IBD5 polymorphisms in inflammatory bowel disease: Association with response to infliximab

AIM: Inflammatory bowel diseases (IBD) are multifactorial pathologies of unknown etiology. One susceptibility locus, IBD5, has been mapped to chromosome 5q31. We analyzed our Spanish cohorts of Crohn's disease (CD) and ulcerative colitis (DC) patients to determine whether this locus is associated with IBD, and to ascertain the main clinical phenotype influenced by this risk factor. The kind of interaction, either genetic heterogeneity or epistasis, between this IBD5 susceptibility region and the NOD2/CARD15 gene mutations was studied as well. Finally, we assessed whether this locus can predict response to infliximab therapy. METHODS: A case control study was performed with 274 CD and 211 UC patients recruited from a single center and 511 healthy ethnically matched controls. Two polymorphisms were genotyped in the IBD5 locus and three in the CARD15/NOD2 gene. RESULTS: Our results evidence association only with CD especially with the fistulizing phenotype and in the absence of NOD2/CARD15 variants (mutant allele frequency in patients vs controls: OR = 2.03, 95% CI = 1.35-3.06, P<0.01). The frequency of the IBD5 homozygous mutant genotype significantly increased in CD patients lacking response to infliximab (RR = 3.88, 95% CI = 1.18-12.0, P<0.05). UC patients overall do not show association with 5q31 polymorphisms, although a similar trend to the one observed in CD is found within the worse prognosis group. CONCLUSION: The IBD5 variants may enhance an individual carrier's risk for CD, mainly in the absence of the NOD2/CARD15 mutations and in fistulizing patients. The data presented suggest the potential role of the 5q31 polymorphisms as markers of response to infliximab.

Hydraulic models for the generation of flood inundation maps are not commonly applied in mountain river basins because of the difficulty in modeling the hydraulic behavior and the complex topography. This paper presents a comparative analysis of the performance of four twodimensional hydraulic models (HEC-RAS 2D, Iber 2D, Flood Modeller 2D, and PCSWMM 2D) with respect to the generation of flood inundation maps. The study area covers a 5-km reach of the Santa B-arbara River located in the Ecuadorian Andes, at 2330 masl, in Gualaceo. The model's performance was evaluated based on the water surface elevation and flood extent, in terms of the mean absolute difference and measure of fit. The analysis revealed that, for a given case, Iber 2D has the best performance in simulating the water level and inundation for flood events with 20- and 50-year return periods, respectively, followed by Flood Modeller 2D, HEC-RAS 2D, and PCSWMM 2D in terms of their performance. Grid resolution, the way in which hydraulic structures are mimicked, the model code, and the default value of the parameters are considered the main sources of prediction uncertainty.

Arboviral diseases are viral infections transmitted to humans through the bites of arthropods,such as mosquitoes,often causing a variety of pathologies associated with high levels of morbidity and *** the past decades,these infections have proven to be a significant challenge to health systems worldwide,particularly following the considerable geographic expansion of the dengue virus(DENV)and its most recent outbreak in Latin America as well as the difficult-tocontrol outbreaks of yellow fever virus(YFV),chikungunya virus(CHIKV),and Zika virus(ZIKV),leaving behind a substantial portion of the population with complications related to these ***,the world is experiencing a period of intense globalization,which,combined with global warming,directly contributes to wider dissemination of arbovirus vectors across the ***,all continents remain on high alert for potential new ***,this review aims to provide a comprehensive understanding of the pathogenesis of the four main arboviruses today(DENV,ZIKV,YFV,and CHIKV)discussing their viral characteristics,immune responses,and mechanisms of viral evasion,as well as important clinical aspects for patient *** includes associated symptoms,laboratory tests,treatments,existing or developing vaccines and the main associated complications,thus integrating a broad historical,scientific and clinical approach.

This paper develops further the theory of symmetrization of fractional Laplacian operators contained in recent works of two of the authors. This theory leads to optimal estimates in the form of concentration comparison inequalities for both elliptic and parabolic equations. The authors extend the theory for the so-called restricted fractional Laplacian defined on a bounded domain Ω of R^N with zero Dirichlet conditions outside of Ω. As an application, an original proof of the corresponding fractional Faber-Krahn inequality is derived. A more classical variational proof of the inequality is also provided.

