Role of soluble factors and three-dimensional culture in in vitro differentiation of intestinal macrophages

AIM: To examine the factor(s) involved in differentiation of intestinal macrophages (IMACs) using a recently established in vitro model. METHODS: To test whether soluble or membrane bound factors induce IMAC-differentiation, freshly elutriated monocytes (MO) were incubated with conditioned media or cell membranes of intestinal epithelial cells (IEC) or cultured with IEC in transwell systems. To determine the importance of an active migration of MO, three- dimensional aggregates from a 1:1-mixture of MO and IEC were examined by immunohistochemistry and flow cytometry. Apoptosis was examined by caspase-3 Western blots. Extracellular matrix production in differentiation models was compared by immunohistochemistry. RESULTS: IMAC differentiation was observed in a complex three-dimensional co-culture model (multicellular spheroid, MCS) with IEC after migration of MO into the spheroids. By co-culture of MO with conditioned media or membrane preparations of IEC no IMAC differentiation was induced. Co-culture of MO with IEC in transwell- cultures, with the two cell populations separated by a membrane also did not result in intestinal-like differentiation of MO. In contrast to IEC-spheroids with immigrating MO in mixed MCS of IEC and MO only a small subpopulation of MO was able to survive the seven day culture period. CONCLUSION: Intestinal-like differentiation of MO in vitro is only induced in the complex three-dimensional MCS model after immigration of MO indicating a roleof cell-matrix and/or cell-cell interactions during the differentiation of IMACs.

Double-balloon-enteroscopy-based endoscopic retrograde cholangiopancreatography in post-surgical patients

AIM: To evaluate double balloon enteroscopy (DBE) in post-surgical patients to perform endoscopic retrograde cholangiopancreatography (ERCP) and interventions. METHODS: In 37 post-surgical patients, a stepwise approach was performed to reach normal papilla or enteral anastomoses of the biliary tract/pancreas. When conventional endoscopy failed, DBE-based ERCP was performed and standard parameters for DBE, ERCP and interventions were recorded. RESULTS: Push-enteroscopy (overall, 16 procedures) reached enteral anastomoses only in six out of 37 post-surgical patients (16.2%). DBE achieved a high rate of luminal access to the biliary tract in 23 of the remaining 31 patients (74.1%) and to the pancreatic duct (three patients). Among all DBE-based ERCPs (86 procedures), 21/23 patients (91.3%) were successfully treated. Interventions included ostium incision or papillotomy in 6/23 (26%) and 7/23 patients (30.4%), respectively. Biliary endoprosthesis insertion and regular exchange was achieved in 17/23 (73.9%) and 7/23 patients (30.4%), respectively. Furthermore, bile duct stone extraction as well as ostium and papillary dilation were performed in 5/23 (21.7%) and 3/23 patients (13.0%), respectively. Complications during DBE-based procedures were bleeding (1.1%), perforation (2.3%) and pancreatitis (2.3%), and minor complications occurred in up to 19.1%. CONCLUSION: The appropriate use of DBE yields a high rate of luminal access to papilla or enteral anastomoses in more than two-thirds of post-surgical patients, allowing important successful endoscopic therapeutic interventions.

Recovery from respiratory failure after decompression laparotomy for severe acute pancreatitis

We present three cases of patients (at the age of 56 years, 49 years and 74 years respectively) with severe acute pancreatitis (SAP), complicated by intra-abdominal compartment syndrome (ACS) and respiratory insufficiency with limitations of mechanical ventilation. The respiratory situation of the patients was significantly improved after decompression laparotomy (DL) and lung protective ventilation was re-achieved. ACS was discussed followed by a short review of the literature. Our cases show that DL may help patients with SAP to recover from severe respiratory failure.

Growth of Large-Area Single- and Bi-Layer Graphene by Controlled Carbon Precipitation on Polycrystalline Ni Surfaces

We report graphene films composed mostly of one or two layers of graphene grown by controlled carbon precipitation on the surface of polycrystalline Ni thin films during atmospheric chemical vapor deposition(CVD).Controlling both the methane concentration during CVD and the substrate cooling rate during graphene growth can signifi cantly improve the thickness *** a result,one-or two-layer graphene regions occupy up to 87%of the fi lm *** layer coverage accounts for 5%11%of the overall fi *** regions expand across multiple grain boundaries of the underlying polycrystalline Ni fi *** number density of sites with multilayer graphene/graphite(>2 layers)is reduced as the cooling rate *** fi lms can also be transferred to other substrates and their sizes are only limited by the sizes of the Ni fi lm and the CVD ***,we demonstrate the formation of fi lms as large as 1 in^(2).These fi ndings represent an important step towards the fabrication of large-scale high-quality graphene samples.

