The concentration of the volatile fatty acids (VFA) is an important indicator of the status of an-aerobic processes, but most of the existing methods require sample pretreatment and are labor-intensive. It was developed and validated a rapid Gas Chromatographic (GC) method to quantify seven VFA (acetic, propionic, isobutyric, butyric, isovaleric, valeric and caproic), acetone, methanol, ethanol and n-butanol by headspace (automatic and manual) and liquid-liquid extraction (LLE) with diethyl ether (only VFA). The determination was made in a Shimadzu Gas Chromatograph equipped with a Flame Ionization Detector (GC/FID), a headspace auto-sampler and an HP-INNOWAX column. Isobutanol and crotonic acid were utilized as internal standards (IS). The validation parameters evaluated were: precision (coefficient of variation—C.V.% for the retention times, from 0.02 to 0.87), linearity (r2 = 0.9291 - 0.9997), limits of detection (from 3.97 to 36.45 mg·L﹣1) and instrumental precision (from 0.01 to 0.53), which provide evidence that the methods are adequate to determine these analytes in samples from anaerobic reactors and from the environment.

The deepwater horizon blowout led to the release of 4.9 million barrels of crude oil into the Gulf of Mexico (GOM). Here the paper analyzes available sediment datasets to determine whether changes in petrogenic contaminants are evident in GOM sediment following the blowout. Locations sampled by EPA (Environmental Protection Agency), British Petroleum (BP) and NOAA (National Oceanic and Atmospheric Administration) were reduced and grouped into 46 similar locations and analyzed. Eleven groups suggested an increase in PAH (polycyclic aromatic hydrocarbon) and nickel, one group showed a statistically significant increase in Phenanthrene. Four locations were analyzed for time trends and differences between initial and peak concentrations of oil range organics (OrO), diesel range organics (DrO), vanadium and nickel. One location had significant increases in OrO, DrO and vanadium and a suggestive increase in nickel. Correlations between OrO/DrO, OrO/V, OrO/Ni, V/Ni were computed (r2 = 0.85, 0.67, 0.64, 0.92, respectively). Overall, the analyses suggest that future monitoring should employ a sampling strategy that coordinates response sampling to previously sampled locations, such that baseline datasets can be used in detection of event associated contamination.

AIM: To evaluate the effect of resistance to activated protein C (aPCr), the most common known inherited thrombophilic disorder, on the risk of intestinal operation of fi brostenosis in patients with Crohn’s disease (CD). METHODS: In a previous study, we assessed the prevalence of aPCr in CD. In a retrospective case- controlled study, 8 of these CD patients with aPCr were now compared with 24 CD patients without aPCr, matched by gender, age at diagnosis and duration of disease in a 1:3 fashion. The primary end point was the occurrence of an intestinal CD-related operation with evidence of fibrostenosis in the bowel resection specimen. rESULTS: The Kaplan-Meier analysis revealed that patients with aPCr had a lower probability of remaining free of operation with f ibrostenosis than patients without aPCr (P = 0.0372; exact log-rank test) resulting in a signifi cantly shorter median time interval from diagnosis of CD to the fi rst operation with fi brostenosis (32 vs 160 mo). At 10 years, the likelihood of remaining free of operation with fi brostenosis was 25% for patients with aPCr and 57.8% for patients without aPCr. CONCLUSION: CD patients with aPCr are at higher risk to undergo intestinal operation of fi brostenosis than those without aPCr. This supports our hypothesis of aPCr being a possible risk factor for fi brostenosis in CD.

AIM: To investigate the mechanism of action of lipophilic antidepressant fluoxetine(FLX) in representative molecular subtypes of breast ***: The anti-proliferative effects and mechanistic action of FLX in triple-negative(SUM149PT) and luminal(T47D and Au565) cancer cells and nontransformed MCF10 A were investigated. reverse phase protein microarray(rPPM) was performed with and without 10 μmol/L FLX for 24 and 48 h to determine which proteins are significantly changed. Viability and cell cycle analysis were also performed to determine drug effects on cell growth. Western blotting was used to confirm the change in protein expression examined by rPPM or pursue other signaling proteins. rESULTS: The FLX-induced cell growth inhibition in all cell lines was concentration- and time-dependent but less pronounced in early passage MCF10 A. In comparison to the other lines,cell growth reduction in SUM149 PT coincided with significant induction of endoplasmic reticulum(Er) stress and autophagy after 24 and 48 h of 10 μmol/L FLX,resulting in decreased translation of proteins along the receptor tyrosine kinase/Akt/mammalian target of rapamycin pathways. The increase in autophagy marker,cleaved microtubule-associated protein 1 light chain 3,in SUM149 PT after 24 h of FLX was likely due to increased metabolic demands of rapidly dividing cells and Er stress. Consequently,the unfolded protein response mediated by double-stranded rNA-dependent protein kinase-like Er kinase resulted in inhibition of protein synthesis,growth arrest at the G1 phase,autophagy,and caspase-7-mediated cell ***: Our study suggests a new role for FLX as an inducer of Er stress and autophagy,resulting in death of aggressive triple negative breast cancer SUM149 PT.

