Association of Vitamin D Levels with the Metabolic Profile and Sexual Maturation Stage of Adolescents

The deficiency of Vitamin D (VD) is widely prevalent worldwide in adults, but many studies show that this deficiency also affects adolescents and may be considered a global public health problem. The VD levels are particularly significant in adolescents since it influences several aspects of growth, development, and puberty. During this phase of life, both girls and boys develop reproductive aspects, and secondary sexual characteristics and the adequate nutritional status appear to be the prerequisite for normal pubertal development. Due to the importance of VD in adolescents, the objective of this study was to investigate the association of the levels of this vitamin with glycemic, lipid, and anthropometric profile and sexual maturation stages of adolescents. Anthropometric (weight and height) and biochemical (fasting glycemia, total cholesterol, HDL-c, LDL-c, and triglycerides) data and VD levels were collected in 67 adolescents. 66% of the adolescents showed VD sufficiency, 28% insufficiency, and 6% showed the deficiency. There was a significant difference in VD levels between the stages of sexual maturation for boys and girls. Overweight patients had lower serum VD levels. The correlation analysis indicated a positive correlation between VD and fasting glycemia and HDL-c, but with no significant difference. A negative correlation was observed between VD and cholesterol, LDL-c and triglycerides, but also with no significant difference. Once VD is crucial for bone health, and as it seems it is necessary to the homeostasis of glycemia, lipids, and body weight, we suggest that more studies should be conducted to confirm the precise role of this vitamin in the promotion and maintenance of health in this population.

Small-scale self-driving cars:A systematic literature review

The autonomous vehicle(AV)technology has the potential to significantly improve safety and efficiency of the transportation and logistics industry.Full-scale AV testing is limited by time,space,and cost,while simulation-based testing often lacks the necessary accuracy of AV and environmental modeling.In recent years,several initiatives have emerged to test autonomous software and hardware on scaled vehicles.This systematic literature review provides an overview of the literature surrounding small-scale self-driving cars,summarizing the current autonomous platforms deployed and focusing on the software and hardware developments in this field.The studies published in English-language journals or conference papers that present small-scale testing of self-driving cars were included.Web of Science,Scopus,Springer Link,Wiley,ACM Digital Library,and TRID databases were used for the literature search.The systematic literature search found 38 eligible studies.Research gaps in the reviewed papers were identified to provide guidance for future research.Some key takeaway emerging from this manuscript are:(i)there is a need to improve the models and neural network architectures used in autonomous driving systems,as most papers present only preliminary results;(ii)increasing datasets and sharing databases can help in developing more reliable control policies and reducing bias and variance in the training process;(iii)small-scaled vehicles to ensure safety is a major benefit,and incorporating data about unsafe driving behaviors and infrastructure problems can improve the accuracy of predictive models.

Background: We currently have international and national guidelines regarding the assessment and monitoring of clinical signs and humane endpoints in animals used in teaching and research, which make the performance of these activities mandatory for any experiment and professional working in this area. Assigning the severity of a research experiment is the result of an analysis of records of observations of the animal’s behavior, and clinical signs. The aim of this study was to describe the importance of carrying out a severity assessment associated with clinical and behavioral monitoring of rodents and rabbits during experimentation to maintain the welfare of these animals undergoing scientific research. Methods: The literature search was carried out using the following terms: “Monitoring”;“Humane endpoints”;“Animal welfare”, “Rodents”;“Rabbits”, and as connectors “and”;“or”, in the following databases: PubMed;LILACS/BIREME and SciELO. Results: A total of 987 articles were identified in the databases, and 20 of these studies were included in this review. Conclusions: Humane endpoint protocols and procedure severity tables are of the utmost importance, both from an ethical point and to refine the results of research conducted on laboratory animals. They should be drawn up jointly by the teams responsible for the project and the maintenance of the animals during the research period, and the data obtained should be published so that the scientific community can have access to it, helping to disseminate these practices, as well as helping to draw up new procedures. Monitoring and evaluating the welfare and clinical condition of animals undergoing scientific research procedures is the responsibility of the professors, researchers, veterinarians, and animal facility coordinators. The Ethics Committee on the Use of Animals must monitor all the activities conducted with the animals, by inspecting the experimental procedures and the physical environment of the laboratory animal facility where the animals are housed.

