Hydroxylation and sulfation of sex steroid hormones in inflammatory liver

Sex steroids, also known as gonadal steroids, are oxidized with hydroxylation by cytochrome P450,glucuronidation by UDP-glucuronosyltransferase, sulfation by sulfotransferase, and O-methylation by catechol Omethyltransferase. Thus, it is important to determine the process by which inflammation influences metabolism of gonadal hormones. Therefore, we investigated the mechanism of metabolic enzymes against high physiologic inflammatory response in vivo to study their biochemical properties in liver diseases. In this study, C57BL/6N mice were induced with hepatic inflammation by diethylnitrosamine(DEN) exposure. We observed upregulation of Cyp19a1, Hsd17b1, Cyplal, Sult1e1 in the DEN-treated livers compared to the control-treated livers using real time PCR. Moreover, the increased Cyp19a1 and Hsd17b1 levels support the possibility that estrogen biosynthesis from androgens are accumulated during inflammatory liver diseases. Furthermore, the increased levels of Cyp1a1 and Cyp1b1 in the hydroxylation of estrogen facilitated the conversion of estrogen to 2-or 4-hydroxyestrogen,respectively. In addition, the substantial increase in the Sultlel enzyme levels could lead to sulfate conjugation of hydroxyestrogen. The present information supports the concept that inflammatory response can sequester sulfate conjugates from the endogenous steroid hormones and may suppress binding of sex steroid hormones to their receptors in the whole body.

The strain behavior of 0.71 Pb(Mg_(1/3)Nb_(2/3))O_(3)ePbTiO_(3)(PMN-PT)single crystal prepared by a solidstate single crystal growth method was investigated on two most widely used orientations,i.e.,(001)-and(011)-oriented.A special emphasis was put on the correlation among longitudinal and transverse strains and the consequent volume *** show that seemingly different strain behavior of(001)-and(011)-oriented crystals has the same origin,and the underlying mechanism excludes possible contribution from the frequently cited polarization *** situ monitoring of electric field induced Poisson’s ratio further suggested that the polarization vectors contributing to the strain properties in PMN-PT are not confined to a unique direction forced by the crystallographic symmetry but possess a span of angular range.

Monofilament type of polyaromatic amide(PA)and carbon nanotube(CNT)composite fibers is presented.A concept of a lyotropic liquid crystal(LLC)constructed via a spontaneous self-assembly is introduced to mitigate the extremely low com-patibility between PA and *** approaches provide an effective co-processing route of PA and CNT simultaneously to fabricate the uniform,continuous,and reliable composite fibers through a ***,the addition of a small amount PA into the dope solution of CNT governs the LLC mesophase not only in a spinneret stage but also in a coagulant ***,the developed PA/CNT composite fibers have the high uniaxial orientational order and the close interfacial packing compared to the pure CNT *** PA/CNT composite fibers achieve the outstanding tensile strength,electrical conductivity,and electrochemical response,while maintaining a *** also exhibit the chemical,mechanical,and thermal *** of these advantages can make flexible,sewable,and washable PA/CNT composite fibers ideal nanocomposite materials for use in next-generation information and energy transporting system by replacing conventional metal electrical conductors.

Changes of tear film lipid layer thickness by 3% diquafosol ophthalmic solutions in patients with dry eye syndrome

AIM: To evaluate the quantitatively changes in lipid layer thickness(LLT) when 3% diquafosol eye drop is used for dry eye patients using the tear film interferometer. METHODS: A total 124 participants(32 males, 92 females;mean age, 28.9 y) diagnosed with dry eye disease(DED) received topical instillation of 4 ophthalmic solutions in one eye: diquafosol, normal saline, 0.1% sodium hyaluronate and 0.3% gatifloxacin, in a masked manner. LLT was measured using an interferometer at baseline and 20 min after the instillation of each ophthalmic ***: Changes of LLT after instillation(nm, mean± standard error) were as follows: 12.6±2.0 for diquafosol(P<0.001), 1.2±2.2 for normal saline(P=0.301), 1.5±2.0 for hyaluronate(P=0.495), and 0.5±3.2 for gatifloxacin(P=0.884).CONCLUSION: Topical instillation of diquafosol increases tear film LLT in DED patients. Diquafosol 3% eye drop might be effective treatment option of evaporative DED with meibomian gland dysfunction.