Genome-wide association studies(GWASs) have been instrumental in understanding complex phenotypic traits. However, they have rarely been used to understand lineage-specific pathways and functions that contribute to the trait. In this study, by integrating lineage-specific enhancers from mesenchymal and myeloid compartments with bone mineral density loci, we were able to segregate osteoblast-and osteoclast(OC)-specific functions. Specifically, in OCs, a PU.1-dependent transcription factor(TF)network was revealed. Deletion of PU.1 in OCs in mice resulted in severe osteopetrosis. Functional genomic analysis indicated PU.1 and MITF orchestrated a TF network essential for OC differentiation. Several of these TFs were regulated by cooperative binding of PU.1 with BrD4 to form superenhancers. Further, PU.1 is essential for conformational changes in the superenhancer region of Nfatc1. In summary, our study demonstrates that combining GWASs with genome-wide binding studies and model organisms could decipher lineage-specific pathways contributing to complex disease states.

Agriculture is essential for providing food and maintaining food security while concurrently delivering multiple other ecosystem services. However,agricultural systems are generally a net source of greenhouse gases and ammonia. They, therefore, need to substantively contribute to climate change mitigation and net zero ambitions. It is widely acknowledged that there is a need to further reduce and mitigate emissions across sectors, including agriculture to address the climate emergency and emissions gap. This discussion paper outlines a collation of opinions from a range of experts within agricultural research and advisory roles following a greenhouse gas and ammonia emission mitigation workshop held in the UK in March 2022. The meeting identified the top mitigation priorities within the UK's agricultural sector to achieve reductions in greenhouse gases and ammonia that are compatible with policy targets. In addition, experts provided an overview of what they believe are the key knowledge gaps, future opportunities and cobenefits to mitigation practices as well as indicating the potential barriers to uptake for mitigation scenarios discussed.

Surface (S)-layer proteins are model systems for studying protein glycosylation in bacteria and simultaneously hold promises for the design of novel, glyco-functionalized modules for nanobiotechnology due to their 2D self-assembly capability. Understanding the mechanism governing S-layer glycan biosynthesis in the Gram-positive bacterium Paenibacillus alvei CCM 2051T is necessary for the tailored glyco-functionalization of its S-layer. Here, the putative oligosaccharyl:S-layer protein transferase WsfB from the P. alvei S-layer glycosylation gene locus is characterized. The enzyme is proposed to catalyze the final step of the glycosylation pathway, transferring the elongated S-layer glycan onto distinct tyrosine O-glycosylation sites. Genetic knock-out of WsfB is shown to abolish glycosylation of the S-layer protein SpaA but not that of other glycoproteins present in P. alvei CCM 2051T, confining its role to the S-layer glycosylation pathway. A transmembrane topology model of the 781-amino acid WsfB protein is inferred from activity measurements of green fluorescent protein and phosphatase A fused to defined truncations of WsfB. This model shows an overall number of 13 membrane spanning helices with the Wzy_C domain characteristic of O-oligosaccharyl:protein transferases (O-OTases) located in a central extra-cytoplasmic loop, which both compares well to the topology of OTases from Gram-negative bacteria. Mutations in the Wzy C motif resulted in loss of WsfB function evidenced in reconstitution experiments in P. alvei ΔWsfB cells. Attempts to use WsfB for transferring heterologous oligosaccharides to its native S-layer target protein in Escherichia coli CWG702 and Salmonella enterica SL3749, which should provide lipid-linked oligosaccharide substrates mimicking to some extent those of the natural host, were not successful, possibly due to the stringent function of WsfB.