对焊接数值模拟中的各种加速计算方法进行了细致研究,并在分段热源法的基础上提出了温度函数法.和分段热源法相比较,温度函数法将焊接数值模拟中的控制变量由热量转变为温度,增加了焊接温度场的可控制性,并提高了模拟温度场在模型宽度和厚度方向与实际焊接温度场的一致性.文中通过铝合金薄板搅拌摩擦对焊的模型对温度函数法进行验证.结果表明,温度函数法得到的残余应力和变形结果与移动热源方法得到的结果具有一致性,并且和实际试验测量结果吻合,计算效率有大幅度的提高,能够达到工程应用的需要.

通过在6056—T6的铝合金薄板上设计不同旋转频率、不同下压量的搅拌摩擦点焊试验,测量焊接过程中的搅拌头扭矩、下压作用力和试板温度曲线来研究焊接工艺参数对扭矩的影响.结果表明,在旋转频率相同而下压量不同的试验中,搅拌头扭矩随下压作用力增大而增大,而引起扭矩增大的最主要原因是下压作用力使搅拌区材料的力学性能发生了改变;在下压量相同而旋转频率不同的焊接试验中,搅拌头扭矩随旋转频率升高而减小,但扭矩和旋转频率的乘积(即焊接热输入功率)却随旋转频率升高而增大,分析认为旋转频率通过温度和材料的应变速率两个途径对搅拌头扭矩产生了影响,而温度的影响作用更显著.

Compressive holographic sensing simplifies quantitative phase imaging

Quantitative phase imaging (QPI) has emerged as method for investigating biological specimen and technical objects. However, conventional methods often suffer from shortcomings in image quality, such as the twin image artifact. A novel computational framework for QPI is presented with high quality inline holographic imaging from a single intensity image. This paradigm shift is promising for advanced QPI of cells and tissues.

Diagnosis of obscure gastrointestinal bleeding by intraoperative enteroscopy in 81 consecutive patients

AIM： To analyze the results and complications of intraoperative enteroscopy （IOE） by investigating a series of selected patients with bleeding suspected to originate from the small intestine. METHODS： Eighty-one patients （mean age： 65 years） including 40 males （49.4%） and 41 females （50.6%） with obscure gastrointestinal bleeding underwent IOE between 1990 and 2004. The patients were identified from a database and data were selected from the patients＇ charts retrospectively. All the patients had undergone at least one non-diagnostic esophagogastroduodenoscopy, colonoscopy, standard enteroscopy and a negative abdominal ultrasound scan before IOE. RESULTS： The median minimal hemoglobin level in the patients was 59＋15g/L and 72.8% of the patients required transfusion of packed erythrOoltes previously. A bleeding source was detected in 68 （84%） of the patients during IOE. Angiodysplasiae were found in 44 patients （54.3%） and 9 patients （11.1%） were affected by ulcers in the small intestine. A tumor in the small intestine was detected in another 6 patients. The treatment consisted of argon-plasma-coagulation, surgical suture or limited resection in most of the patients. CONCLUSION： Intra-operative enteroscopy is still used for the diagnosis of suspected small bowel bleeding. Recent developments such as wireless capsule endoscopy and double balloon enteroscopy, may lead to the replacement of IOE in the future.

Do Near-Solar-System Supernovae Enhance Volcanic Activities on Earth and Neighbouring Planets on Their Paths through the Spiral Arms of the Milky Way, and What Might Be the Consequences for Estimations of Earth’s History and Predictions for Its Future?

Recent observations of young volcanism on the near-Earth terrestrial planets require a new understanding. Magmatic/volcanic episodes on Venus, Mars and Mercury, as well as on Earth’s Moon, are apparently contemporaneous thermal events that accompanied increased magmatic/volcanic activity on Earth, following a 300-Myr cycle. A collateral galactic thermal source in the Milky Way appears to be needed that would predominantly affect the interior of the planets and, perhaps indirectly, Earth’s biosphere, compared to other galactic sources, such as intense cosmic rays or large, rocky bolides. The search for such a source leads to near-Earth supernovae, with their neutrino output, and to the question of whether those neutrinos could act as energy transmitters to heat up the body of Earth, and also enhance its short-term magmatic processes;for example, Cenozoic anorogenic volcanism. This observation challenges present assumptions and paradigms about Earth’s history, and requires the following reconsiderations: 1) the real origin of the end-Cretaceous mass extinction;2) the general radioactive age determinations of rocks;and 3) geodynamic modelling using additional, external heat sources.
