Tumor microenvironment-activatable neuropeptide-drug conjugates enhanced tumor penetration and inhibition via multiple delivery pathways and calcium deposition

Peptide-drug conjugates have achieved considerable development and application as a novel strategy for targeted delivery of anticancer drugs. Bioactive peptides induced calcium deposition can irreversibly assist inhibition of tumors. However, active regulation of calcium level through signal transduction of bioactive substances has not been reported yet. In this study, novel neuropeptide-doxorubicin conjugates(NP-DOX) with lysosome-specific acid response were described for neuropeptide Y_1 receptor(Y_1R)-overexpressed triple-negative breast cancer. The delivery mechanism of NP-DOX was clarified that diverse pathways were involved, including intracellular and intercellular transport. Importantly, up-regulation of Y_1 R-mediated intracellular calcium level via second messenger inositol triphosphate was presented in NP-DOX treated MDA-MB-231 cells. In vivo antitumor efficacy demonstrated that NP-DOX showed less organ toxicity and enhanced tumor inhibition benefited from its controlled release and Y_1R-mediated calcium deposition, compared with free DOX. This bioconjugate is a proof-of-concept confirming that neuropeptide-mediated control of signaling responses in neuropeptide-drug conjugates enables great potential for further applications in tumor chemotherapy.

Anti-abrasion collagen fiber-based membrane functionalized by UiO-66-NH_(2)with ultra-high efficiency and stability for oil-in-water emulsions separation

Membrane separation strategies offer promising platform for the emulsion ***,the low mechanical strength of membrane separation layers and the trade-off between separation flux and efficiency present significant *** this study,we report a CfM@UiO-66-NH_(2)membrane with high separation flux,efficiency and stability,through utilizing a robust anti-abrasion collagen fiber membrane(CfM)as the multifunctional support and UiO-66-NH_(2)by an in-situ growth as the separation *** high mechanical strength of the CfM compensated for the weakness of the separation layer,while the charge-breaking effect of UiO-66-NH_(2),along with the size sieving of its constituent separating layers and the capillary effect of the collagen fibers,contributed to the potential for efficient ***,the CfM@UiO-66-NH_(2)membrane exhibited superhydrophilic properties,making it suitable for separating oil-in-water microemulsions and nanoemulsions stabilized by anionic *** membrane demonstrated remarkable separation efficiencies of up to 99.960%and a separation flux of370.05 L·m^(-2)·h^(-1).Moreover,it exhibits stability,durability,and abrasion resistance,maintaining excellent separation performance even when exposed to strong acids and alkalis without any damage to its structure and *** six cycles of reuse,it achieved a separation flux of 417.97 L·m^(-2)·h^(-1)and a separation efficiency of 99.747%.furthermore,after undergoing 500 cycles of strong abrasion,the separation flux remained at 124.39 L·m^(-2)·h^(-1),with a separation efficiency of 99.992%.These properties make it suitable for the long-term use in harsh operating *** attribute these properties to the electrostatic effect resulting from the amino group on UiO-66-NH_(2)and its in-situ growth on the CfM,which forms a size-screening separation *** work highlights the potential of the CfM@UiO-66-NH_(2)membrane as an environmentally friendly size-screening

Clinical characteristics of acute non-varicose upper gastrointestinal bleeding and the effect of endoscopic hemostasis

BACKGROUND Acute non-variceal upper gastrointestinal bleeding(ANVUGIB)constitutes a prevalent emergency within Gastroenterology,encompassing 80%-90%of all gastrointestinal hemorrhage *** condition is distinguished by its abrupt onset,swift progression,and notably elevated mortality *** To gather clinical data from patients with ANVUGIB at our hospital in order to elucidate the clinical characteristics specific to our institution and analyze the therapeutic effectiveness of endoscopic *** We retrospectively retrieved the records of 532 patients diagnosed with ANVUGIB by endoscopy at our hospital between March 2021 and March 2023,utilizing our medical record *** pertaining to general patient information,etiological factors,disease outcomes,and other relevant variables were meticulously collected and *** Among the 532 patients diagnosed with ANVUGIB,the male-to-female ratio was 2.91:1,with a higher prevalence among ***,43.6%of patients presented with black stool as their primary complaint,while 27.4%had hematemesis as their initial *** admission,17%of patients exhibited both hematemesis and black stool,while most ANVUGIB patients primarily complained of overt gastrointestinal *** routine blood examinations at admission revealed that 75.8%of patients had anemia,with 63.4%experiencing moderate to severe anemia,and 1.5%having extremely severe anemia(hemoglobinf patients experienced bleeding without a definitive trigger,24.2%had a history of using gastric mucosa-irritating medications,24.2%developed bleeding after alcohol consumption,2.8%attributed it to improper diet,1.7%to emotional excitement,and 2.3%to fatigue preceding the bleeding ***-induced ANVUGIB was more prevalent in the elderly than middle-aged and young individuals,while bleeding due to alcohol consumption showed the opposite ***,diet-related bleeding was more common

Effects of processing parameters on fabrication defects,microstructure and mechanical properties of additive manufactured Mg–Nd–Zn–Zr alloy by selective laser melting process

