Quality evaluation of nothofagus pumilio seeds linked to forest management and climatic events

Background Forest ecosystems undergo significant transformations due to harvesting and climate fluctuations, emphasizing the critical role of seeding in natural regeneration and long-term structural preservation. Climate change further amplifies these dynamics, affecting phenology across species and regions. In Tierra del Fuego(Argentina), nothofagus pumilio(lenga) forests represent the most important timber resource, and it is managed through different silvicultural strategies. This species demonstrates notable post-disturbance regeneration, yet seed fall exhibits signifi-cant variability, leading to variations in seed quality(e.g., viability). This study aims to assess fluctuations in n. pumilio seed quality, determine how it varies concerning forest management strategies, annual productivity, and the co-occurrence of climatic phenomena including El ni?o-Southern Oscillation(EnSO) and the Southern Annular Mode(SAM).Results Viable seeds represented 18.4% of the total, notably higher in unharvested than in managed areas. Con-versely, empty seeds were more prevalent in harvested areas(> 75%). Seed quality exhibited significant differences across silvicultural treatments, except for insect-predated seeds, which had similar proportions across all areas, though dispersed retention showed higher predation. When considering years with varying production levels, high-production years favoured full and viable seeds, particularly in unharvested forests and aggregated retention, while low-production years saw reduced viability across all treatments. Quadratic models revealed that viability increased with seed production, where unharvested forests achieved the highest values. Climate variability influenced seed proportions, where EnSO+/SAM+ promoting more full and viable seeds, while EnSO–/SAM+ favoured non-predated seeds, especially in unharvested *** Seed quality varies among treatments and years with different levels of seeding. Rydberg excitons(analogues of Rydberg atoms in condensed matter systems)are highly excited bound electron-hole states with large Bohr *** interaction between them as well as exciton coupling to light may lead to strong optical nonlinearity,with applications in sensing and quantum information ***,we achieve strong effective photon–photon interactions(Kerr-like optical nonlinearity)via the Rydberg blockade phenomenon and the hybridisation of excitons and photons forming polaritons in a Cu2O-filled *** pulsed resonant excitation polariton resonance frequencies are renormalised due to the reduction of the photon-exciton coupling with increasing exciton *** analysis shows that the Rydberg blockade plays a major role in the experimentally observed scaling of the polariton nonlinearity coefficient as∝n4.4±1.8 for principal quantum numbers up to n=*** high principal quantum numbers studied in a polariton system for the first time are essential for realisation of high Rydberg optical nonlinearities,which paves the way towards quantum optical applications and fundamental studies of strongly correlated photonic(polaritonic)states in a solid state system.

Progressive osseous heteroplasia(POH)is an ultra-rare autosomal dominant disabling disorder characterized by heterotopic ossification(HO).It is caused by heterozygous inactivating mutations in the GnAS(guanine nucleotide-binding protein alpha-stimulating activity polypeptide)***,the molecular mechanisms underlying HO remain poorly *** a treatment for POH is not yet available,the identification of the mechanisms driving POH in affected tissues using gene expression may be of great help to underestand the molecular basis of POH and develop new therapeutic approaches.

Data-driven models for ground and excited states for Single Atoms on Ceria

Ceria-based single-atom catalysts present complex electronic structures due to the dynamic electron transfer between the metal atoms and the semiconductor oxide *** these materials implies retrieving all states in these electronic ensembles,which can be limiting if done via density functional ***,we propose a data-driven approach to obtain a parsimonious model identifying the appearance of dynamic charge transfer for the single atoms(SAs).We first constructed a database of(701)electronic configurations for the group 9–11 metals on CeO_(2)(100).Feature Selection based on predictive Elastic net and Random Forest models highlights eight fundamental variables:atomic number,ionization potential,size,and metal coordination,metal–oxygen bond strengths,surface strain,and Coulomb *** these variables a Bayesian algorithm yields an expression for the adsorption energies of SAs in ground and low-lying excited *** work paves the way towards understanding electronic structure complexity in metal/oxide interfaces.

Humanity is in the grip of another pandemic beyond coronavirus disease(COVID).Although it is not spreading as rapidly as the viral pandemic,it is insidious and probably presents a greater threat in the long *** the large-scale use of antibiotics,we have driven the emergence of antimicrobial resistance(AMR)in bacteria,and these resistant bacteria are increasing in number and prevalence,threatening our ability to treat common infections and reducing our opportunities to use other life-saving treatments such as chemotherapy and surgery.A recent study showed that bacterial AMR was associated with nearly 5 million deaths in2019,with about 1.3 million being directly attributable to resistance[1].These numbers are likely to increase.

