AIM： To assess whether portacaval anastomosis （PCA） in rats affects the protein expression and/or activity of glutaminase in kidneys, intestines and in three brain areas of cortex, basal ganglia and cerebellum and to explain the neurological alterations found in hepatic encephalopathy （HE）. METHODS： Sixteen male Wistar rats weighing 250-350 g were grouped into sham-operation control （n=8） or portacaval shunt （n = 8）. Twenty-eight days after the procedure, the animals were sacrificed. The duodenum, kidney and brain were removed, homogenised and mitochondria were isolated. Ammonia was measured in brain and blood. Phosphate-activated glutaminase （PAg） activity was determined by measuring ammonia production following incubation for one hour at 37 ℃ with O-phthalaldehyde （OPA） and specific activity expressed in units per gram of protein （pkat/g of protein）. Protein expression was measured by immunoblotting. RESULTS： Duodenal and kidney PAg activities together with protein content were significantly higher in PCA group than in control or sham-operated rats （duodenum PAg activity was 976.95±268.87μkat/g of protein in PCA rats vs 429.19±126.92.μkat/g of protein in shamoperated rats; kidneys PAg activity was 1259.18±228.79 μkat/g protein in PCA rats vs 669.67±400.8 μkat/g of protein in controls, P〈0.05; duodenal protein content： 173% in PCA vs sham-operated rats; in kidneys the content of protein was 152% in PCA vs sham-operated rats）. IBD5 polymorphisms in inflammatory bowel disease: Association with response to infliximab

AIM: Inflammatory bowel diseases (IBD) are multifactorial pathologies of unknown etiology. One susceptibility locus, IBD5, has been mapped to chromosome 5q31. We analyzed our Spanish cohorts of Crohn's disease (CD) and ulcerative colitis (DC) patients to determine whether this locus is associated with IBD, and to ascertain the main clinical phenotype influenced by this risk factor. The kind of interaction, either genetic heterogeneity or epistasis, between this IBD5 susceptibility region and the NOD2/CARD15 gene mutations was studied as well. Finally, we assessed whether this locus can predict response to infliximab therapy. METHODS: A case control study was performed with 274 CD and 211 UC patients recruited from a single center and 511 healthy ethnically matched controls. Two polymorphisms were genotyped in the IBD5 locus and three in the CARD15/NOD2 gene. RESULTS: Our results evidence association only with CD especially with the fistulizing phenotype and in the absence of NOD2/CARD15 variants (mutant allele frequency in patients vs controls: OR = 2.03, 95% CI = 1.35-3.06, Pgous mutant genotype significantly increased in CD patients lacking response to infliximab (RR = 3.88, 95% CI = 1.18-12.0, Pgh a similar trend to the one observed in CD is found within the worse prognosis group. CONCLUSION: The IBD5 variants may enhance an individual carrier's risk for CD, mainly in the absence of the NOD2/CARD15 mutations and in fistulizing patients. The data presented suggest the potential role of the 5q31 polymorphisms as markers of response to infliximab.

Mammillaria pectinifera is an endemic, short-globose cactus species, included in the IUCN list as a threatened species with only 18 remaining populations in the Tehuacfin-Cuicatlan Valley in central Mexico. We evaluated the population genetic diversity and structure, connectivity, recent bottlenecks and population size, using nuclear microsatellites. M. pectinifera showed high genetic diversity but some evidence of heterozygote deficiency （F15）, recent bottlenecks in some populations and reductions in population size. Also, we found low population genetic differentiation and high values of connectivity for M. pectinifera, as the result of historical events of gene flow through pollen and seed dispersal. M. pectinifera occurs in sites with some degree of disturbance leading to the isolation of its populations and decreasing the levels of gene flow among them. Excessive deforestation also changes the original vegetation damaging the natural habitats. This species will become extinct if it is not properly preserved. Furthermore, this species has some ecological features that make them more vulnerable to disturbance such as a very low growth rates and long life cycles. We suggest in situ conservation to prevent the decrease of population sizes and loss of genetic diversity in the natural protected areas such as the Tehuacan-Cuicatlan Biosphere Reserve. In addition, a long-term ex situ conservation program is need to construct seed banks, and optimize seed germination and plant establishment protocols that restore disturbed habitats. Furthermore, creating a supply of living plants for trade is critical to avoid further extraction of plants from nature.

PSTCNN: Explainable COVID-19 diagnosis using PSO-guided self-tuning CNN

Since 2019,the coronavirus disease-19(COVID-19)has been spreading rapidly worldwide,posing an unignorable threat to the global economy and human *** is a disease caused by severe acute respiratory syndrome coronavirus 2,a single-stranded RNA virus of the genus *** virus is highly infectious and relies on its angiotensin-converting enzyme 2-receptor to enter *** the increase in the number of confirmed COVID-19 diagnoses,the difficulty of diagnosis due to the lack of global healthcare resources becomes increasingly *** learning-based computer-aided diagnosis models with high generalisability can effectively alleviate this *** tuning is essential in training such models and significantly impacts their final performance and training ***,traditional hyperparameter tuning methods are usually time-consuming and *** solve this issue,we introduce Particle Swarm Optimisation to build a PSO-guided Self-Tuning Convolution Neural Network(PSTCNN),allowing the model to tune hyperparameters ***,the proposed approach can reduce human ***,the optimisation algorithm can select the combination of hyperparameters in a targeted manner,thus stably achieving a solution closer to the global ***,the PSTCNN can obtain quite excellent results,with a sensitivity of 93.65%±1.86%,a specificity of 94.32%±2.07%,a precision of 94.30%±2.04%,an accuracy of 93.99%±1.78%,an F1-score of 93.97%±1.78%,Matthews Correlation Coefficient of 87.99%±3.56%,and Fowlkes-Mallows Index of 93.97%±1.78%.Our experiments demonstrate that compared to traditional methods,hyperparameter tuning of the model using an optimisation algorithm is faster and more effective.

Home range variability and philopatry in Cinereous vultures(Aegypius monachus)breeding in Iberia

Large scavengers are strongly dependent on environmental conditions and carrion distribution and abundance,so season and breeding-related factors may influence the spatial ecology of species such as the Cinereous Vulture(Aegypius monachus),the largest European *** holds one of the biggest populations worldwide,but some aspects of the spatial ecology of the species in this region remain *** this study,17 adult Cinereous Vultures were gPS-tracked in order to study their spatial ecology during the adult *** average monthly home ranges(95%Kernel Density Estimation,KDE)and core areas(50%KDE)were 6543±19,935 km^(2)and 1174±4004 km^(2),*** average monthly home range fidelity ranged between 50 and 73%.Differences in movement-related variables between the seasonal periods(incubation,chick-rearing and non-breeding)were *** the chick-rearing period,the monthly accumulated distance was higher than during the other periods:3316±1108(chick-rearing)vs.1621±622(incubation)vs.1726±1159 km per month(nonbreeding).Additionally,large home range sizes were more frequent during the chick-rearing *** are two likely causes for these seasonal ***,chick-rearing entails a higher energetic expenditure by the parental individuals in foraging activities,so larger movements and foraging areas are expected during this ***,the flight is favoured during spring and summer due to environmental *** chick-rearing and warm months is a great evolutionary advantage for soaring-gliding raptors,as it allows them to cover larger areas with low energy ***,six individuals tagged as nestlings highlight the philopatric behaviour of the species:vultures settle their breeding areas 54±51 km from their natal nest(range=9–138 km).
