“Cachacas”(Sugarcane Spirit)Aged Quantitation of Phenolic Compounds,Antibacterial and Antioxidant Activity

The “cachaça” is currently one of the fastest growing agro-industrial industries in the country as the drink considered as a symbol of Brazilian nationality. This study aimed to quantify the phenolic compounds, evaluate the antioxidant and antibacterial activity of different samples of aged “cachaça”. The physic-chemical analysis was performed at the Brandy Quality Laboratory of the Federal University of Lavras, and the determination of phenolic compounds was performed by High Performance Liquid Chromatography (HPLC). The antimicrobial activity evaluation was held at the Food Mycology Laboratory through the cavity agar diffusion technique and by steam using the micro-organisms Escherichia coli, Staphylococcus aureus, Salmonella choleraesuis, Listeria monocytogenes and Pseudomonas aeruginosa. The antioxidant activity was evaluated using the DPPH radical sequestering method. The values obtained for the phenolic compounds ranged from 0.41 to 9.69 mg/L, observing the predominance of syringaldehyde, gallic acid and vanillic acid. The phenolic extracts showed a satisfactory antibacterial activity for both Gram-negative and Gram-positive but did not show inhibition against the bacteria Pseudomonas aeruginosa. The antioxidant activity was evidenced by the DPPH test for cachaças tested and the “cachaça” aged in oak barrel showed the highest activity, followed by “cachaça” aged in jequitibá, amburana and balm barrels.

The compositions of essential oils from Chenopodium ambrosioides L. and Philodendron bipinnatifidum Schott were determined, and the their potential effects on the nutrition and mortality of Diabrotica speciosa were studied. The extraction of the oils was performed by hydrodistillation (2 h) using a modified Clevenger apparatus and the oils were subsequently subjected to analysis by gas chromatography/flame ionization detector (CG/FID) and gas chromatography/mass spectrometry (GC-MS). A completely randomized design with five treatments and four replications was adopted. The bean plants were sprayed with solutions of the oils dissolved in aqueous Tween 80 solutions at concentrations of 0 (water + Tween 80), 0.5%, 1.0%, 1.5% and 2.0% and then furnished to the insects with no choice available. Seven days after the application, the percentage of leaves with injury, degree of defoliation, the preference index for consumption and the percent of mortality of insects were evaluated. Neither of the essential oils caused a reduction in foliar injury, but antifeeding activity was observed, causing reduced feeding and increasing the mortality of adult D. speciosa insects.

The essential oils from Mentha viridis L. and Mentha pulegium L. were characterized by gas chromatography coupled to mass spectrometry (GC-MS). These oils were obtained by hydrodistillation and presented linalool (40.70%), carvone (13.52%) and α-terpinene (8.56%) as the principal constituents in the essential oil from Mentha viridis L. Pulegone (50.01%), menthol (31.90%) and menthone (16.56%) were the principal constituents in the essential oil from Mentha pulegium L. These essential oils (in concentrations ranging from 3.91 to 500 μL·mL-1) showed satisfactory activities against Escherichia coli, Listeria monocytogenes, Salmonella choleraesuis and Staphylococcus aureus. The antioxidant activities with 2-deoxyribose and phosphomolybdenum and the reducing power (in concentrations ranging from 0.78 to 1000 μL·mL-1) were determined. The antioxidant activity was observed for the two oils evaluated by the phosphomolybdenum and 2-deoxyribose methods, whereas the essential oil from M. viridis presented low antioxidant activity in the reducing power assay.

The objective of this study was to characterize and verify the in vitro antitumor activity of essential oils (EOs) extracted from the leaves and flowers of Callistemon viminalis. The EOs were extracted by hydrodistillation using a modified Clevenger apparatus. The identification and quantification of constituents were performed on a gas chromatograph coupled to a mass spectrometer and a gas chromatograph with a flame ionization detector. The antitumoral activity was evaluated by a colorimetric assay (MTS) using different cell lines derived from human tumors (breast, lung, glioblastoma, and melanoma). The major constituents of the EOs of leaves and flowers were similar, only quantitative differences being observed. The compounds 1,8-cineole, α-pinene and α-terpineol were found in concentrations of 50.4%, 25.8% and 8.7% in the EOs obtained from the leaves and 48.8%, 24.5% and 3.9% in the EOs obtained from the flowers, respectively. The cytotoxic activity of the EOs was observed only in melanoma cultures (HT144). Cultures treated for 48 h with EOs from leaves and flowers (200 μg·mL-1) reduced the viability by 40% and 25%, respectively. Thus, the antiproliferative activity of the EO from leaves was more pronounced than the EO from flowers in cells derived from melanoma.

The extraction of essential oils was performed by using the technique of hydrodistillation modified Clevenger apparatus, identification and quantification of the constituents was achieved by Gas Chromatography coupled to Mass Spectrometry, and Gas Chromatography equipped with a flame ionization detector. Assessment of allelopathic activity was evaluated with the use of the method that assesses the direct contact of essential oils on germination and vigor of lettuce seeds. The major constituents that characterize the essential oil from the flowers of Callistemon viminalis were 1, 8-cineole, α-pinene and limonene at concentrations of 66.9%, 16.0% and 10.0%, respectively. The essential oil presented allelopathic activity at intensities that varied proportionately to the concentration of the essential oil, with a reduction in the percentage of germination and the germination speed index (GSI) of lettuce seeds and in the dry mass and length of shoots and roots of lettuce seedlings.

