KLF4 facilitates chromatin accessibility remodeling in porcine early embryos

Chromatin accessibility remodeling driven by pioneer factors is critical for the development of early embryos.Current studies have illustrated several pioneer factors as being important for agricultural animals,but what are the pioneer factors and how the pioneer factors remodel the chromatin accessibility in porcine early embryos is not clear.By employing low-input DNase-seq(liDNase-seq),we profiled the landscapes of chromatin accessibility in porcine early embryos and uncovered a unique chromatin accessibility reprogramming pattern during porcine preimplantation development.Our data revealed that KLF4 played critical roles in remodeling chromatin accessibility in porcine early embryos.Knocking down of KLF4 led to the reduction of chromatin accessibility in early embryos,whereas KLF4 overexpression promoted the chromatin openness in porcine blastocysts.Furthermore,KLF4 deficiency resulted in mitochondrial dysfunction and developmental failure of porcine embryos.In addition,we found that overexpression of KLF4 in blastocysts promoted lipid droplet accumulation,whereas knockdown of KLF4 disrupted this process.Taken together,our study revealed the chromatin accessibility dynamics and identified KLF4 as a key regulator in chromatin accessibility and cellular metabolism during porcine preimplantation embryo development.

To editor:Uterine rupture,despite its low incidence,is a severe obstetric complication that is associated with adverse maternal and neonatal outcomes.Previous uterine scarring is a known risk factor for uterine rupture.Uterine scars can form from hysterotomy after abdominal/hysteroscopic myomectomy,cesarean delivery,or hysteroscopic surgery.Over the past few decades,ultrasound-guided high-intensity focused ultrasound(USgHIFU)has emerged as a non-invasive therapeutic tool and has found wide application for the safe and effective management strategy of uterine fibroids and adenomyosis.

Continual learning fault diagnosis:A dual-branch adaptive aggregation residual network for fault diagnosis with machine increments

As a data-driven approach, Deep Learning(DL)-based fault diagnosis methods need to collect the relatively comprehensive data on machine fault types to achieve satisfactory performance. A mechanical system may include multiple submachines in the real-world. During condition monitoring of a mechanical system, fault data are distributed in a continuous flow of constantly generated information and new faults will inevitably occur in unconsidered submachines, which are also called machine increments. Therefore, adequately collecting fault data in advance is difficult. Limited by the characteristics of DL, training existing models directly with new fault data of new submachines leads to catastrophic forgetting of old tasks, while the cost of collecting all known data to retrain the models is excessively high. DL-based fault diagnosis methods cannot learn continually and adaptively in dynamic environments. A new Continual Learning Fault Diagnosis method(CLFD) is proposed in this paper to solve a series of fault diagnosis tasks with machine increments. The stability–plasticity dilemma is an intrinsic issue in continual learning. The core of CLFD is the proposed Dual-branch Adaptive Aggregation Residual Network(DAARN).Two types of residual blocks are created in each block layer of DAARN: steady and dynamic blocks. The stability–plasticity dilemma is solved by assigning them with adaptive aggregation weights to balance stability and plasticity, and a bi-level optimization program is used to optimize adaptive aggregation weights and model parameters. In addition, a feature-level knowledge distillation loss function is proposed to further overcome catastrophic forgetting. CLFD is then applied to the fault diagnosis case with machine increments. Results demonstrate that CLFD outperforms other continual learning methods and has satisfactory robustness.

Synchronous chirp mode extraction: A promising tool for fault diagnosis of rolling element bearings under varying speed conditions

As critical components in modern aerospace productions,rolling element bearings(REBs)generally work under varying speed conditions,which brings great challenges to their operating health monitoring.Some novel time–frequency decomposition(TFD)algorithms are established recently to extract nonlinear features from the non-stationary signals effectively,which are promising for realizing fault diagnosis of REBs under varying speed conditions.However,numerous personal experiences must be incorporated and the anti-noise performance of these methods needs to be further enhanced.Given these issues,a synchronous chirp mode extraction(SCME)-based REB fault diagnosis method is proposed for the health monitoring of REBs under varying speed conditions in this study.It mainly consists of following two parts.(a)The shaft rotational frequency(SRF)is initially estimated from the low-frequency band of the vibration signal.Simultaneously,an adaptive refining strategy is incorporated to obtain a suitable bandwidth parameter.(b)A cycle-one-step estimation frame is constructed to extract synchronous modes from the envelope waveform of the vibration signal.Meanwhile,a synchronous mode spectrum(SMS)is generated using the information of the extracted synchronous modes,which is a novel REBs fault diagnosis technique with tacholess and resampling-free.In contrast to the current TFD algorithms,the proposed method needs fewer input parameters and owns a well anti-noise performance because there is no iterative optimization in the procedure of construction of SMS.As a result,the health conditions of REBs are evaluated by detecting the exhibited features in the SMS.Simulations and experiments are conducted to validate the effectiveness of the proposed method in terms of REB fault diagnosis.Analysis results demonstrate that the proposed method outperforms the current TFD algorithm and the conventional order tracking technique for fault diagnosis of REB under varying speed conditions.