Therapeutic Properties of Flavonoids in Treatment of Cancer through Autophagic Modulation:A Systematic Review

Cancers have high morbidity and mortality rates worldwide.Current anticancer therapies have demonstrated specific signaling pathways as a target in the involvement of carcinogenesis.Autophagy is a quality control system for proteins and plays a fundamental role in cancer carcinogenesis,exerting an anticarcinogenic role in normal cells and can inhibit the transformation of malignant cells.Therefore,drugs aimed at autophagy can function as antitumor agents.Flavonoids are a class of polyphenolic secondary metabolites commonly found in plants and,consequently,consumed in diets.In this review,the systematic search strategy was used,which included the search for descriptors"flavonoids"AND"mTOR pathway"AND"cancer"AND"autophagy",in the electronic databases of PubMed,Cochrane Library,Web of Science and Scopus,from January 2011 to January 2021.The current literature demonstrates that flavonoids have anticarcinogenic properties,including inhibition of cell proliferation,induction of apoptosis,autophagy,necrosis,cell cycle arrest,senescence,impaired cell migration,invasion,tumor angiogenesis and reduced resistance to multiple drugs in tumor cells.We demonstrate the available evidence on the roles of flavonoids and autophagy in cancer progression and inhibition.(Registration No.CRD42021243071 at PROSPERO)

Abiotic stress on seed germination and plant growth of Zeyheria tuberculosa

Zeyheria tuberculosa(Vell.)Bureau ex Verl.is among the Brazilian flora threatened with extinction.It has high-quality wood and can be used to recover herbicidecontaminated areas.However,abiotic stresses can harm seed germination and seedling development under adverse conditions and need to be understood to improve autochthonous or indigenous biodiversity conservation and strengthen reforestation projects.This study evaluated the effects of temperature,water and saline stress on Z.tuberculosa seed germination and water stress on seedling growth.Three laboratory experiments evaluated germination under:(1)different temperatures(20,25,30,35 and 40℃);(2)different water potentials simulated with polyethylene glycol(PEG6000)at 25 and 30℃;and,(3)different osmotic potentials simulated with NaCl at 25 and 30℃.In addition,a greenhouse experiment subjected seedlings to five water regimes(20%,35%,50%,65%and 80%of maximum substrate water holding capacity).In the laboratory,seedling biometry and germination were evaluated,while physiological and morphological parameters were assessed in the greenhouse.Physiological potential of seeds increased at 25 and 30℃.Germination and vigor decreased as water and saline stresses increased.Germination tolerance thresholds for water and salt stresses were-0.6 and-0.4 MPa,respectively.In the greenhouse,the most restrictive water regimes reduced seedling physiological and morphological parameters.Seedling development was compromised at moisture levels below50%field capacity.

Are there differences in anaerobic relative muscle power between upper and lower limbs in adolescent swimmers:A blinded study

Success in speed swimming depends on the efficiency of the anaerobic system for the production of cellular energy,especially during muscle power production.In the adolescent athletes much is unknown with regards to the relationships between relative power of upper and lower limbs with speed swimming performance.The aim the present study was to identify differences in relative muscle power of upper and lower limbs in adolescent swimmers and relate these to speed swimming performances.Sixty adolescents,of both sexes(50%female,50%male,30 swimmers and 30 controls),were recruited.The relative upper limb power(ULP[W/kg])was assessed by a medicine ball test and the relative lower limbs power(LLP[W/kg])by a jump test on a jumping platform.Lean mass of the upper and lower limbs was assessed by dual-energy X-ray absorptiometry(DXA)(g).Sport performance was assessed during national level competition(50-m swimming time[in seconds]).Biological maturation(BM)was indexed by years from attainment of peak height velocity.ULP(W/kg)was higher than LLP(W/kg)in both groups(p<0.05).Upper and lower limb lean mass(g)correlated significantly with ULP(W/kg)and LLP(W/kg)in both groups(p<0.05).ULP(W/kg)and LLP(W/kg)correlated with 50-m swimming performance(s),in both sexes(p<0.05).Advanced BM was associated with ULP(W/kg)and LLP(W/kg)in both groups(p<0.05),and with 50-m swimming performance(s)in both sexes(p<0.05).We concluded that ULP(W/kg)is higher than LLP(W/kg)in adolescent swimmers.Upper and lower limb lean mass and BM were both positively associated with increased ULP(W/kg)and LLP(W/kg).