Long-term effects of gestational diabetes mellitus on the pancreas of female mouse offspring

BACKGROUND Prolonged fetal exposure to hyperglycemia may increase the risk of developing abnormal glucose metabolism and type-2 diabetes during childhood,adolescence,and adulthood;however,the mechanisms by which gestational diabetes mellitus(GDM)predisposes offspring to metabolic disorders remain unknown.AIM To quantify the nerve axons,macrophages,and vasculature in the pancreas from adult offspring born from mouse dams with GDM.METHODS GDM was induced by i.***.administration of streptozotocin(STZ)in ICR mouse dams.At 12 wk old,fasting blood glucose levels were determined in offspring.At 15 wk old,female offspring born from dams with and without GDM were sacrificed and pancreata were processed for immunohistochemistry.We quantified the density of sensory[calcitonin gene-related peptide(CGRP)]and tyrosine hydroxylase(TH)axons,blood vessels(endomucin),and macro-phages(CD68)in the splenic pancreas using confocal microscopy.RESULTS Offspring mice born from STZ-treated dams had similar body weight and blood glucose values compared to offspring born from vehicle-treated dams.However,the density of CGRP+and TH+axons,endomucin+blood vessels,and CD68+macrophages in the exocrine pancreas was significantly greater in offspring from mothers with GDM vs control offspring.Likewise,the microvasculature in the islets was significantly greater,but not the number of macrophages within the islets of offspring born from dams with GDM compared to control mice.CONCLUSION GDM induces neuronal,vascular,and inflammatory changes in the pancreas of adult progeny,which may partially explain the higher propensity for offspring of mothers with GDM to develop metabolic diseases.

A Reliability-Based Strategy for the Analysis of Single Proton Exchange Membrane Fuel Cells

The development of power conversion systems based on fuel cells has been demanding reliability studies since the requirements associated with cost and durability of these technological products have become fundamental to their acceptance by the energy market. The experimental part of the reliability study presented in this work consisted of performing life tests with single proton exchange membrane fuel cells (PEMFCs). The proposed reliability analysis methodology covered the application of qualitative and quantitative techniques. In the qualitative approach, a Failure Mode and Effect Analysis was developed in order to identify and evaluate all potential failures associated with the operation of fuel cells. In the quantitative approach, a statistical analysis was applied to the sample data generated in long-term steady-state tests of these devices. A two-parameter exponential distribution was fitted to data and the maximum likelihood estimate for the mean time to failure (MTTF) of the fuel cells was calculated. It is important to point out that the tests performed under the scope of this study were the first long-term experiments performed with the fuel cells produced in the laboratories of IPEN-CNEN/SP, Brazil. Although the results indicated that fuel cell performance and durability were still at a level below the targets normally established for similar commercial devices, the improvement of the main components of PEMFCs has been the objective of several projects developed at the institute. Thus, the main benefit brought by this study is the proposed methodology, which can be implemented as part of a reliability growth analysis of the fuel cells and can be integrated into the design process of these devices.

Simulation of Biogas Production from Solid Organic Wastes

The design and building of new alternative fuel plants is an increasing necessity to replace old technology and non-renewable fossil fuels. To optimize the performance of these plants and to obtain an economically feasible production of these types of fuels, it is necessary to have a total control of each variable involved in the process of production and how these factors affect the yield of fuel production. In this paper it is proposed a model of a digester to generate gas using a Vensim software designed to generate simulations in dynamic state. This simulation was developed using differential equations to model the behavior at each stage of the process and auxiliary conditions to complement the mathematical description of the model. The main factors in the biogas production are the retention time and the methanogen mortality ratio. For retention time lower than 10 h the process loses effectiveness due to bacterial growth is not completed efficiently, but a high retention time involves a bigger reactor and the yield of production decreases considerably for retention time higher than 40 h. The best yields were obtained for a mortality ratio in methanogen and acidogenic bacteria lower than 0.2 and a retention time of 30 h with a final production of 3.33 L by each kilogram of biomass.

Computational Simulation to Predict the Syngas Composition Produced during Charcoal Gasification

Charcoal gasification could mitigate the energetic problems in the rural zones since these regions have considerable amounts of wood, which is the base of such a fuel available. This paper presents some CFD (computational fluid dynamic) predictions of the experimental results obtained from the fixed bed gasification of charcoal made in a pilot-scale downdraft reactor using air, which was designed and built by the Research Group in Clean development Mechanisms and Energy Management, from the National University of Colombia. The quality of the syngas obtained from the process was evaluated through the CO and CO2 percentages measured in its composition. The performance at various air flow rates (measured at the system entrance, through an analog flow meter) is evaluated with the help of 11 thermocouples, which give the information to create a temperature profile, and three load cells to measure the solid fuel conversion rate. To simulate the process, the information from temperature profile, charcoal proximate analysis, air flow meter and load cells were taken as inputs and the syngas composition was obtained as the result from the calculation. The domain was defined as 2D with an axis-symmetric description, using quads as mesh elements. The calculation and results were performed in a CFD commercial code widely used for this type of simulations: ANSYS FLUENT. The predictions made by the software were validated with the experimental results obtained in the laboratory.
