Spinal cord injury remains a major cause of disability in young adults,and beyond acute decompression and rehabilitation,there are no pharmacological treatments to limit the progression of injury and optimize recovery in this *** the thorough investigation of the complement system in triggering and propagating cerebral neuroinflammation,a similar role for complement in spinal neuroinflammation is a focus of ongoing *** this work,we survey the current literature investigating the role of complement in spinal cord injury including the sources of complement proteins,triggers of complement activation,and role of effector functions in the *** study relevant data demonstrating the different triggers of complement activation after spinal cord injury including direct binding to cellular debris,and or activation via antibody binding to damage-associated molecular *** effector functions of complement have been implicated in spinal cord injury,and we critically evaluate recent studies on the dual role of complement anaphylatoxins in spinal cord injury while emphasizing the lack of pathophysiological understanding of the role of opsonins in spinal cord *** this pathophysiological review,we systematically review the different translational approaches used in preclinical models of spinal cord injury and discuss the challenges for future translation into human *** review emphasizes the need for future studies to dissect the roles of different complement pathways in the pathology of spinal cord injury,to evaluate the phases of involvement of opsonins and anaphylatoxins,and to study the role of complement in white matter degeneration and regeneration using translational strategies to supplement genetic models.

Clinical presentation and early predictors for poor outcomes in pediatric myocarditis:A retrospective study

BACKGROUND Myocarditis is an important cause of morbidity and mortality in children, leading to long-term sequelae including chronic congestive heart failure, dilated cardiomyopathy, heart transplantation, and death. The initial diagnosis of myocarditis is usually based on clinical presentation, but this widely ranges from the severe sudden onset of a cardiogenic shock to asymptomatic patients. Early recognition is essential in order to monitor and start supportive treatment prior to the development of severe adverse events. Of note, many cases of fulminant myocarditis are usually misdiagnosed as otherwise minor conditions during the weeks before the unexpected *** To provide diagnostic clues to make an early recognition of pediatric *** investigate early predictors for poor *** We conducted a retrospective cross-sectional single-center study from January 2008 to November 2017 at the Pediatric Department of our institution, including children < 18-years-old diagnosed with myocarditis. Poor outcome was defined as the occurrence of any of the following facts: death, heart transplant, persistent left ventricular systolic dysfunction or dilation at hospital discharge(early poor outcome), or after 1 year of follow-up(late poor outcome). We analyzed different clinical features and diagnostic test findings in order to provide diagnostic clues for myocarditis in children. Multivariable stepwise logistic regression analysis was performed using all variables that had been selected by univariate analysis to determine independent factors that predicted a poor early or late outcome in our study *** A total of 42 patients [69% male; median age of 8(1.5-12) years] met study inclusion criteria. Chest pain(40%) was the most common specific cardiac symptom. In pancreas transplantation, complications can arise at each step of the process,from the initial selection of donors and recipients through the surgical techniqueitself and the post-operative period, when lifelong immunosuppression isrequired. In the early steps, careful retrieval and preservation of the pancreas arecrucial for the viability of the organ and ultimate success of the transplant. Thepancreas is a low-flow gland, making it highly sensitive to transplantationconditions and presenting risk of pancreatitis due to periods of ischemia. The twogroups of donors - after brain death (DBD) or after cardiac arrest (DCD) - requiredifferent strategies of retrieval and preservation to avoid or reduce the risk ofcomplications developing during and after the transplantation. For DBD donortransplantation, multiorgan retrieval and cold preservation is the conventionaltechnique. Asystole donor (DCD) transplantation, in contrast, can benefit from thenewest technologies, such as hypothermic and especially normothermicpreservation machines (referred to as NECMO), to optimize organ *** latter has led to an increase in the pool of donors by facilitating recuperationof organs for transplantation that would have been discarded otherwise.

Deciphering how T-cell antigen receptor signals are modulated by coinhibitors is a fundamental goal in immunology and of considerable clinical interest because blocking coinhibitory signals via therapeutic antibodies have become a standard cancer immunotherapeutic *** of the attention devoted to T-cell immunoglobulin and mucin domain-3(TIM3;also known as HAVCR2 or CD366)molecules stems from their expression on exhausted T cells in settings of chronic viral infection and ***,T cells expressing high levels of both PD-1 and TIM3 coinhibitors appear more dysfunctional than those expressing PD-1 *** therapies intending to block both PD-1 and TIM3 are thus actively being explored in the cancer treatment *** interaction with Galectin-9(GAL-9)and carcinoembryonic antigen-related cell adhesion molecule 1(CEACAM-1),the tyrosines found in the TIM3 cytoplasmic tail are phosphorylated.1 Because these conserved tyrosines do not form a recognizable inhibitory signaling motif,the mechanism by which TIM3 transmits inhibitory signals has not been ***,TIM3 also has costimulatory activity in T cells.2,3 Published biochemical studies attempting to unveil the mode of action of TIM3 have relied on approaches addressing one candidate effector at a time with limited quantitative insight,and most used transformed *** mice expressing an affinity Twin-Strep-tag(OST)at the TIM3-protein C-terminus(TIM3OST mice)(Figs.1a and S1a)and affinity purification coupled with mass spectrometry(AP-MS),we herein defined the composition and dynamics of the signaling protein complex(signalosome)used by TIM3 in primary effector T *** results provide a more complete model of TIM3 signaling and explain its paradoxical coinhibitory and costimulatory functions.