Tumor necrosis factor inhibitors(anti-TNFs)are widely used therapies for the treatment of inflammatory bowel diseases(IBD);however,their administration is not risk-free.Heart failure(HF),although rare,is a potential adverse event related to administration of these medications.However,the exact mechanism of development of HF remains obscure.TNFαis found in both healthy and damaged hearts.Its effects are concentration-and receptor-dependent,promoting either cardio-protection or cardiomyocyte apoptosis.Experimental rat models with TNFαreceptor knockout showed increased survival rates,less reactive oxygen species formation,and improved diastolic left ventricle pressure.However,clinical trials employing anti-TNF therapy to treat HF had disappointing results,suggesting abolishment of the cardioprotective properties of TNFα,making cardiomyocytes susceptible to apoptosis and oxidation.Thus,patients with IBD who have risk factors should be screened for HF before initiating anti-TNF therapy.This review aims to discuss adverse events associated with the administration of anti-TNF therapy,with a focus on HF,and propose some approaches to avoid cardiac adverse events in patients with IBD.

Amprolium exposure alters mice behavior and metabolism in vivo

Background: Thiamine deficiency(TD) models have been developed, mainly using the thiamine analog pyrithiamine. Other analogs have not been used in rodents. We aimed to evaluate the effects and mechanisms of intraperitoneal(ip) amprolium-induced TD in mice. We also evaluated the associated pathogenesis using antioxidant and anti-inflammatory compounds(Trolox, dimethyl sulfoxide).Methods: Male mice were separated into two groups, one receiving a standard diet(control animals), and the other a TD diet(deficient groups) for 20 days. Control mice were further subdivided into three groups receiving daily ip injections of saline(NaCl0.9%; Cont group), Tolox(Tr group) or dimethyl sulfoxide(DMSO; Dmso group). The three TD groups received amprolium(Amp group), amprolium and Trolox(Amp+Tr group), or amprolium and DMSO(Amp+Dmso group). The animals were subjected to behavioral tests and then euthanized. The brain and viscera were analyzed.Results: Amprolium exposure induced weight loss with hyporexia, reduced the behavioral parameters(locomotion, exploratory activity, and motor coordination), and induced changes in the brain(lower cortical cell viability) and liver(steatosis). Trolox co-treatment partially improved these conditions, but to a lesser extent than DMSO.Conclusions: Amprolium-induced TD may be an interesting model, allowing the deficiency to develop more slowly and to a lesser extent. Amprolium exposure also seems to involve oxidative stress and inflammation, suggested as the main mechanisms of cell dysfunction in TD.

Glial cells are crucial for maintaining central nervous system(CNS)homeostasis.They actively participate in immune responses,as well as form functional barriers,such as blood-brain barrier(BBB),which restrict the entry of pathogens and inflammatory mediators into the CNS.In general,viral infections during the gestational period can alter the embryonic and fetal environment,and the related inflammatory response may affect neurodevelopment and lead to behavioral dysfunction during later stage of life,as highlighted by our group for Zika virus infection.Severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)induces a cytokine storm and,during pregnancy,may be related to a more severe form of the coronavirus disease-19(COVID-19)and also to higher preterm birth rates.SARS-CoV-2 can also affect the CNS by inducing neurochemical remodeling in neural cells,which can compromise neuronal plasticity and synaptic function.However,the impact of SARS-CoV-2 infection during pregnancy on postnatal CNS,including brain development during childhood and adulthood,remains undetermined.Our group has recently highlighted the impact of COVID-19 on the expression of molecular markers associated with neuropsychiatric disorders,which are strongly related to the inflammatory response.Thus,based on these relationships,we discussed the impact of SARS-CoV-2 infection either during pregnancy or in critical periods of neurodevelopment as a risk factor for neurological consequences in the offspring later in life,focusing on the potential role of glial cells.Thus,it is important to consider future and long-term public health concerns associated with SARS-CoV-2 infection during pregnancy.

This study hypothesized that different land-use affect the microbial enzymatic stoichiometry and C-,N-,and P-acquisition in Brazilian semiarid soils.Thus,the enzymesβ-glucosidase(C-acquiring enzyme),urease(N-acquiring enzyme),and acid phosphatase(P-acquiring enzyme)were assessed in soil samples collected at 0−5 and 5−10 cm depth from a tropical dry forest,a protected area with Angico,a protected area with Ipê,scrub area,and an agricultural area with maize.The values of C-,N-,and P-acquiring enzymes were used to calculate the enzymatic C:N,C:P,and N:P ratios.The values of C:P and N:P ratios were higher at 0−5 cm depth,while no significant variation,between soil depth,was observed for C:N ratio.The values of C-and N-acquiring enzymes were higher at 0−5 cm in tropical dry forest areas and Angico forest,respectively.In all land use types,the values of vectors L and A were higher than 1°and 45°,respectively.This study showed that both land-use and soil depth influence the enzymatic stoichiometry,showing higher values of C-and N-acquiring enzymes in native and protected forests at soil surface.
