Brassinosteroids （BRs） are essential hormones for growth and development of plant. In rice, BRs regulate multiple developmental processes and affect many important traits such as height, leaf angle, fertility and seed filling. We identified brassinosteroid-regulated proteins in rice using proteomic approaches and performed functional analysis of some BR-regulated proteins by overexpression experiments. Using two-dimensional difference gel electrophoresis （2-D DIGE） followed by protein identification by mass spectrometry, we compared proteomic differences in the shoots and roots of the BR-insensitive mutant d61-4 and BR-deficient mutant brd1-3. We identified a large number of proteins differentially expressed in the mutants compared with wild type control. These include a glycine-rich RNA-binding protein （OsGRP1） and a DREPP2 protein, which showed reduced levels in the BR mutants. Overexpression of these two proteins partially suppressed the dwarf phenotype of the Arabidopsis BR-insensitive mutant bri1-5. In contrast to the reduced protein level, the RNA level of OsGRP1 was not significantly affected in the BR mutants or by BR treatment, suggesting BR regulation of OsGRP1 at the posttranslational level. This study identifies many BR-regulated proteins and demonstrates that OsGRP1 functions downstream in the BR signal transduction pathway to promote cell expansion.

Changes of Reactive Oxygen Species and Related Enzymes in Mitochondrial Respiration During Storage of Harvested Peach Fruits

Peach fruits [Prumus persica （l.） Batsch, cv. Yuhuasanhao] were used as materials to investigate the changes of reactive oxygen species （ROS） and related enzymes in mitochondria respiration during storage and then their influence on senescence of harvested Peach fruits was studied. The results showed that low temperature （5℃） strongly inhibited the reduction of firmness and the increase in respiration rate. During storage at ambient temperature （20℃）, ROS had a cumulative process while malondialdehye （MDA） content continued to increase in associated with enhanced membrane lipid peroxidation. lipoxygenase （lOX） activity was strongly inhibited under the low temperature condition. The activities of succinic dehydrogenase （SDH）, cytochrome C oxidase （CCO）, and Ca^2＋-ATPase declined to a certain extent at ambient temperature, while they showed higher activities at low temperature, which may be related to lower membrane lipid peroxidation at low temperature. Higher Ca^2＋ content at ambient temperature may be responsible for impairment of mitochondrial function, thus, leading to fruit senescence. The results showed that under low temperature condition, the low accumulation of ROS and the low level of membrane lipid peroxidation could maintain the function of mitochondria that would help to delay the senescence of peach fruits. These suggested a close relationship existed between ROS metabolism and mitochondrial respiration. It can be inferred that the low temperature helps to delay senescence of peach fruits via suppression of ROS and related enzymes, maintain better homeostasis of Ca^2＋ in mitochondria and thus better mitochondrial functions.

Nothofagus pumilio forests in Tierra del Fuego are the southernmost forests in the world,where extreme climate conditions represent a challenge to attain sustainable forest *** forestry was proposed as an alternative to increase the species conservation in managed ***,we synthetized results related to the implementation of a variable retention harvesting based on a combination of aggregate patches and dispersed retention during the last 18 years comparing with other silviculture proposals(e.g.,shelterwood cuts)and control treatments(primary unmanaged forests).We summarized the results for(i)sawmill operations,(ii)timber yield,(iii)overstory stability,(iv)forest structure,(v)microclimate and natural cycles,(vi)natural regeneration dynamics(flowering,seeding,foraging,recruitment,growth,and mortality),and(vii)biodiversity(mammals,understory plants,mistletoes,birds,arthropods,mosses,lichens,and fungi).In general,aggregate patches maintained forest structure and micro-environmental variables,and slightly increased biodiversity and forest reproduction variables compared to unmanaged primary *** the contrary,dispersed retention decreased forest structure variables and greatly increased biodiversity(richness and abundance)when it was compared to unmanaged primary *** conditions are influenced by variable retention harvesting,but direction and magnitude of the effect depend and differ according to retention *** this,biodiversity taxa greatly differed among groups depending on retention *** general,the species assemblages in aggregate patches were similar to those found in primary unmanaged forests,while they were significantly modified in the dispersed *** occurred due to(i)local extinction of some original species,(ii)the introduction of native species from the surrounding environments,or(iii)the invasion of exotic *** silvicultural method has been a useful tool to conserve biodiversity and ec

Synthesis of polymer protected Au NPs for silver ions detection

Herein, a novel probe based on poly(N,N′-methylenebisacylamide) protected Au NPs(PDMAM-Au NPs) was developed for determination of silver ions. The thiol-terminated PDMAM was synthesized by the reversible addition fragmentation chain transfer radical polymerization. Then, the PDMAM-Au NPs were prepared by a simple, one-pot and green process. It has been observed that existence of the silver ions evoked a small red-shift to the emission peak of PDMAM-Au NPs and an obvious increase in UV absorption. The detection limit of the present method for silver ions was 0.5 ?mol/l, with a linear range of 1.0–1000 ?mol/l. Moreover, the possible mechanism for enhanced UV absorption intensity with enlarged diameter of PDMAM-Au NPs was explored and discussed briefly. The results demonstrated that silver ions could be detected by the PDMAM-Au NPs based colorimetric assays, further opening up new ways for the visualization and quantification of silver ions involved in our daily life.

