Expression of an endoglucanase from Tribolium castaneum （TcEG 1 ） in saccharomyces cerevisiae

Insects are a largely unexploited resource in prospecting for novel cellulolytic enzymes to improve the production of ethanol fuel from lignocellulosic biomass. The cost of lignocellulosic ethanol production is expected to decrease by the combination of cellulose degradation （saccharification） and fermentation of the resulting glucose to ethanol in a single process, catalyzed by the yeast saccharomyces cerevisiae transformed to express efficient cellulases. While s. cerevisiae is an established heterologous expression system, there are no available data on the functional expression of insect cellulolytic enzymes for this species. To address this knowledge gap, s. cerevisiae was transformed to express the full-length cDNA encoding an endoglucanase from the red flour beetle, Tribolium castaneum （TcEG 1）, and evaluated the activity of the transgenic product （rTcEG 1）. Expression of the TcEG1 cDNA in s. cerevisiae was under control of the strong glyceraldehyde-3 phosphate dehydrogenase promoter. Cultured transformed yeast secreted rTcEG1 protein as a functional β-1,4-endoglucanase, which allowed transformants to survive on selective media containing cellulose as the only available carbon source. Evaluation of substrate specificity for secreted rTcEG1 demonstrated endoglucanase activity, although some activity was also detected against complex cellulose substrates. Potentially relevant to uses in biofuel production rTcEG1 activity increased with pH conditions, with the highest activity detected at pH 12. Our results demonstrate the potential for functional production of an insect cellulase in s. cerevisiae and confirm the stability of rTcEG1 activity in strong alkaline environments.

District energy systems(DEs)offer an optimal solution for decarbonising the heating and cooling sector while attaining multiple additional *** first step to analyse the potential of DEs in both new and existing markets is through rapid assessments(RA).Currently,publicly available models lack rapid assessments of the technical-economic and environmental potential of *** RA model was developed within the framework of UNEP’s District Energy in Cities Initiative to identify DEs’s potential spending low time and monetary *** this light,the study presents a model for conducting a rapid assessment applied to the case of Temuco,*** show that a total of 55 MW DH(district heating)capacity is required to cover the heating demand.A wood-chip boiler of 25 MW capacity and a gas boiler of 30 MW capacity are considered in the *** total CAPEX of the project is around 25 billion CLP,with a NPV of 10.5 billion CLP and an IRR of 14%.The project is also estimated to achieve an annual reduction of 24,382 tons of PM10 and 23,692 tons of *** model was validated against an independent study conducted by an international consulting company,and the results were found to be in close proximity with the ***,the model can be an effective tool for performing rapid assessments of DEs projects in the region and subjecting attractive projects to more detailed pre-feasibility analysis.

Oral allopurinol to prevent hyperamylasemia and acute pancreatitis after endoscopic retrograde cholangiopancreatography

AIM:To assess the efficacy of allopurinol to prevent hyperamylasemia and pancreatitis after endoscopic retrograde cholangiopancreatography(PEP).METHODs:One hundred and seventy patients were enrolled and randomized to two groups:a study group(n=85)who received 300 mg of oral allopurinol at 15 h and 3 h before endoscopic retrograde cholangiopancreatography(ERCP)and a control group(n=85)receiving an oral placebo at the same *** Outcome Measurements included serum amylase levels and the number severity of the episodes of *** amylase levels were classified as normal(semia(>151 IU/L).Episodes of PEP were classified following Ranson's criteria and CT severity ***:Gender distribution was similar between *** age was 53.5±18.9 years for study group and 52.8±19.8 years for ***,the distribution of benign pathology was similar between *** was more common in the control group(P=0.003).Mild PEP developed in two patients from the study group(2.3%)and eight(9.4%) from control group(P=0.04),seven episodes were observed in high-risk patients of the control group(25%) and one in the allopurinol group(3.3%,P=0.02).Risk factors for PEP were precut sphincterotomy(P=0.02),pancreatic duct manipulation(P=0.002)and multiple procedures(P=0.000).There were no deaths or side ***:Oral allopurinol before ERCP decreased the incidences of hyperamylasemia and pancreatitis in patients submitted to high-risk procedures.

