Metabolic-associated fatty liver disease(MAFLD)is a new disease definition,and is proposed to replace the previous name,nonalcoholic fatty liver disease(NAFLD).Globally,MAFLD/NAFLD is the most common liver disease,with an incidence rate ranging from 6%to 35%in adult *** pathogenesis of MAFLD/NAFLD is closely related to insulin resistance(IR),and the genetic susceptibility to acquired metabolic stress-associated liver ***,the gut microbiota in MAFLD/NAFLD is being revaluated by scientists,as the gut and liver influence each other via the gut-liver *** is a novel form of programmed cell death caused by iron-dependent lipid *** evidence suggests that ferroptosis has a key role in the pathological progression of MAFLD/NAFLD,and inhibition of ferroptosis may become a novel therapeutic strategy for the treatment of *** review focuses on the main mechanisms behind the promotion of MAFLD/NAFLD occurrence and development by the intestinal microbiota and *** outlines new strategies to target the intestinal microbiota and ferroptosis to facilitate future MAFLD/NAFLD therapies.

【目的】本研究将CRISPR/Cas12a系统与反转录重组酶介导等温核酸扩增技术(RT-RAA)结合，拟建立一种基于CRISPR/Cas12a-RT-RAA的猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus, PRRSV)快速检测方法。【方法】分析不同基因型PRRSV全基因组序列，在3′-UTR端保守区设计重组酶介导等温核酸扩增技术(RAA)引物、crRNA及DNA报告底物分子，筛选RT-RAA最佳引物对。表达纯化AsCas12a蛋白，并以质粒为标准品，经RT-RAA扩增后，将产物加入CRISPR/Cas12a系统，建立PRRSV的CRISPR/Cas12a-RT-RAA检测方法，分别以PRRSV、猪圆环病毒2型(PCV2)、猪流行性腹泻病毒(PEDV)、猪瘟病毒(CSFV)、猪伪狂犬病病毒(PRV)核酸为模板验证建立方法的特异性，以不同浓度的pMD18T-PRRSV为模板验证建立方法的敏感性。以5.62×10～6、5.62×10～3、5.62拷贝数/μL的质粒为模板进行重复性试验，最后进行临床效果评价，将所建立的方法与实时荧光定量逆转录PCR方法进行比较。【结果】本研究成功表达并纯化获得具有良好剪切活性的AsCas12a蛋白，蛋白分子质量大小为196 ku;所建立的CRISPR/Cas12a-RT-RAA方法的检测下限可达5.62拷贝/μL;该方法具有良好的特异性，可特异性检测出PRRSV,不能检出其他猪病病毒PCV2、PEDV、CSFV和PRV,并具有良好的重复性。应用该方法对121份猪组织样品进行检测，与实时荧光定量逆转录PCR的阳性符合率为93.75%,阴性符合率为99.05%,总符合率为99.17%。【结论】本研究成功建立基于CRISPR/Cas12a-RT-RAA的PRRSV快速检测方法，该方法特异性强、灵敏性高、重复性好且不依赖复杂设备，可在现场和基层实验室使用。

Objective:Functional constipation(FC)is a common intestinal disease *** the presence of criteria such as Roman IV,there is no standardized diagnosis and treatment algorithm in Hong Kong that combines both Western and Chinese medicine *** study integrates current effective and safe diagnosis and treatment methods for FC and provides a clear and scientific pathway for clinical professionals and ***:A systematic search of the PubMed,Cochrane Library,and China National Knowledge Infrastructure databases was performed from their inception to June 30th,2022,collecting the current evidence about the efficacious integrative management for *** organized a meeting of professionals in fields relevant to treatment and management of FC to develop a consensus agreement on clinical pathway ***:We developed a clinical pathway for the treatment of FC based on the most recent published guidelines and consultation with *** pathway includes a hierarchy of recommendations for every step of the clinical process,including clinical intake,diagnostic examination,recommended labs,diagnostic flowchart,and guidance for selection of therapeutic ***:This pathway establishes clinical standards for the diagnosis and treatment of FC using Chinese medicine and Western medicine;it will help to provide high-quality medical services in Hong Kong for patients with FC.

目的探讨SPARC(骨素)、cwcv和Kazal样结构域蛋白多糖1(Spock1)在肥大型与扩张型心肌病发生中的作用与分子机制。方法基于CNKI数据库明确Spock1在机体各组织中的表达情况,并获得Spock1参与信号通路的关联基因,对此关联基因进行GO和KEGG富集分析,对比同类基因与心脏疾病的关系,从而分析获得Spock1参与肥大型与扩张型心肌病的具体关联信息。结果Spock1在心脏中较高表达,并且其可能通过Adipogenesis信号通路参与肥大型心肌病(HCM)和扩张型心肌病(DCM)的发生与发展。结论Spock1可能与肥大型和扩张型心肌病等相关心血管疾病的发生密切相关,并可能成为心脏疾病发生发展的生物标记物,甚至可成为治疗心脏疾病的新靶点基因。

A new simple pyrene-substituted acylhydrazone derivative, 4-（3,4-dioctyloxy） phenyl-l-pyrene acylhydrazone （PAH-8）, was designed and synthesized. The PAH-8 can form thermo-reversible organogel in DMSO, and shows gelation-induced enhanced fluorescence emission. Xerogel exhibits ribbon-like fibrous aggregates with widths of 0.5- 1 μm. The PAH-8 organogel indicates photo-responsive behaviors due to the trans-cis isomerizations of-C = N- bond upon exposure to visible or UV light. Upon visible light irradiation, the partial trans-cis isomerization of the -C=N- bond causes the fiber morphology to disappear, resulting in gel-sol transition, whereas the PAH-8 organogel exhibits few photoisomerizations from trans to cis transition without breakage of the gel state upon UV light irradiation.

