Increased connectivity of hiPSC-derived neural networks in multiphase granular hydrogel scaffolds

To reflect human development,it is critical to create a substrate that can support long-term cell survival,differentiation,and *** are promising materials for 3D ***,a bulk structure consisting of dense polymer networks often leads to suboptimal microenvironments that impedes nutrient exchange and cell-to-cell ***,granular hydrogel-based scaffolds were used to support 3D human induced pluripotent stem cell(hiPSC)-derived neural networks.A custom designed 3D printed toolset was developed to extrude hyaluronic acid hydrogel through a porous nylon fabric to generate hydrogel *** and hydrogel granules were combined using a weaker secondary gelation step,forming self-supporting cell laden *** three and seven days,granular scaffolds supported higher cell viability compared to bulk hydrogels,whereas granular scaffolds supported more neurite bearing cells and longer neurite extensions(65.52±11.59μm)after seven days compared to bulk hydrogels(22.90±4.70μm).Long-term(three-month)cultures of clinically relevant hiPSC-derived neural cells in granular hydrogels supported well established neuronal and astrocytic colonies and a high level of neurite extension both inside and beyond the *** approach is significant as it provides a simple,rapid and efficient way to achieve a tissue-relevant granular structure within hydrogel cultures.

Utilization of ph-driven methods to fortify nanoemulsions with multiple polyphenols

Emergence of the colistin resistance gene,mcr-1,has attracted worldwide *** the prevalence of mcr-1-positive Escherichia coli(MCRPEC)strains in human carriage showing a significant decrease between 2016 and 2019,genetic differences in MCRPEC strains remain largely *** therefore conducted a comparative genomic study on MCRPEC strains from fecal samples of healthy human subjects in 2016 and *** identified three major differences in MCRPEC strains between these two time ***,the insertion sequence ISApl1 was often deleted and the percentage of mcr-1-carrying IncI2 plasmids was increased in MCRPEC strains in ***,the antibiotic resistance genes(ARGs),aac(3)-IVa and blaCTX-M-1,emerged and coexisted with mcr-1 in ***,MCRPEC strains in 2019 contained more virulence genes,resulting in an increased proportion of extraintestinal pathogenic ***(ExPEC)strains(36.1%)in MCRPEC strains in 2019 compared to that in 2016(10.5%),implying that these strains could occupy intestinal ecological niches by competing with other commensal *** results suggest that despite the significant reduction in the prevalence of MCRPEC strains in humans from 2016 to 2019,MCRPEC exhibits increased resistance to other clinically important ARGs and contains more virulence genes,which may pose a potential public health threat.

Advanced bioactive glue tethering Lubricin/PRG4 to promote integrated healing of avascular meniscus tears

Meniscus injuries are extremely common with approximately one million patients undergoing surgical treatment annually in the U.S. alone, but no regenerative therapy exist. Previously, we showed that controlled applications of connective tissue growth factor (CTGF) and transforming growth factor beta 3 (TGFβ3) via fibrin-based bio-glue facilitate meniscus healing by inducing recruitment and stepwise differentiation of synovial mesenchymal stem/progenitor cells. here, we first explored the potential of genipin, a natural crosslinker, to enhance fibrin-based glue’s mechanical and degradation properties. In parallel, we identified the harmful effects of lubricin on meniscus healing and investigated the mechanism of lubricin deposition on the injured meniscus surface. We found that the pre-deposition of hyaluronic acid (hA) on the torn meniscus surface mediates lubricin deposition. Then we implemented chemical modifications with heparin conjugation and CD44 on our bioactive glue to achieve strong initial bonding and integration of lubricin pre-coated meniscal tissues. Our data suggested that heparin conjugation significantly enhances lubricin-coated meniscal tissues. Similarly, CD44, exhibiting a strong binding affinity to lubricin and hyaluronic acid (hA), further improved the integrated healing of hA/lubricin pre-coated meniscus injuries. These findings may represent an important foundation for developing a translational bio-active glue guiding the regenerative healing of meniscus injuries.

Rolling in Nature and Robotics: A Review

This paper presents a review of recent rolling robots including Rollo from helsinki University of Technology, Spherical Mobile Robot from the Politecnico of Bad, Sphericle from the University of Pisa, Spberobot from Michigan State University, August from Azad University of Qazvin and the University of Tehran, Deformable Robot from Ritsumeijan University, Kickbot from the Massachusetts Institute of Technology, Gravitational Wheeled Robot from Kinki University, Gyrover from Carnegie Mellon University, Roball from the Université de Sherbrooke, and Rotundus from the Angstroem Space Technology Center. Seven rolling robot design principles are presented and discussed （Sprung central member, Car driven, Mobile masses, hemispherical wheels, Gyroscopic stabilisation, Ballast mass - fixed axis, and Ballast mass - moving axis）. Robots based on each of the design principles are shown and the performances of the robots are tabulated. An attempt is made to grade the design principles based on their suitability for movement over an unknown and varied but relatively smooth terrain. The result of this comparison suggests that a rolling robot based on a mobile masses principle would be best suited to this specific application. Some wonderful rolling organisms are introduced and defined as ＂active＂ or ＂passive＂ depending on whether they generate their own rolling motion or external forces cause their rolling.

Toward the implementation of a universal angle-based optical indoor positioning system

There is an emerging market today for indoor positioning systems capable of working alongside global navigation satellite systems, such as the global positioning system, in indoor environments. Many systems have been proposed in the literature but all of them have fundamental flaws that hold them back from widescale implementation. We review angle-of-arrival （AOA） and angle-difference- of-arrival （ADOA） optical indoor positioning systems which have been proven to be robust, accurate, and easily implementable. We build an AOA/ADOA optical indoor positioning system out of a simple commercial high-speed camera and white light light emitting diodes （LEDs） which operate over a working area of 1 m3, and compare its performance to other indoor positioning methods. The AOA and ADOA systems achieve positioning with low errors of 1.2 and 3.7 cm, respectively.
