In vivo testing of mucus-permeating nanoparticles for oral insulin delivery using Caenorhabditis elegans as a model under hyperglycemic conditions

The aim was to evaluate the potential of mucus-permeating nanoparticles for the oral administration of insulin.These nanocarriers,based on the coating of zein nanoparticles with a polymer conjugate containing PEG,displayed a size of 260 nm with a negative surface charge and an insulin payload of 77 mg/mg.In intestinal pig mucus,the diffusivity of these nanoparticles(PPA-NPs)was found to be 20-fold higher than bare nanoparticles(NPs).These results were in line with the biodistribution study in rats,in which NPs remained trapped in the mucus,whereas PPA-NPs were able to cross this layer and reach the epithelium surface.The therapeutic effcacy was evaluated in Caenorhabditis elegans grown under high glucose conditions.In this model,worms treated with insulin-loaded in PPA-NPs displayed a longer lifespan than those treated with insulin free or nanoencapsulated in NPs.This finding was associated with a signifcant reduction in the formation of reactive oxygen species(ROS)as well as an important decrease in the glucose and fat content in worms.These effects would be related with the mucus-permeating ability of PPA-NPs that would facilitate the passage through the intestinal peritrophic-like dense layer of worms(similar to mucus)and,thus,the absorption of insulin.

Early life 1000 days: opportunities for preventing adult diseases

With the rapid socioeconomic development, improvements in living standards, and changes in lifestyle, adult diseases, such as cardiovascular diseases, type 2 diabetes mellitus (T2DM), hypertension, obesity, and so on, are increasing dramatically over the past decades, which have caused a huge economic and health burden at both population and individual levels in China.[1] The increased burden of adult diseases has become the main public health issue, and prevention of adult diseases in China has become a formidable task during recent years. In 2016, China has put forward the concept of whole life-course health and efforts have been made on shifting the focus from treating diseases to prevention. In view of the poor effects of prevention of diseases in adulthood, it is suggested that prevention and management of adult diseases should be extended to early life, which is the critical window for physiological programming and has profound and enduring effects on the whole life-cycle health.[2,3,4] In 2018, an article named "To Prevent Cardiovascular Disease, Pay Attention to Pregnancy Complications" published in Journal of the American Medical Association (JAMA) also illustrated that pregnancy is a window to future health[5].

Quality assessment of the Topo-Iberia CGPS stations and data quality's effects on postfit ionosphere-free phase residuals

This paper analyzed GPS data from the Topo-Iberia network spanning almost 12 years(2008-2020).The data quality information for all 26 Topo-Iberia stations is provided for the first time,complementing the Spanish Geological Survey’s storage work.Data analyses based on quality indicators obtained using TEQC have been carried out.The guidelines and data quality information from the IGS stations have been considered as the quality references,with the stations ALJI,EPCU,and TIOU standing out as the worst stations,while on the contrary,FUEN,PALM,PILA,and TRIA meet the quality requirements to become an IGS station.The relationship between the GPS data quality and their GAMIT-and Gipsy X-derived postfit ionosphere-free phase residuals has also been investigated,and the results reveal an inversely proportional relationship.It has been found that the stations showing an increase in elevation of the horizon line,also show an increase in cycle slips and multipath,are among the poorest quality stations,and among those with the highest postfit RMS of phase residuals.Moreover,the evolution of the vegetation around the antenna should be considered as it could cause a progressive loss of quality,which is not complying with the IGS standards.The quality assessment shows that the Topo-Iberia stations are appropriate for geodetic purposes,but permanent monitoring would be necessary to avoid the least possible loss of data and quality.In addition,a method to characterize the GNSS data quality is proposed.

Boundary Control of Coupled Non-Constant Parameter Systems of Time Fractional PDEs with Different-Type Boundary Conditions

This paper addresses a boundary state feedback control problem for a coupled system of time fractional partial differential equations(PDEs)with non-constant(space-dependent)coefficients and different-type boundary conditions(BCs).The BCs could be heterogeneous-type or mixed-type.Specifically,this coupled system has different BCs at the uncontrolled side for heterogeneous-type and the same BCs at the uncontrolled side for mixed-type.The main contribution is to extend PDE backstepping to the boundary control problem of time fractional PDEs with space-dependent parameters and different-type BCs.With the backstepping transformation and the fractional Lyapunov method,the Mittag-Leffler stability of the closed-loop system is obtained.A numerical scheme is proposed to simulate the fractional case when kernel equations have not an explicit solution.

Intensive care unit-acquired weakness(ICU-AW)is a common complication in critically ill patients and is associated with a variety of adverse outcomes.These include the need for prolonged mechanical ventilation and ICU stay;higher ICU,in-hospital,and 1-year mortality;and increased in-hospital costs.ICU-AW is associated with multiple risk factors including age,underlying disease,severity of illness,organ failure,sepsis,immobilization,receipt of mechanical ventilation,and other factors related to critical care.The pathological mechanism of ICUAW remains unclear and may be considerably varied.This review aimed to evaluate recent insights into ICU-AW from several aspects including risk factors,pathophysiology,diagnosis,and treatment strategies;this provides new perspectives for future research.