Advancing efficient and affordable electrocatalysts to boost the oxygen evolution reaction(OER)is pivotal for sustainable green hydrogen ***,we propose the fabrication of nickel-iron alloy nanoparticles-encapsulated on N-doped vertically aligned graphene array on carbon cloth(NiFe@NVG/CC)as a highly active three-dimensional(3D)catalyst electrode for *** 1 M KOH,such NiFe@NVG/CC demonstrates outstanding catalytic performance,necessitating merely overpotential of 245 mV for achieving a current density of 10 mA·cm^(−2),a remarkably low Tafel slope of 36.2 mV·dec^(−1).Furthermore,density functional theory calculations validate that the incorporate of N species into graphene can reinforce the electrocatalytic activity though reducing the reaction energy barrier during the conversion of*O to*OOH *** outstanding performance and structural benefits of NiFe@NVG/CC offer valuable insights for the development of innovative and efficient electrocatalysts for water oxidation.

Identifying pathogenetic variants and inferring their impact on protein-protein interactions sheds light on their functional consequences on *** by the availability of experimental data on the consequences of protein interaction,most existing methods focus on building models to predict changes in protein binding ***,we introduced MIPPI,an end-to-end,interpretable transformer-based deep learning model that learns features directly from sequences by leveraging the interaction data from *** was specifically trained to determine the types of variant impact(increasing,decreasing,disrupting,and no effect)on protein-protein *** demonstrate the accuracy of MIPPI and provide interpretation through the analysis of learned attention weights,which exhibit correlations with the amino acids interacting with the ***,we showed the practicality of MIPPI in prioritizing de novo mutations associated with complex neurodevelopmental disorders and the potential to determine the pathogenic and driving ***,we experimentally validated the functional impact of several variants identified in patients with such ***,MIPPI emerges as a versatile,robust,and interpretable model,capable of effectively predicting mutation impacts on protein-protein interactions and facilitating the discovery of clinically actionable variants.

In order to get direct evidence for the effect of intermolecular hydrogen bonding on the organogels, one arnide group in N-（3, 4, 5-octyloxybenzoyl）-N＇-（4＇-aminobenzoyl）hydrazine（D8） was replaced by a Schiff base group, forming N-（3,4,5-octyloxybenzoyl）-N＇-（4＇-amidobenzoyl） acylhydrazone（T8SchA）. D8 and T8SchA organogels in cyclohexane show the same hexagonal columnar structure. And the hydrogen bonding was demonstrated to be still interacting in the organogels. However, although the molecular geometry of D8 was well retained in T8SchA, the molecular dipole moment of T8SchA is bigger than that of D8 due to the reduction of the number of hydrogen bonds. Thus, the decreased gelling stability of T8SchA compared to that of D8 can only be attributed to the reduction of the number of intermolecular hydrogen bonds, which provides direct evidence that intermolecular hydrogen bonding plays an important role in stabilising organogels.

AIM: To investigate the anti-fibrosis effect of IκB kinase-beta inhibitor (IKK2 inhibitor IMD0354) in liver fibrosis. METHODS: Twenty male C57BL6 mice were divided into four groups. Five high-fat fed mice were injected with lipopolysaccharide (LPS, 10 mg/kg) intraperitoneally and five high-fat fed mice were without LPS injection to build models of liver injury, and the intervention group (five mice) was injected intraperitoneally with IKK2 inhibitor (IMD 30 mg/kg for 14 d), while the remaining five mice received a normal diet as controls. Hepatic function, pathological evaluation and liver interleukin-6 (IL-6) expression were examined. Western blotting and real-time polymerase chain reaction were used to detect the expressions of nuclear factor-κB (NF-κB), alpha-smooth muscle actin (α-SMA), tumor growth factor-beta1 (TGF-β1), tumor necrosis factor-alpha (TNF-α), typeⅠand type Ⅲ collagen proteins and mRNA. RESULTS: A mouse model of liver injury was successfully established, and IMD decreased nuclear transloca-tion of NF-κB p65 in liver cells. In the IMD-treated group, the levels of alanine aminotransferase (103 ± 9.77 μ/L vs 62.4 ± 7.90 μ/L, P < 0.05) and aminotransferase (295.8 ± 38.56 μ/L vs 212 ± 25.10 μ/L, P < 0.05) were significantly decreased when compared with the model groups. The histological changes were significantly ameliorated. After treatment, the expressions of IL-6 (681 ± 45.96 vs 77 ± 7.79, P < 0.05), TGF-β1 (Western blotting 5.65% ± 0.017% vs 2.73% ± 0.005%, P < 0.05), TNF-α (11.58% ± 0.0063% vs 8.86% ± 0.0050%, P < 0.05), typeⅠcollagen (4.49% ± 0.014% vs 1.90% ± 0.0006%, P < 0.05) and type Ⅲ collagen (3.46% ± 0.008% vs 2.29% ± 0.0035%, P < 0.05) as well as α-SMA (6.19 ± 0.0036 μ/L vs 2.16 ± 0.0023 μ/L, P < 0.05) protein and mRNA were downregulated in the IMD group compared to the fibrosis control groups (P < 0.05).