Quantization and Event-Triggered Policy Design for Encrypted Networked Control

This paper proposes a novel event-driven encrypted control framework for linear networked control systems(NCSs),which relies on two modified uniform quantization policies,the Paillier cryptosystem,and an event-triggered strategy.Due to the fact that only integers can work in the Pailler cryptosystem,both the real-valued control gain and system state need to be first quantized before encryption.This is dramatically different from the existing quantized control methods,where only the quantization of a single value,e.g.,the control input or the system state,is considered.To handle this issue,static and dynamic quantization policies are presented,which achieve the desired integer conversions and guarantee asymptotic convergence of the quantized system state to the equilibrium.Then,the quantized system state is encrypted and sent to the controller when the triggering condition,specified by a state-based event-triggered strategy,is satisfied.By doing so,not only the security and confidentiality of data transmitted over the communication network are protected,but also the ciphertext expansion phenomenon can be relieved.Additionally,by tactfully designing the quantization sensitivities and triggering error,the proposed event-driven encrypted control framework ensures the asymptotic stability of the overall closedloop system.Finally,a simulation example of the secure motion control for an inverted pendulum cart system is presented to evaluate the effectiveness of the theoretical results.

Adaptive Space Expansion for Fast Motion Planning

The sampling process is very inefficient for sam-pling-based motion planning algorithms that excess random sam-ples are generated in the planning space.In this paper,we pro-pose an adaptive space expansion(ASE)approach which belongs to the informed sampling category to improve the sampling effi-ciency for quickly finding a feasible path.The ASE method enlarges the search space gradually and restrains the sampling process in a sequence of small hyper-ellipsoid ring subsets to avoid exploring the unnecessary space.Specifically,for a con-structed small hyper-ellipsoid ring subset,if the algorithm cannot find a feasible path in it,then the subset is expanded.Thus,the ASE method successively does space exploring and space expan-sion until the final path has been found.Besides,we present a particular construction method of the hyper-ellipsoid ring that uniform random samples can be directly generated in it.At last,we present a feasible motion planner BiASE and an asymptoti-cally optimal motion planner BiASE*using the bidirectional exploring method and the ASE strategy.Simulations demon-strate that the computation speed is much faster than that of the state-of-the-art algorithms.The source codes are available at https://***/shshlei/ompl.

Impact of frailty on outcomes of elderly patients undergoing percutaneous coronary intervention: A systematic review and metaanalysis

BACKGROUND Frailty is a common condition in elderly patients who receive percutaneous coronary intervention(PCI).However,how frailty affects clinical outcomes in this group is unclear.AIM To assess the link between frailty and the outcomes,such as in-hospital complic-ations,post-procedural complications,and mortality,in elderly patients post-PCI.METHODS The PubMed/MEDLINE,EMBASE,Cochrane Library,and Web of Science databases were screened for publications up to August 2023.The primary outcomes assessed were in-hospital and all-cause mortality,major adverse cardiovascular events(MACEs),and major bleeding.The Newcastle-Ottawa Scale was used for quality assessment.RESULTS Twenty-one studies with 739693 elderly patients undergoing PCI were included.Frailty was consistently associated with adverse outcomes.Frail patients had significantly higher risks of in-hospital mortality[risk ratio:3.45,95%confidence interval(95%CI):1.90-6.25],all-cause mortality[hazard ratio(HR):2.08,95%CI:1.78-2.43],MACEs(HR:2.92,95%CI:1.85-4.60),and major bleeding(HR:4.60,95%CI:2.89-7.32)compared to non-frail patients.CONCLUSION Frailty is a pivotal determinant in the prediction of risk of mortality,development of MACEs,and major bleeding in elderly individuals undergoing percutaneous coronary intervention.