Mg–3Nd–0.2Zn–0.4Zr(NZ30K,wt.%)alloy is a new kind of high-performance metallic *** combination of the NZ30K Magnesium(Mg)alloy and selective laser melting(SLM)process seems to be an ideal solution to produce porous Mg degradable ***,the microstructure evolution and mechanical properties of the SLMed NZ30K Mg alloy were not yet studied ***,the fabrication defects,microstructure,and mechanical properties of the SLMed NZ30K alloy under different processing parameters were *** results show that there are two types of fabrication defects in the SLMed NZ30K alloy,gas pores and unfused *** the increase of the laser energy density,the porosity sharply decreases to the minimum first and then slightly *** minimum porosity is 0.49±0.18%.While the microstructure varies from the large grains with lamellar structure inside under low laser energy density,to the large grains with lamellar structure inside&the equiaxed grains&the columnar grains under middle laser energy density,and further to the fine equiaxed grains&the columnar grains under high laser energy *** lamellar structure in the large grain is a newly observed microstructure for the NZ30K Mg *** laser energy density leads to finer grains,which enhance all the yield strength(YS),ultimate tensile strength(UTS)and elongation,and the best comprehensive mechanical properties obtained are YS of 266±2.1 MPa,UTS of 296±5.2 MPa,with an elongation of 4.9±0.68%.The SLMed NZ30K Mg alloy with a bimodal-grained structure consisting of fine equiaxed grains and coarser columnar grains has better elongation and a yield drop phenomenon.

Gender Pay Gap in the Gig Economy

The rapid ascent of digital platforms and the gig economy has prompted concerns about the gender pay *** results show that in the gig economy,gender continues to be a crucial determinant of workers'earnings,with women earning 85%of what men earn on a monthly ***,in comparison to the traditional waged employment during the same period,the gender pay gap in the gig economy has *** some gig jobs(e.g.,ride-hailing services,delivery services,online education)exhibit certain occupational segregation,women in gig economy work are no longer concentrated in low-paying roles,thereby challenging the occupational crowding hypothesis prevalent in traditional *** the gig economy,the vast majority of the gender pay gaps arise from factors within occupations,and occupational segregation only has a very limited impact on the earnings ***,the gender pay gap among platform gig workers can be mostly explained by observable factors,which implies that compared to traditional employment,the gig economy exhibits a lower level of implicit gender discrimination in ***,we investigate new factors that determine the gender pay gap in the gig *** exhibit a stronger aversion to algorithmic control,a preference for job flexibility,and a tendency toward more isolated and less socially interactive work environmentsall of which serve to widen the gender pay gap that might have otherwise *** results of this research suggest that despite the criticisms surrounding the gig economy,it continues to exert a positive influence on China's labor market。

Efficient and selective upcycling of waste polylactic acid into acetate using nickel selenide

The conversion of waste polylactic acid（PLA） plastics into high-value-added chemicals through electrochemical methods is a promising and sustainable ***,developing efficient and highly selective catalysts for lactic acid oxidation reaction（LAOR） and understanding the reaction process are ***,we report the electrooxidation of waste PLA to acetate at a high current density of 100 mA cm-2 with high faraday efficiency（～95%） and excellent stability（>100 h） over a nickel selenide nanosheet *** addition,a total faraday efficiency of up to 190% was achieved for carboxylic acids,including acetic acid and formic acid,by coupling with the cathodic CO2reduction *** situ experimental results and theoretical simulations revealed that the catalytic activity center of LAOR was dynamically formed NiOOH species,and the surface-adsorbed SeOxspecies accelerated the formation of Ni3+species,thus promoting catalytic *** mechanism of lactic acid electrooxidation was further *** acid was dehydrogenated to produce pyruvate first and then formed CH3CO due to preferential C-C bond cleavage,resulting in the presence of *** work demonstrated a sustainable method for recycling waste PLA and CO2into high-value-added products.

Enhancing mechanisms of N-doped biomass carbon on the vanadium-based catalyst for furan degradation at low temperature

Polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans(PCDD/fs)have attracted widespread concern due to their high toxicity,and their difficult manipulation in laboratories has made the research process ***,in our work,furan is selected as the model compound owing to the same structure of a central oxygenate *** catalytic oxidation is regarded as an effective and applicable method for the abatement of PCDD/fs,the synthesis of low-temperature catalysts is still a challenging problem in practical *** this situation,we prepared a novel V_(2)O_(5)/TiO_(2)catalyst modified with N-doped hierarchical porous carbon(NHPC)via a wet impregnation *** V/T-1%NHPC catalyst could achieve expectant low-temperature performances with 50%furan conversion at 150℃and a complete conversion at 200℃,which decreased 23℃and 40℃compared to the V/T catalyst ***,the addition of NHPC presented lifting chemical stability during long-time *** addition of NHPC in V/T catalysts decreased the formation of crystalline V_(2)O_(5) and increased the percentages of V^(5+)and O_(lat),which improved the utilization of vanadium ions and the catalytic ***,the higher binding energy shift of O_(lat) implied more reaction possibility with other oxidise ***,this work proved the lifting catalytic activity by the interaction between catalysts and NHPC,and proposed the promoting effects of the N *** results showed that the content of the pyridinic N and graphitic N in NHPC changed after combining with V/T catalyst,which played crucial roles in the excellent catalytic ***,this work provides comprehensive research of the V/T-1%NHPC catalyst toward furan oxidation at low temperature and explain the effects of N-doped biomass carbon in catalytic activity clearly,which gave a new thought to design low-temperature catalysts in PCDD/fs ***,the internal functional mech
