Reviewing Self-Injuries Behavior in Macaques: The Role of Early Experience and Genetics in an Integrated Non-Human Model

Genetic differences in non-human primates and the context where they were reared are important in the determination of their behaviors. Some studies suggest that Self-Injurious Behavior results from stress or anxiety, but some macaques appear to be more vulnerable to acquiring Self-Injurious Behavior. Vulnerability and risk for socially reared macaques of developing Self-Injurious Behavior increased when they experienced early adverse events and suffered from consequent stress. Stressful events can cause alterations in the neuroendocrine and neuropeptide systems associated with the regulation of stress and anxiety. Dysregulation in these systems contributes to the occurrence of anxious episodes that lead to self-harm. This could be used by those animals to counteract the effects of anxiety, causing euphoria by releasing endogenous opioids. In developing an integrative approach of Self-Injurious Behavior, we propose the diathesis-stress model to illustrate how the environment and genetics are influenced as well, taking rhesus monkeys as an example. Rhesus macaques and humans have essentially the same gene 5-HTT as well as functional polymorphism. The allelic variation in the region promoting the serotonin transporter gene (5-HTT) was associated with significant differences in the number of depressive symptoms and contributions to social reward and punishment. Growing up without the mother and the rearing environment are important risk factors for developing abnormal behaviors. Presence of Infectious Hypodermal and Haematopoietic Necrosis Virus (IHHNV) in Native Shrimps from Southern Mexico

A survey for Infectious Hypodermal and Haematopoietic Necrosis Virus (IHHNV) and White Spot Syndrome Virus (WSSV) was performed during two catching seasons of native shrimps in the Yucatan Coast in the Gulf of Mexico. The mtDNA COI barcode identified two endemic species;the southern pink shrimp Penaeus notialis (Pérez Fantante, 1967) and the northern pink shrimp Penaeus brasiliensis (Latreille, 1987) (previously described as Farfantepenaeus brasiliensis). The prevalence of IHHNV was of 18.18% in 2016, and of 8.57% in 2017. All organisms tested negative for WSSV. This is the first identification of wild shrimps in the state of Yucatan Mexico by mtDNA COI barcode as well as the first identification of IHHNV in such species. The presence of IHHNV in wild shrimps populations has a potential of persisting in the coast of Yucatan with putative detrimental effect on local fisheries because once established in natural waters and hosts;such pathogens are almost impossible to eradicate.

Immersion Tools as a Strategy for Adolescents Mental Health: A Qualitative Study

Background: Information and communication technologies have revolutionized education, due to development of didactic strategies to take advantage of them for maximizing learning. The aim was to analyze the influence of immersion tools as a strategy to improve adolescents’ mental health. Methods: Qualitative research was employed, and participants’ experiences, opinions, values, behaviors, emotions, feelings were recorded. Twenty six students of both sexes, aged 13 to 15 years, from the three grades of secondary school were selected by convenience, and supported by the school’s psychologist. A workshop on emotional intelligence (EI) based on educational technology was implemented using a video;students practiced a reflexive process to understand themselves and their surroundings, creating a video + audio as a product of the workshop. In it, students analyzed several problems they face and their relationship with their contextual situation. Data collection was through semi-structured interviews before and after the workshop on EI. Data analysis was according to Souza Minayo, and the theoretical framework according to Goleman, Tapia et al., Gorostieta et al., and Vigotsky. Results: Two categories emerged: Category I, EI as a risk factors transformer from living with addictions to protection factors in adolescents. Category II, EI development through immersion tools to improve mental health in adolescents. Conclusions: Immersion tools favor user to be closer, with a humanist focus where real necessities are studied after the vivid experience of the problem to solve or get a better situation. The goal is that students experience real situations inside controlled surroundings, in such a way that they learn from their experiences and acquire knowledge through self-learning. Soil Carbon Storage in Experimental Forest Plantations with Tropical Species