Experimental analysis of infragravity waves in two eroded microtidal beaches

This work aims to contribute to the characterization and understanding ofinfragravity waves on two beaches with erosion problems. For this reason, we have used an array of ADCP and a pressure sensor to measure wave parameters and pressure inside and outside of the surf zone during the dry and rainy period in the beaches of Galerazamba and Manzanillo del Mar （both dissipative and eroded beaches） located in the Colombian Caribbean coast. Based on these measurements, we have carried out a spectral analysis in order to identify the frequency components that characterize the wave and its energy; thus, we identified the characteristic frequencies of iufragravity waves to finally filter the infragravity signal on each beach in different seasonal periods. Among the results of the Welch spectrum applied to surface elevation time series, we found that, the frequencies＇ energy of the sea-swell band decreases due to bottom friction and wave breaking as the wave approaches the shore, while the frequencies＇ energy of the infragravity band increases significantly. In addition, for the wavelet analysis, we could observe how the energy of the infragravity band, especially the lowest frequencies gain energy as the waves approaches the coast. Furthermore, based on the infragravity wave obtained from the extreme wave event registered during the field campaign we can conclude that the contribution of this signal is important in the erosion problems presented in the beaches of Galerazamba and Manzanillo del Mar. Finally, these results show the need to realize other studies that allow us to understand deeply, the role of infragravity waves on the morphological changes that occurs in these beaches.

AIM: To explore methylation of DAPK, THBS1, CDH-1, and p14 genes, and Helicobacter pylori(H. pylori) status in individuals harboring esophageal columnar ***: Distal esophageal mucosal samples obtained by endoscopy and histologically diagnosed as gastric-type(non-specialized) columnar metaplasia, were studied thoroughly. DNA was extracted from paraffin blocks, and methylation status of deathassociated protein kinase(DAPK), thrombospondin-1(THBS1), cadherin-1(CDH1), and p14 genes, was examined using a methyl-sensitive polymerase chain reaction(MS-PCR) and sodium bisulfite modification protocol. H. pylori cag A status was determined by ***: In total, 68 subjects(33 females and 35 males), with a mean age of 52 years, were included. H. pylori cag A positive was present in the esophageal gastric-type metaplastic mucosa of 18 individuals. DAPK, THSB1, CDH1, and p14 gene promoters were methylated by MS-PCR in 40(58.8%), 33(48.5%), 46(67.6%), and 23(33.8%) cases of the 68 esophageal samples. H. pyloristatus was associated with methylation of DAPK(P = 0.003) and THBS1(P = 0.019).CONCLUSION: DNA methylation occurs in cases of gastric-type(non-specialized) columnar metaplasia of the esophagus, and this modification is associated with H. pylori cag A positive infection.

Landslides in intertropical mountainous areas of Mexico are a natural hazard that could potentially generate a disaster. Despite this, many areas lack landslide inventories, making it difficult to monitor and efficiently manage the associated risks for local populations. For this reason, the aim of this research was to undertake a multitemporal landslide inventory and analyze its conditioning factors in the Pico de Tancítaro stratovolcano, which, in recent decades, has presented several high-risk geomorphological events with disastrous consequences for nearby localities. The spatial distribution of landslides and its conditioning factors were mapped at a 1:50,000 scale for the 1995-2020 period, through visual interpretation of orthophotos and satellite images(Landsat, SPOT, and SENTINEL). The conditioning factors that link the most to landslides were Quaternary andesite and basaltic andesite rocks;volcanic cones, mountain river valleys, and very steep to steep slopes(30° to >45°), oriented to the South, North, and East;Andosols and Leptosols;and the oyamel fir and mixed forests. 505 landslide events classified into slides and flows that occurred in 1995, 2004, 2010, and 2015 were identified. New scars generated by the reactivation of landslide processes were also observed. The landslides in the study area were triggered by torrential rains caused by hurricanes and tropical storms;such as hurricanes Alex and Patricia. The inventory and analysis of conditioning factors highlighted the distribution and dynamics of slides and flows in the study area. In addition, the most important factors for studies of landslide susceptibility, hazards, and risks in the Pico de Tancítaro stratovolcano were identified, contributing to the management of geomorphological risks in intertropical mountain areas.