Determination of Radioactive Contaminant <sup>137sup>Cs (Fission Product) and Comparison with Natural <sup>40sup>K Present in Mexican strawberries

several previous papers have been published about radioactive contamination in Mexican soil, marine sediments and foodstuffs. Ever since, results have been according to expectation, mainly consisting of natural isotopes in small concentration. Continuous research leads with the surprising finding of fission product sup>137sup>Cs (tsub>1/2sub> = 30 years) in strawberries produced in different states, about 2000 km away from each other. Never before, such contaminant has been observed in foodstuffs, produced in this country. The aim of this work is to report the presence of sup>137sup>Cs in Mexican strawberries and compare the activity of such contaminant with the natural radioactivity of sup>40sup>K (tsub>1/2sub> = 1.28 × 10sup>9sup> years), through gamma spectrometry. Eventually, the specific activity of sup>137sup>Cs observed in samples FM4, FM5 and FBC is respectively 0.7, 0.8 and 0.3 Bq/kg. The limits established in CODEX-sTAN-193-1995 for sup>137sup>Cs are 1000 Bq/kg, so all samples meet the recommendation. It’s presence now in vegetables result with enough interest to be known and considered in the near future to measure the possible effect that radioactive contamination could reach after nearly 80 years from second world war, where no nuclear test has been performed, nuclear accident occurred, neither nuclear weapon has ever been used.

The Immune system and the Role of Inflammation in Perinatal Depression

Major depression during pregnancy is a common psychiatric disorder that arises from a complex and mul- tifactorial etiology. Psychosocial stress, sex, hormones, and genetic vulnerability increase the risk for triggering mood disorders. Microglia and toll-like receptor 4 play a crucial role in triggering wide and varied stress-induced responses mediated through activation of the inflammasome; this leads to the secretion of inflammatory cytokines, increased serotonin metabolism, and reduction of neurotransmitter availability along with hypothalamic-pituitary-adrenal axis hyperactivity. Dysregulation of this intricate neu- roimmune communication network during pregnancy modifies the maternal milieu, enhancing the emergence of depressive symptoms and negative obstetric and neu- ropsychiatric outcomes. Although several studies have clearly demonstrated the role of the innate immune system in major depression, it is still unclear how the placenta, the brain, and the monoaminergic and neuroendocrine systems interact during perinatal depression. Thus, in the present review we describe the cellular and molecular interactions between these systems in major depression during preg- nancy, proposing that the same stress-related mechanisms involved in the activation of the NLRP3 inflammasome in microglia and peripheral myeloid cells in depressed patients operate in a similar fashion in the neuroimmune placenta during perinatal depression. Thus, activation of Toll-like receptor 2 and 4 signaling and the NLRP3 inflammasome in placental immune cells may promote a shift of the Thl/Th2 bias towards a predominant Thl/Thl7 inflammatory response, associated with increased secretion of pro-inflammatory cytokines, among other secreted autocrine and paracrine mediators, which play a crucial role in triggering and/or exacerbating depressive symptoms during pregnancy.

Lightweight Approach to Connect Business Rules with Aspects

Aspectual connection that compose business rule, refers to the code in charge of not only triggering the application of the rules at certain events, but also gathering the necessary information for their application and incorporating their results in the rest of the core application functionality. We propose an approach to connect business rule with aspect systematically. This approach is agile, programming language-independent, business rules type independent and it can be used in different stages of development. This approach considers the identification, analysis and construction of connections with aspects, (aspectual connection). We use our experience and previous works to define the initial steps, and complete them with the analysis of interactions and propose methods to implement aspectual connections code. We have developed a tool that supports this approach.