随着中国经济的发展,儿童与青少年的消费规模不断增加,他们已然跃升成为巨大的消费群体。本研究主要通过代际消费决策风格、家庭理财教育方式和自我差异来探究中学生群体的消费社会化。研究—根据前人研究重新修订了消费决策风格量表,并具备较良好的信效度,可适用于中学生和成人群体,能作为后续研究的测量工具。研究二在国内各地的初、高中正式发放家长和中学生子女的消费决策风格、家庭理财教育方式和自我差异的问卷,共收集了2 15对有效数据。研究结果发现:(1)代际消费决策风格整体相关程度较高,父母和孩子都表现出了较高的质量认知导向和忠诚习惯性导向消费决策风格,但在品牌认知、休闲娱乐和决策困扰存在差异;(2)在父母和孩子不同的消费决策风格关系中,相应的理财教育方式会起到中介作用,如焦虑牢骚理财教育方式中介代际冲动性购买导向消费决策风格;(3)自我差异对质量认知导向、价格认知导向、冲动性购买导向的代际消费决策风格之间存在调节作用。

AIM To validate the effects of receptor interacting protein kinase-3(RIP3) deletion in non-alcoholic fatty liver disease(NAFLD) and to clarify the mechanism of *** Wild-type(WT) and RIP3 knockout(KO) mice werefed normal chow and high fat(HF) diets for 12 wk. The body weight was assessed once weekly. After 12 wk, the liver and serum samples were extracted. The liver tissue expression levels of RIP3, microsomal triglyceride transfer protein, protein disulfide isomerase, apolipoprotein-B, X-box binding protein-1, sterol regulatory element-binding protein-1c, fatty acid synthase, cluster of differentiation-36, diglyceride acyltransferase, peroxisome proliferator-activated receptor alpha, tumor necrosis factor-alpha(TNF-α), and interleukin-6 were assessed. Oleic acid treated primary hepatocytes from WT and RIP3 KO mice were stained with Nile red. The expression of inflammatory cytokines, including chemokine(C-X-C motif) ligand(CXCL) 1, CXCL2, and TNF-α, in monocytes was *** RIP3 KO HF diet fed mice showed a significant gain in body weight, and liver weight, liver to body weight ratio, and liver triglycerides were increased in HF diet fed RIP3 KO mice compared to HF diet fed WT mice. RIP3 KO primary hepatocytes also had increased intracellular fat droplets compared to WT primary hepatocytes after oleic acid treatment. RIP3 overexpression decreased hepatic fat content. Quantitative real-time polymerase chain reaction analysis showed that the expression of very-low-density lipoproteins secretion markers(microsomal triglyceride transfer protein, protein disulfide isomerase, and apolipoprotein-B) was significantly suppressed in RIP3 KO mice. The overall NAFLD Activity Score was the same between WT and RIP3 KO mice; however, RIP3 KO mice had increased fatty change and decreased lobular inflammation compared to WT mice. Background: Single port laparoscopic cholecystectomy (SPLC) is a widely performed advanced technique in laparoscopic surgery which has many benefits compare to conventional three port laparoscopic cholecystectomy (TPLC). The purpose of this study was to compare the patients’ satisfaction of SPLC and TPLC after one year of operation by using questionnaire which not only objective factors such as results of operation and hospital days but also subjective factor such as social and cosmetic quality of life. Materials and Methods: This study analyzed the data of 74 patients (SPLC = 42, TPLC = 32) who underwent the laparoscopic cholecystectomy between March and july 2013. The patients were asked to complete a patient-assessment questionnaire measuring the postoperative social and cosmetic quality-of-life values at the 12-month mark. Results: Statistically significant differences in the age, sex distribution, and body mass index are absent between the two groups. All the average scores of the different parameters of the social-activity aspect—satisfaction with physical condition, limitation of nutrition, stamina, postoperative pain level, returning to social life, impairment of social life, degree of complications, and cost-effectiveness of the surgery—are not statistically significant in both groups. Alternatively, all the results of the cosmetic aspect—scar influence on charms (p p = 0.015), and patient’s score of scars (p < 0.001)—show statistically significant differences. Conclusion: The SPLC is a safe and an attractive approach regarding the patients of this study. It offers a more effective cosmetic result that may be conveyed by a greater patient satisfaction.