Forest ecosystems represent a third of the total organic carbon (TOC) stored in the soil. However, studies still need to be carried out to generate reliable information on its soil carbon storage potential in an experimental plantation. Therefore, the objective of this research was to determine the storage of total organic carbon (TOC) in the soil in two experimental forest plantations with tropical species, located in the state of Nayarit, México. The research was carried out in two experimental plantations with tropical species, located at the Santiago Ixcuintla Experimental Field (CESIX) and at the El Verdine?o Experimental Site (SEEV). The established species are Swietenia macrophylla King (mahogany), Cedrela odorata L. (cedar) and Tabebuia rosea Bertoil (pink trumpet-tree). In addition, data from dasometric variables were used in order to define if any of these variables had a similar variation to the soil TOC stock by species. The sampling was done under a randomized block experimental design in each experimental forest plantation (CESIX and SEEV). Nine composite samples were collected in the soil under the tree species at a depth of 0 - 30 cm, and 1.5 m distance from the tree. To determine the soil TOC, an elemental analyzer with a thermal conductivity detector Thermo Scientific (Flash 2000) was used. The results showed that TOC presented mean values of 103.60 and 219.98 t·ha-1 at a depth of 0 - 30 cm in CESIX and SEEV respectively. The highest TOC was obtained in soil under mahogany;142.95 t·ha-1 in CESIX and 284.41 t·ha-1 in SEEV. The analysis of the differences in TOC between the two plantations was highly significant. 背景与目的林奇综合征(lynch syndrome,LS)是一种遗传性疾病,LS患者罹患结直肠癌、子宫内膜癌和其他与DNA错配修复基因发生种系突变相关肿瘤的风险较高。LS的传统遗传诊断方法为对与疑似综合征相关的基因进行桑格测序。二代测序技术(next-generation sequencing,NGS)可同时对大量遗传性癌基因进行测序。本研究旨在探究林奇综合征患者中是否存在其他遗传性种系病理癌基因变异。方法我们选取先前已利用桑格测序遗传诊断法确诊的84例LS先证者作为研究对象,通过杂交捕获一组94个商业化的遗传癌基因,随后对这些捕获的样本进行NGS测序。按照美国医学遗传学和基因组学标准,对这些变异的临床意义进行分类。利用桑格测序验证NGS的结果。在可能的情况下,利用桑格测序进一步在先证者的亲属中进行新发现种系变异基因的遗传分析。结果我们从84个家族中鉴定出5(6%)个至少含有2个种系致病变异的家族,这些变异具有高或中等的诱发不同显性癌症的风险,这些家族的种系致病变异如下:[MLH1-BRCA2-NBN]、[MLH1-BRCA1]、[MSH2-ATM]、[MSH6-NF1]和[MLH1-FANCA]。有趣的是,这5个家族中只有1个家族表现出与新的致病变异相关的临床表型。含有3个致病变异基因[MLH1-BRCA2-NBN]的家族显示出LS、乳腺和卵巢癌综合征相关肿瘤的高度聚集特性。结论我们的研究表明,癌症易感基因中,同时发生多个致病变异是LS病例的显著特点。在大部分的病例中,继发性致病变异与临床表型不相关。将来需要进一步验证这些结论并阐明其临床影响。只有表现出复杂的临床表型家族才有必要重新对LS进行分析。

Novel Nitrobenzazolo[3,2-a]quinolinium Salts Induce Cell Death through a Mechanism Involving DNA Damage, Cell Cycle Changes, and Mitochondrial Permeabilization

This study reports the capacity of three nitro substituted benzazolo[3,2-a]quinolinium salts NBQs: NBQ 95 (NSC-763304), NBQ 38 (NSC 763305), and NBQ 97 (NSC-763306) as potential antitumor agents. NBQ’s are unnatural alkaloids possessing a positive charge that could facilitate interaction with cell organelles. The anticancer activities of these compounds were evaluated through the National Cancer Institute (NCI) 60 cell line screening which represents diverse histologies. The screening was performed at 10 μM on all cell lines. Results from the NCI screening indicated cytotoxicity activity on six cell lines. In order to explore a possible mechanism of action, a detailed biological activity study of NBQ 95 and NBQ 38 was performed on A431 human epidermoid carcinoma cells to determine an apoptotic pathway involving, cell cycle changes, DNA fragmentation, mutations, mitochondrial membrane permeabilization and caspases activation. DNA fragmentation, cell cycle effects, mutagenesis, mitochondrial permeabilization and activation of caspases were determined by fluorimetry and differential imaging. Our data showed that A431 growth was inhibited with an average IC50 of 30 mM. In terms of the mechanism, these compounds interacted with DNA causing fragmentation and cell cycle arrest at sub G0/G1 stage. Mutagenesis was higher for NBQ 38 and moderate for NBQ 95 Mitochondrial permeabilization was observed with NBQ 38 and slightly for NBQ 95. Both compounds caused activation of Caspases 3 and 7 suggesting an apoptotic cell death pathway through an intrinsic mechanism. This study reports evidence of the toxicity of these novel compounds with overlapping structural and mechanistic similarities to ellipticine, a known anti-tumor compound.

MicroRNAs: New players in endometriosis

Endometriosis is an estrogen-dependent inflammatory disorder that limits the quality of life of affected women. This pathology affects 10% of reproductive-age women, although the prevalence in those patients experiencing pain, infertility or both is as high as 35%-50%. Endometriosis is characterized by endometrial-like tissue outside the uterus, primarily on the pelvic peritoneum, ovaries and the pouch of Douglas. Despite extensive research endeavours, a unifying theory regarding the exact etiopathogenic mechanism of this high prevalent and incapacitating condition is still lacking, although it has been suggested that epigenetics could be involved. Micro RNAs(mi RNAs), one of the epigenetic players, are small non-coding RNAs that can act as post-transcriptional regulators of gene expression, reducing the expression of their target m RNAs either inhibiting its translation or promoting its degradation. Mi RNA expression profiles are specific of tissue and cell type. Abnormal mi RNA expression has been described in different pathological conditions, such as a myriad of oncological, cardiovascular and inflammatory diseases and gynecological pathologies. In endometriosis, mi RNA expression patterns of eutopic endometrium from patients and control women and from different endometriotic lesions have been described. These small non-coding molecules have become attractive candidates as novel biomarkers for an early non-invasive diagnosis of the disease, which could suppose a valuable benefit to the patients in terms of improvement of prognosis and reduction of the ratio of recurrence. In this systematic review we will focus on the role of mi RNAs in the pathophisiology of endometriosis.