No Reference Image Sharpness Assessment Based on Global Color Difference Variation

Image quality assessment(IQA) model is designed to measure the image quality in consistent with subjective ratings by computational models. In this research, a valid no reference IQA(NR-IQA) model for color image sharpness assessment is proposed based on local color difference map in a color space. In the proposed model, the absolute color difference variation and relative color difference variation are combined to evaluate sharpness in YIQ color space(a color coordinate system for the development of the United States color television system). The difference between sharpest and blurriest spot of an image is represented by the absolute color difference variation, and relative color difference variation expresses the variation in the image content. Extensive experiments are performed on five publicly available benchmark synthetic blur databases and two real blur databases, and the results prove that the proposed model work better than the other state-of-the-art and latest NR-IQA models for the prediction accuracy on blurry images. Besides, the model maintains the lowest computational complexity.

An uncertainty investigation for liner impedance eduction methods

Impedance eduction methods have been developed for decades to meet the increasing need for high-quality impedance data in the design and optimization of acoustic liners.To this end,it is important to fully investigate the uncertainty problem,to which only limited attention has been devoted so far.This paper considers the possibility of acoustically-induced structural vibration as a nonnegligible uncertainty or error source in impedance eduction experiments.As the frequency moves away from the resonant frequency,with the increase in the value of cavity reactance,the acoustic particle velocity inside liner orifices possibly decreases to the extent comparable to the vibration velocity of liner facing sheet.Thus,the acoustically-induced vibration,although generally being weak except at the inherent structural frequencies,may considerably affect the impedance eduction results near the anti-resonant frequency where the liner has poor absorption.To demonstrate the effect of structural vibration,the vibration velocity of liner facing sheet is estimated from the experimentally educed admittance of the liner samples whose orifices are sealed with tape.Further,a three-dimensional numerical model is set up,in which normal particle velocity is introduced over the solid portion of liner facing sheet to imitate structural vibration,rather than directly solving the acoustic-structural coupling problem.As shown by the results,the vibration of liner facing sheet,whose velocity is as small as estimated by the experiment,can result in anomalous deviation of the educed impedance from the impedance model near the anti-resonant frequency.The trend that the anomalous deviation varies with frequency is numerically captured.

BACKGROUND Prohibitin 1(PHB1)has been identified as an antiproliferative protein that is highly conserved and ubiquitously expressed,and it participates in a variety of essential cellular functions,including apoptosis,cell cycle regulation,prolifera-tion,and survival.Emerging evidence indicates that PHB1 may play an important role in the progression of hepatocellular carcinoma(HCC).However,the role of PHB1 in HCC is controversial.AIM To investigate the effects of PHB1 on the proliferation and apoptosis of human HCC cells and the relevant mechanisms in vitro.METHODS HCC patients and healthy individuals were enrolled in this study according to the inclusion and exclusion criteria;then,PHB1 levels in the sera and liver tissues of these participates were determined using ELISA,RT-PCR,and immunohistoche-mistry.Human HepG2 and SMMC-7721 cells were transfected with the pEGFP-PHB1 plasmid and PHB1-specific shRNA(shRNA-PHB1)for 24-72 h.Cell prolif-eration was analysed with an MTT assay.Cell cycle progression and apoptosis were analysed using flow cytometry(FACS).The mRNA and protein expression levels of the cell cycle-related molecules p21,Cyclin A2,Cyclin E1,and CDK2 and the cell apoptosis-related molecules cytochrome C(Cyt C),p53,Bcl-2,Bax,caspase 3,and caspase 9 were measured by real-time PCR and Western blot,respectively.RESULTS Decreased levels of PHB1 were found in the sera and liver tissues of HCC patients compared to those of healthy individuals,and decreased PHB1 was positively correlated with low differentiation,TNM stage III-IV,and alpha-fetoprotein≥400μg/L.Overexpression of PHB1 significantly inhibited human HCC cell proliferation in a time-dependent manner.FACS revealed that the overexpression of PHB1 arrested HCC cells in the G0/G1 phase of the cell cycle and induced apoptosis.The proportion of cells in the G0/G1 phase was significantly increased and the proportion of cells in the S phase was decreased in HepG2 cells that were transfected with pEGFP-PHB1 compared with untreated control and empty vector-trans
