An increasing number of microRNAs （miRNAs） have been shown to play crucial regulatory roles in the process of plant development. Here, we used high-throughput sequencing combined with computational analysis to characterize miRNAomes from the ovules of wild-type upland cotton and a fiberless mutant during fiber initiation. Comparative miR- NAome analysis combined with northern blotting and RACE-PCR revealed seven fiber initiation-related miRNAs expressed in cotton ovules and experimentally validated targets of these miRNAs are involved in different cellular responses and metabolic processes, including transcriptional regulation, auxin and gibberellin signal transduction, actin bundles, and lignin biosynthesis. This paper describes a complex regulatory network consisting of these miRNAs expressed in cotton ovules to coordinate fiber initiation responses. In addition, 36 novel miRNAs and two conserved miRNAs were newly iden- tified, nearly doubling the number of known cotton miRNA families to a total of 78. Furthermore, a chromatin remodeling complex subunit and a pre-mRNA splicing factor are shown for the first time to be miRNA targets. To our knowledge, this study is the first systematic investigation of fiber initiation-related miRNAs and their targets in the developing cotton ovule, deepening our understanding of the important regulatory functions of miRNAs in cotton fiber initiation.

Postharvest broccoli is prone to yellowing during storage,which is the key factor leading to a reduction in *** explore appropriate control methods,it is important to understand the mechanisms of *** analyzed the genes related to the metabolism of chlorophyll,carotenoids,and flavonoids and the transcription factors(TFs)involved in broccoli yellowing using transcriptome sequencing *** stored at 10℃showed slight yellowing on postharvest day 5 and serious symptoms on day *** were significant changes in chlorophyll fluorescence kinetics,mainly manifesting as a decrease in the Fv/Fm value and an increase in nonphotochemical quenching,during the yellowing *** sequencing profiles from samples of fresh broccoli and broccoli with slight and severe yellowing revealed 6,5,and 4 differentially expressed genes involved in chlorophyll metabolism,carotenoid biosynthesis,and flavonoid biosynthesis,*** transcription factor gene ontology categories showed that the MYB,bHLH,and bZip gene families were involved in chlorophyll *** addition,the transcription factor families included NACs and ethylene response factors(ERFs)that regulated carotenoid *** transcription polymerase chain reaction further confirmed that bHLH66,PIF4,LOB13,NAC92,and APL were vital transcription factors that potentially regulated the CAO and HYD genes and were involved in chlorophyll metabolism and the carotenoid biosynthetic *** flavonoid biosynthetic pathway was mainly regulated by MYBs,NACs,WRKYs,MADSs,and *** results of the differentially expressed gene(DEG)and pigment content analyses indicated that the transcriptome data were accurately and positively associated with broccoli yellowing.

A target in layered medium can be located by the ridge-like distribution time reversal and reverse time migration(TR-RTM) mixed method. However, this method cannot distinguish between acoustic field distributions of the interface and target for the wider acoustic pulse signals, which may result in inaccurate location of the target. A snapshot TR-RTM mixed method is proposed to solve this problem. The principle of snapshot TR-RTM mixed method is first given. Experiments are then carried out, and a mountain-like acoustic field distribution is obtained by processing experimental data. The results show that the location of the peak is that of the target, and the ratio of the scattered signal and interface reflection signal(signal-to-interference ratio) is improved by about four times after processing. Furthermore, this method can effectively suppress the interface reflection signal and enhance the target scattering signal. Therefore, it can achieve effective detection and location of a target in a layered medium.

The“tailing”effect caused by residual non-aqueous phase liquids(NAPLs)in porous aquifers is one of the frontiers in pollution hydrogeology *** on the current knowledge that the residual NAPLs is mainly controlled by the pore structure of soil,this study established a method for evaluating the residual saturation of NAPLs by investigating the fractal dimension of porous *** this study,the soil column experiments of residual light NAPLs(LNAPLs)in sandy aquifer with different ratios of sands and soil were carried out,and the correlation between the fractal dimension of the medium,the residual of LNAPLs and the soil structure parameters are statistically analyzed,and its formation mechanism and main control factors are *** results show that:Under our experimental condition:(1)the fractal dimension of the medium has a positive correlation with the residual saturation of NAPLs generally,and the optimal fitting function can be described by a quadratic model:S_(R)=192.02 D2-890.73 D+1040.8;(2)the dominant formation mechanism is:Smaller pores in the medium is related to larger fractal dimension,which leads to higher residual saturation of NAPLs;stronger heterogeneity of the medium is related to larger fractal dimension,which also leads to higher residual saturation of NAPLs;(3)the micro capillary pores characterized by fine sand are the main controlling factors of the formation *** is concluded that both the theory and the method of using fractal dimension of the medium to evaluate the residual saturation of NAPLs are *** study provides a new perspective for the research of“tailing”effect of NAPLs in porous media aquifer.

Being the largest and most accessible organ of the human body,the skin could offer a window to diabetes-related complications on the ***,skin microvasculature is typically assessed by histological analysis,which is not suited for applications to large populations or longitudinal *** introduce ultra-wideband rasterscan optoacoustic mesoscopy(RSOM)for precise,non-invasive assessment of diabetes-related changes in the dermal microvasculature and skin micro-anatomy,resolved with unprecedented sensitivity and detail without the need for contrast *** unique imaging contrast,we explored a possible role for RSOM as an investigational tool in diabetes healthcare and offer the first comprehensive study investigating the relationship between different diabetes complications and microvascular features in *** applied RSOM to scan the pretibial area of 95 participants with diabetes mellitus and 48 age-matched volunteers without diabetes,grouped according to disease complications,and extracted six label-free optoacoustic biomarkers of human skin,including dermal microvasculature density and epidermal parameters,based on a novel image-processing *** then correlated these biomarkers to disease severity and found statistically significant effects on microvasculature parameters as a function of diabetes *** discuss how label-free RSOM biomarkers can lead to a quantitative assessment of the systemic effects of diabetes and its complications,complementing the qualitative assessment allowed by current clinical metrics,possibly leading to a precise scoring system that captures the gradual evolution of the disease.