Ni nanoparticles confined by yolk-shell structure of CNT-mesoporous carbon for electrocatalytic conversion of CO_(2): Switching CO to formate

Electrochemical reduction of CO_(2)(CO_(2)ER) to formate has been a promising route to produce value-added chemicals.Developing low-cost and efficient electrocatalysts with high product selectivity is still a grand challenge.Herein,a novel Ni nanoparticles-anchored CNT coated by mesoporous carbon with yolk-shell structure (CNT/Ni@mC) catalysis was designed for CO_(2)ER.Ni nanoparticles were confined in the cavity between CNT and mesoporous carbon shell and the confined space can be controlled by tuning the amount of silica precursor.The mesoporous carbon shell and confined space are beneficial to charge transmission during CO_(2)ER.In contrast to previous studies,the CNT/Ni@mC catalyst presents selectivity toward formate rather than CO.Electrochemical in situ attenuated total reflection Fourier transform infrared spectroscopy measurements indicate the presence of a COO* intermediate that converts to formate under CO_(2)ER conditions.The well-defined structural feature of the confined space of the Ni-based catalyst for selective CO_(2)ER to formate may facilitate in-depth mechanistic understandings on structural factors that affect CO_(2)ER performance.

The Initial Mesoscale Vortexes Leading to the Formation of Tropical Cyclones in the Western North Pacific

A statistical analysis of the initial vortexes leading to tropical cyclone(TC)formation in the western North Pacific(WNP)is conducted with the ECMWF ERA5 reanalysis data from 1999 to 2018.It is found that TCs in the WNP basically originate from three kinds of vortexes,i.e.,a mid-level vortex(MV),a low-level vortex(LV),and a relatively deep vortex with notable vorticity in both the lower and middle troposphere(DV).Among them,LV and DV account for 47.9%and 24.2%of tropical cyclogenesis events,respectively,while only 27.9%of TCs develop from the MV,which is much lower than that which occurs in the North Atlantic and eastern Pacific.Such a difference might be ascribed to the active monsoon systems in the WNP all year round.Due to the nearly upright structure of mid-level convergence in the early pre-genesis stage,TC genesis efficiency is the highest in DV.Compared with MV,LV generally takes a shorter time to intensify to a TC because of the higher humidity and the stronger low-level cyclonic circulation,which is related to air-sea interaction and boundary-layer convergence.Further examination of the relationship between tropical cyclogenesis and large-scale flow patterns indicate that the TC genesis events associated with LV are primarily related to the monsoon shear line,monsoon confluence region,and monsoon gyre,while those associated with MV are frequently connected with easterly waves and wave energy dispersion of preexisting TC.Compared with other flow patterns,tropical cyclones usually form and intensify faster in the monsoon confluence region.

Cerebral amyloid angiopathy-related inflammation: current status and future implications

Cerebral amyloid angiopathy-related inflammation (CAA-RI) is a rare but increasingly recognized subtype of CAA. CAA-RI consists of two subtypes: inflammatory cerebral amyloid angiopathy and amyloid β (Aβ)-related angiitis. Acute or subacute onset of cognitive decline or behavioral changes is the most common symptom of CAA-RI. Rapid progressive dementia, headache, seizures, or focal neurological deficits, with patchy or confluent hyperintensity on T2 or fluid-attenuated inversion recovery sequences and evidence of strictly lobar microbleeds or cortical superficial siderosis on susceptibility-weighted imaging imply CAA-RI. The gold standard for diagnosis is autopsy or brain biopsy. However, biopsy is invasive;consequently, most clinically diagnosed cases have been based on clinical and radiological data. Other diagnostic indexes include the apolipoprotein E ε4 allele, Aβ and anti-Aβ antibodies in cerebral spinal fluid and amyloid positron emission tomography. Many diseases with similar clinical manifestations should be carefully ruled out. Immunosuppressive therapy is effective both during initial presentation and in relapses. The use of glucocorticoids and immunosuppressants improves prognosis. This article reviews the pathology and pathogenesis, clinical and imaging manifestations, diagnostic criteria, treatment, and prognosis of CAA-RI, and highlights unsolved problems in the existing research.

Temporal stability and directional change in a color cline of a marine snail from NW Spain

The evolution and maintenance of color clines is a classic topic of research in evolutionary ecology.However,studies analyzing the temporal dynamics of such clines are much less frequent,due to the difficulty of obtaining reliable data about past color distributions along environmen-tal gradients.In this article,we describe a case of decades-long temporal stability and directional change in a color cline of the marine snail Littorina saxatilis along the coastal inlet of the Ria de Vigo(NW Spain).L.saxatilis from this area shows a clear color cline with 3 distinct areas from the innermost to the more wave-exposed localities of the Ria:the inner,protected localities show an abundance of fawn-like individuals;the intermediate localities show a high diversity of colors;and the outer,wave-exposed localities show populations with a high frequency of a black and lineated morph.We compare data from the 1970s and 2022 in the same localities,showing that the cline has kept relatively stable for at least over half a century,except for some directional change and local variability in the frequency of certain morphs.Multiple regression analyses and biodiversity measures are presented to provide clues into the selective pressures that might be involved in the maintenance of this color cline.Future research avenues to properly test the explanatory power of these selective agents as well as the possible origins of the cline are discussed.