According to the main international clinical guidelines,the recommended treatment for locally-advanced rectal cancer is neoadjuvant chemoradiotherapy followed by ***,doubts have been raised about the appropriate definition of clinical complete response(cCR)after neoadjuvant therapy and the role of surgery in patients who achieve a *** resection is associated with significant morbidity and decreased quality of life(QoL),which is especially relevant given the favourable prognosis in this patient subset. Accordingly, therehas been a growing interest in alternative approaches with less morbidity,including the organ-preserving watch and wait strategy, in which surgery isomitted in patients who have achieved a cCR. These patients are managed with aspecific follow-up protocol to ensure adequate cancer control, including the earlyidentification of recurrent disease. However, there are several open questionsabout this strategy, including patient selection, the clinical and radiologicalcriteria to accurately determine cCR, the duration of neoadjuvant treatment, therole of dose intensification (chemotherapy and/or radiotherapy), optimal followupprotocols, and the future perspectives of this approach. In the present review,we summarize the available evidence on the watch and wait strategy in thisclinical scenario, including ongoing clinical trials, QoL in these patients, and thecontroversies surrounding this treatment approach.

Facial Asymmetry Correction in Facial Palsy Patients with silhouette sutures

Introduction: over the last few decades several techniques static and dynamics, have been performed to improve facial asymmetry and functionality alter suffering facial paralysis. Methods: we present a pilot study to show and evaluate the benefits of a new form of facial suspension, with silhouette sutures. We performed two patients with total and complete facial palsy due to otical tuberculosis in one case and to parotid carcinoma in the other. Results: one year after surgery, both patients have improved facial asymmetry, mastication and speech production which have lead to a higher self-esteem and major social interaction. Conclusions: static facial suspension with silhouette sutures is a non invasive alternative to dynamic techniques in patients who don’t want or can’t undergo more complex surgeries.

Albumin-bound paclitaxel as new treatment for metastatic cholangiocarcinoma: A case report

BACKGROUND Cholangiocarcinomas are rare and very aggressive *** patients have advanced-stage or unresectable disease at presentation,and the systemic therapies have limited ***-bound paclitaxel(nab-paclitaxel)is a solvent-free taxane that has been approved for the treatment of some cancers such as breast,non-small cell lung and pancreatic cancer,however it has not been applied to treat *** have both preclinical and clinical evidence of the efficacy of nab-paclitaxel in cholangiocarcinoma,yet no phase 3 trials have been *** sUMMARY A 63-year-old man was diagnosed in December 2016 with stage III B intrahepatic *** was performed,followed by adjuvant chemotherapy treatment with capecitabine and gemcitabine;although,the gemcitabine was suspended due to allergic reaction after two *** April 2019,metastatic cholangiocarcinoma relapse was diagnosed,and a first-line treatment with FOLFOX scheme was *** cycles were administered,producing an initial clinical improvement and decrease in blood tumor marker *** and serological progression was noted in september *** a second-line treatment,FOLFIRI was not recommended due to risk of worsening the patient’s tumor-related diarrhea.A combination therapy with gemcitabine was not feasible,as the patient had previously suffered from an allergic reaction to this *** decided to use nab-paclitaxel as a second-line treatment,and four cycles were *** clinical and serological responses were observed,and a radiological mixed response was also *** Advanced cholangiocarcinoma could be treated with nab-paclitaxel monotherapy,which should be studied in combination with other types of treatment(chemotherapy,fibroblast growth factor receptor inhibitors).

Implications of Klotho in vascular health and disease

Cardiovascular disease(CVD) is a prevalent condition in general population and the first cause of death overall. Klotho, a pleiotropic protein related to longevity that acts as a co-receptor of the fibroblast growth factor 23, has been proposed as a key regulator of the development of CVD. In the few clinical studies made, it has been observed a relationship between low levels of soluble Klotho and the occurrence and severity of CVD, as well as a reduction of cardiovascular risk when they are high. Also, different polymorphisms of human Klotho gene have been related to the incidence of cardiovascular events. Moreover, several experimental studies indicate that this protein acts in the maintenance of vascular homeostasis. Klotho improves endothelial dysfunction through promotion of NO production and mediates antiinflammatory and anti-aging effects such as suppression of adhesion molecules expression, attenuation of nuclear factor-kappa B or inhibition of Wnt signaling. Furthermore,this protein is related to the attenuation of vascular calcification as well as prevention of cardiac hypertrophy. The expression of this protein in the vascular wall implies a new scenario for the treatment of vascular disorders. The purpose of this review is to provide an overview of the relationship between the Klotho protein and CVD, in addition to its role in the maintenance of functional vascular integrity.
