P < 0.05) upon infection. Gene ontology analysis was consistent with previous studies on H. pylori *** Gene expression data suggest that infection with H. pylori causes a decrease in glycan synthesis, resulting in shorter and simpler glycan structures.

Purpose:Recurrent dislocation of shoulder(RDS)is a common injury in high demand professionals,like athletes and military *** treatment for the patients with bankart lesion is the arthroscopic *** present study compares the outcomes of two different techniques of arthroscopic bankart repair i.e.a standard two anterior portals technique and a single anterior portal technique in patients with ***:Patients with traumatic RDS met the inclusion criteria were managed with bankart repair using either two anterior portals(Group A)or a single anterior portal(Group b)*** were evaluated before the intervention and at the mean follow-up of approximately two years using Rowe score,Oxford shoulder score and Tegner activity ***:The mean age of the patients in Groups A(n=34)and b(n=37)was 29.64 years and 29.05 years respectively(p=0.66).The dominant shoulder was involved in 27 patients in Group A and 22 patients in Group b(p=0.069).The operative time in Group A and b was 68.52 min and 46.35 min,respectively(pboth groups compared with the pre-operative ***,the final outcome scores were not significantly different between the both *** median Tegner's score preoperatively and at follow-up was 7 and 6,respectively in Groups A and ***:Single anterior portal technique is an effective treatment modality,yielding a similar outcome as two anterior portals technique in the management of RDS.

The impact of perioperative blood transfusions on short-term outcomes following hepatectomy

Testing for hepatitis b virus alone does not increase vaccine coverage in non-immunized persons

AIM To determine whether hepatitis b virus(HbV)-testing could serve as a gateway to vaccinate non-immunized individuals in a low-prevalent *** Non-immunized subjects participating in a multi-center, HbV-testing campaign in Paris, France were identified and contacted via telephone 3-9 mo after testing in order to determine vaccination status. Vaccination coverage was evaluated in per-protocol(for all respondents) and intent-to-treat analysis(assuming all non-responders did not vaccinate).RESULTS In total, 1215/4924(24.7%) enrolled subjects with complete HbV serology were identified as nonimmunized and eligible for analysis. There were 99/902 successfully contacted subjects who had initiated HbV vaccination after screening: per-protocol, 11.0%(95%CI: 9.0-13.2); intent-to-treat, 8.2%(95%CI: 6.7-9.8). In multivariable analysis, vaccination was more likely to be initiated in individuals originating from moderate or high HbV-endemic countries(P bout the reasons for not initiating HbV vaccination, the most frequent response was "will be vaccinated later"(33.4%), followed by "did not want to vaccinate"(29.8%), and "vaccination was not proposed by the physician"(21.5%). Sub-group analysis indicated a stark contrast in vaccination coverage across centers, ranging from 0%-56%.CONCLUSION HbV-vaccination after HbV screening was very low in this study, which appeared largely attributed to physician-patient motivation towards vaccination. Increased vaccination coverage might be achieved by emphasizing its need at the organizational level.

Although spermatozoa are formed during spermatogenesis in the testis, testicular spermatozoa are immature and cannot swim or fertilize. These critical spermatozoal functions are acquired in the epididymis where a specific luminal environment is created by the blood-epididymal barrier; proteins secreted by epididymal principal cells bind to maturing spermatozoa and regulate the maturational process of the spermatozoa. In the epididymis, epithelial cell-cell interactions are mediated by adhering junctions, necessary for cell adhesion, and by tight junctions, which form the blood-epididymal barrier. The regulation of these cellular junctions is thought to represent a key determinant in the process of sperm maturation within the epididymis. Tight junctions between adjacent principal cells permit the formation of a specific microenvironment in the lumen of the epididymis that is essential for sperm maturation. Although we have made significant progress in understanding epididymal function and the blood-epididymal barrier, using animal models, there is limited information on the human epididymis. If we are to understand the normal and pathological conditions attributable to human epididymal function, we must clearly establish the physiological, cellular and molecular regulation of the human epididymis, develop tools to characterize these functions and develop clinical strategies that will use epididymal functions to improve treatment of infertility. （Asian J Androl 2007 July; 9： 463- 475）

Retinopathy is a human eye disease that causes changes in retinal blood vessels that leads to bleed,leak fluid and vision *** of retinopathy are blurred vision,changes in color perception,red spots,and eye pain and it cannot be detected with a naked *** this paper,a new methodology based on Convolutional Neural Networks(CNN)is developed and proposed to intelligent retinopathy prediction and give a decision about the presence of retinopathy with automatic diabetic retinopathy screening with accurate *** CNN model is trained by different images of eyes that have retinopathy and those which do not have *** fully connected layers perform the classification process of the images from the dataset with the pooling layers minimize the coherence among the adjacent *** feature loss factor increases the label value to identify the patterns with the kernel-based *** performance of the proposed model is compared with the related methods of DREAM,KNN,GD-CNN and *** results show that the proposed CNN performs better.