Reverse-transformation austenite structure control with micro/nanometer size

To control the reverse-transformation austenite structure through manipulation of the micro/nanometer grain structure, the influences of cold deformation and annealing parameters on the microstructure evolution and mechanical properties of 316l austenitic stainless steel were investigated. The samples were first cold-rolled, and then samples deformed to different extents were annealed at different temperatures. The microstructure evolutions were analyzed by optical microscopy, scanning electron microscopy (SEM), magnetic measurements, and X-ray diffraction (XRD); the mechanical properties are also determined by tensile tests. The results showed that the fraction of stain-induced martensite was approximately 72% in the 90% cold-rolled steel. The micro/nanometric microstructure was obtained after reversion annealing at 820-870A degrees C for 60 s. Nearly 100% reversed austenite was obtained in samples annealed at 850A degrees C, where grains with a diameter ae 500 nm accounted for 30% and those with a diameter > 0.5 mu m accounted for 70%. The micro/nanometer-grain steel exhibited not only a high strength level (approximately 959 MPa) but also a desirable elongation of approximately 45%.

The global avalanche characteristics （the sum- of-squares indicator and the absolute indicator） measure the overall avalanche characteristics of a cryptographic Boolean function. Sung et al. （1999） gave the lower bound on the sum- of-squares indicator for a balanced Boolean function satisfy- ing the propagation criterion with respect to some vectors. In this paper, if balanced Boolean functions satisfy the propaga- tion criterion with respect to some vectors, we give three nec- essary and sufficient conditions on the auto-correlation distri- bution of these functions reaching the minimum the bound on the sum-of-squares indicator. And we also find all Boolean functions with 3-variable, 4-variable, and 5-variable reaching the minimum the bound on the sum-of-squares indicator.

Effects of triclosan on gonadal differentiation and development in the frog Pelophylax nigromaculatus

Previous studies have reported that triclosan（TCS） could possess an androgenic *** aimed to investigate the effects of TCS on gonadal differentiation and development in the frog Pelophylax nigromaculatus, a sensitive amphibian species to androgenic chemicals.P. nigromaculatus tadpoles at stage 24 were exposed to TCS（3, 30, and 300 nmol/l） to stage46 in a semi-static exposure system. At the end of exposure, gonadal morphology and histology, sex ratio and gonadal expression of sex-biased genes were examined in P. nigromaculatus. In each TCS treatment group, we found several individuals whose gonads exhibited morphological and/or histological abnormalities. Gonadal histological abnormalities were characterized by few oocytes and many somatic cells. Although the percentage of the individuals with abnormal gonads was low（7.8%） among all animals treated with TCS,statistical test revealed the sex ratios in the 3 and 300 nmol/l TCS treatment groups were significantly different from the solvent control. In the 30 nmol/l TCS treatment group,abnormal gonads were also observed, although the sex ratio was not changed compared with the solvent control, which was possibly due to the smaller sample size in this group. In all the TCS treatment groups, the sex ratios were not obviously male-biased, but the expression levels of some sex-biased genes were significantly altered by TCS. Altogether,our results suggest that TCS, even at environmentally relevant concentrations, could disrupt gonadal differentiation and development in P. nigromaculatus, but we are not sure whether the disrupting effects were associated with masculinization or feminization.

Asperuloside Promotes Apoptosis of Cervical Cancer Cells through Endoplasmic Reticulum Stress-Mitochondrial Pathway

Objective: To investigate the effects of asperuloside on cervical cancer based on endoplasmic reticulum(ER) stress and mitochondrial pathway. Methods: Different doses(12.5–800 μg/ml) of asperuloside were used to treat cervical cancer cell lines Hela and CaSki to calculate the half maximal inhibitory concentration(IC_(50)) of asperuloside. The cell proliferation was analyzed by clone formation assay. Cell apoptosis, intracellular reactive oxygen species(ROS) and mitochondrial membrane potential were determined by flow cytometry. The protein expressions of cleaved-caspase-3, Bcl-2, Bax, Cyt-c, cleaved-caspase-4 and glucose-regulated protein 78(GRP78) were analyzed by Western blot. And the inhibitor of ER stress, 4-phenyl butyric acid(4-PBA) was used to treat cervical cancer cells to further verify the role of ER stress in the apoptosis of cervical cancer cells induced by asperuloside. Results: Asperuloside of 325, 650, and 1300 μg/ml significantly inhibited the proliferation and promoted apoptosis of Hela and CaSki cells(Pll doses of asperuloside significantly increased intracellular ROS levels, reduced mitochondrial membrane potential, significantly reduced Bcl-2 protein expression level, and increased Bax, Cyt-c, GRP78 and cleaved-caspase-4 expressions(Pl/l 4-PBA treatment significantly promoted cell proliferation and reduced apoptosis(Pl asperuloside could reverse 4-PBA-induced increased cell proliferation, decreased apoptosis and cleaved-caspase-3,-4 and GRP78 protein expressions(Plusion: Our study revealed the role of asperuloside in cervical cancer, suggesting that asperuloside promotes apoptosis of cervical cancer cells through ER stress-mitochondrial pathway.