In the present study,the green synthesis of potassium nanoparticles(K-NPs)was assessed using aqueous extract of Sideroxylon *** potassium nanoparticles were analyzed by UV-visible spectroscopic techniques,X-ray spectrometers of energy dispersive(SEM-EDS)and dynamic light *** results showed high values at 3.5 keV confirming the formation of potassium nanoparticles and the SEM analysis showed an agglomerated particles size between 360 to 200 nm with a spherical *** K-NPs showed an effective antibacterial activity against the test organisms mainly with Bacillus cereus,Enterobacter aerogenes,Fusarium solani and Botrytis *** further studies about nanotoxicity of K-NPs are needed to confirm their potential in the control of the pathogen microorganisms under field conditions.

AIM： To assess if the included vascular endothelial growth factor （VEGF） polymorphisms rs3025035, rs3025021 and rs2010963 are associated to proliferative retinopathy in a Mexican population with type 2 diabetes mellitus （T2DM）. METHODS： A case-control study was conducted in adult individuals with T2DM associated to proliferative retinopathy or non-proliferative retinopathy from Oct. 2014 to Jun. 2015 from the Retina Department of the Asociation to Prevent Blindness in Mexico. The selected patients were adults with a diagnosis of T2DM ≥5y. All subjects had a comprehensive ocular examination and the classification of the retinopathy severity was made considering the Early Treatment Diabetic Retinopathy Study （ETDRS） standardization protocols. Genomic DNA was extracted from whole fresh blood. All samples were genotyped by qPCR for selected VEGF polymorphisms. Hardy-Weinberg equilibrium was calculated by comparing Chi-square values between the expected and the observed values for genotype ***： In total 142 individuals were enrolled, 71 individuals with T2DM and associated proliferative retinopathy and 71 individuals with non-proliferative retinopathy. One-sided Fisher’s exact test was performed for rs3025021 [OR （95% CI）=0.44（0.08-2.2）; P=0.25] and rs2010963 [OR （95% CI）=0.63（0.25-1.6）; P=0.23]. The minor allelic frequencies obtained were 26% for rs3025021, 10% for rs3025035 and 61% for rs2010963. This paper presents an upper limb exoskeleton that allows cognitive(through electromyography signals)and physical user interaction(through load cells sensors)for passive and active exercises that can activate neuroplasticity in the rehabilitation process of people who suffer from a neurological *** the exoskeleton to be easily accepted by patients who suffer from a neurological injury,we used the ISO9241-210:2010 as a methodology design *** the first steps of the design process,design requirements were collected from previous usability tests and ***,as a second step,a technological solution is proposed,and as a third step,the system was evaluated through performance and user *** part of the technological solution and to allow patient participation during the rehabilitation process,we have proposed a hybrid admittance control whose input is load cell or electromyography *** hybrid admittance control is intended for active therapy exercises,is easily implemented,and does not need musculoskeletal modeling to ***,electromyography signals classification models and features were evaluated to identify the best settings for the cognitive human–robot interaction.

To determine which patients with Early Inflammatory Polyarthritis (EIP) with less than a year of evolution of the disease without a definitive diagnosis, progressed to rheumatoid arthritis according to ACR1987 criteria, we developed a predictive model for classification of early rheumatoid arthritis, based on clinical characteristics by observation of theonset of the disease and to relate with certain laboratory variables. A total of 54 patients with arthritis of less than one year of evolution were evaluated. We conducted a physical examination and the following parameters were determined: DAS 28, HAQ, Rheumatoid Factor (RF), Citrullinated Peptide (anti-CCP) and C-Telopeptide (CTx-II);radiology of hands and feet was also carried out, and was assessed by the Sharp method modified by van der Heijde. The patient follow-up was performed every 3 months for 12 months, classifying them according to the development of self-limiting, persistent non-erosive, and persistent erosive arthritis, and according to definite diagnosis. We estimated the relative risk and 95% confidence intervals for the predictor variables considered. Overall, 80.4% of patients with EIP evolved to persistent arthritis. Most persistent arthritis was diagnosed as rheumatoid arthritis (67.4%). However 51.6% (16/31) were anti-CCP positive, 21/31 (67.7%) were RF, and 11/31 (35%) were CTx-II positive. The basal nodes and RF were able to predict the persistence of activity and symmetry;rheumatoid nodules predict the development of erosions. It is important to note that patients who had an high initial average by the Sharp method modified by van der Heijde tend to have a greater increase in erosion at 6 months compared to those who had an initial low average (Pearson correlation coefficient 0.40, p < 0.015). An initial erosive disease increases the risk of radiological progression.