为进一步明确麦二叉蚜Schizaphis graminum唾液蛋白Sg1655在调控小麦防御反应中的作用,基于麦二叉蚜唾液腺转录组数据,利用PCR技术克隆获取Sg1655开放阅读框,利用生物信息学软件对其序列进行分析;通过实时荧光定量PCR(real-time quantitative PCR,RT-qPCR)技术检测Sg1655在麦二叉蚜不同组织及不同龄期中的表达;利用细菌III型分泌系统将Sg1655导入到小麦叶片内,检测其瞬时表达对小麦胼胝质积累的影响。结果表明,Sg1655开放阅读框全长375 bp,编码125个氨基酸残基,n端1~21位为信号肽序列,预测蛋白分子量为14.90 kD。麦二叉蚜Sg1655氨基酸序列与玉米蚜Rhopalosiphum maidis同源序列相致性最高,为89.52%;麦二叉蚜Sg1655与玉米蚜同源序列聚为一个分支,亲缘关系最近。Sg1655在麦二叉蚜各组织中均有表达,其中在唾液腺中表达量最高,Sg1655在麦二叉蚜各个龄期均有表达,且在不同龄期之间无显著差异。小麦叶片内Sg1655瞬时表达可显著抑制小麦胼胝质积累,表明该蛋白可作为潜在效应子参与抑制小麦防御反应。

Predictors of bowel damage in the long-term progression of Crohn’s disease

BACKGROUnD Crohn’s disease(CD)is a chronic inflammatory bowel disorder that progresses to bowel damage(BD)over *** image-based index,the Lémann index(LI),has been developed to measure cumulative *** To characterize the long-term progression of BD in CD based on changes in the LI and to determine risk factors for long-term *** This was a single-center longitudinal cohort *** who had participated in prospective studies on the accuracy of magnetic resonance imaging using endoscopy as a gold standard and who had a follow-up of at least 5 years were reevaluated after 5-12 *** Seventy-two patients were *** increased in 38 patients(52.8%),remained unchanged in 9 patients(12.5%),and decreased in 25 patients(34.7%).The small bowel score and surgery subscale significantly increased(P=0.002 and P=0.001,respectively),whereas the fistulizing subscale significantly decreased(P=0.001).Baseline parameters associated with BD progression were ileal location(P=0.026),CD phenotype[stricturing,fistulizing,or both(P=0.007,P=0.006,and P=0.035,respectively)],disease duration>10 years(P=0.019),and baseline LI stricturing score(P=0.049).no correlation was observed between BD progression and baseline clinical activity,biological markers,or severity of endoscopic *** BD,as assessed by the LI,progressed in half of the patients with CD over a period of 5-12 *** main determinants of BD progression were ileal location,stricturing/fistulizing phenotype,and disease duration.

The development of a new generation of nonantibody protein drug delivery systems requires site-directed conjugation strategies to produce homogeneous,reproducible and scalable *** that,the genetic addition of cysteine residues into solvent-exposed positions allows the thiol-mediated cysteine coupling of therapeutic drugs into protein-based ***,the high reactivity of unpaired cysteine residues usually reduces protein stability,consequently imposing the use of more methodologically demanding purification *** is especially relevant for disulfide-containing nanocarriers,as previously observed in ***,although many protein scaffolds and targeting ligands are also rich in disulfide bridges,the use of these methodologies over emerging non-antibody carrier proteins has been completely ***,we report the development of a simple and straightforward procedure for a onestep production and site-directed cysteine conjugation of disulfide-containing non-antibody thiolated carrier proteins(THIOCAPs).This method is validated in a fluorescent C-X-C chemokine receptor 4(CXCR4)-targeted multivalent nanocarrier containing two intramolecular disulfide bridges and one reactive cysteine residue strategically placed into a solvent-exposed position(THIO-T22-GFP-H6)for drug conjugation and in a humanized alternative intended for clinical applications(T22-HSnBT-H6).Thus,we produce very stable,homogeneous and fully functional antitumoral nanoconjugates(THIO-T22-GFP-H6-MMAE and T22-HSnBT-H6-MMAE)that selectively eliminate target cancer cells via ***,the developed methodology appears as a powerful tool for the rational engineering of emerging non-antibody,cell-targeted protein nanocarriers that contain disulfide bridges together with a solvent-exposed reactive cysteine(THIOCAP).This should pave the way for the development of a new generation of stable,homogeneous and efficient nanomedicines.
