The solute carrier family 12(SLC12)of cation-chloride cotransporters(CCCs)comprises potassium chloride cotransporters(KCCs,***1,KCC2,KCC3,and KCC4)-mediated Cl^(-)extrusion,and sodium potassium chloride cotransporters(n[K]CCs,nKCC1,nKCC2,and nCC)-mediated Cl^(-)*** CCCs play vital roles in cell volume regulation and ion ***-of-function or loss-of-function of these ion transporters can cause diseases in many *** recent years,there have been considerable advances in our understanding of CCCs'control mechanisms in cell volume regulations,with many techniques developed in studying the functions and activities of *** approaches to directly measure CCC activity involve assays that measure the transport of potassium substitutes through the *** techniques include the ammonium pulse technique,radioactive or nonradioactive rubidium ion uptakeassay,and thallium ion-uptake ***'activity can also be indirectly observed by measuring gaminobutyric acid(GABA)activity with patch-clamp electrophysiology and intracellular chloride concentration with sensitive microelectrodes,radiotracer^(36)Cl^(-),and fluorescent *** techniques include directly looking at kinase regulatory sites phosphorylation,flame photometry,22nat uptake assay,structural biology,molecular modeling,and high-throughput drug *** review summarizes the role of CCCs in genetic disorders and cell volume regulation,current methods applied in studying CCCs biology,and compounds developed that directly or indirectly target the CCCs for disease treatments.

Diversity of Insect Flower Visitors on Macadamia within a Monoculture Orchard in Murang’a County, Central Kenya

Macadamia is cultivated for its nutritious edible kernel. In Kenya, the crop is predominantly grown by smallholder farmers mainly for export markets. Macadamia trees also enhance agro-forestry conservation, in the East African farmlands, by providing habitat and floral resources to beneficial arthropods such as insect pollinators. Allogamy of macadamia flowers is largely dependent on insects that pollinate and consequently influence the nut set, retention and yield. However, there is limited information on macadamia insect flower visitors in Kenya. This article assessed the diversity of insects that forage flowers of macadamia crop, and further evaluated their temporal distribution, in Murang’a county, central Kenya. Insect flower visitors were sampled weekly using a butterfly net for twelve consecutive months, from January 2021 to December 2021. Sixty-one insect species were recorded foraging macadamia flowers in Murang’a county, central Kenya. There was a statistical difference in the Simpson diversity index among weather seasons, dry, long rain, cold and short rain (p Apis mellifera), a key pollinator species, being the most abundant flower visitor. This study provides strong baseline information, to scientists and farmers, on probable macadamia pollinator species, in central Kenya. .

Dioxin-like molecules have been associated with endocrine disruption and liver *** better understand aryl hydrocarbon receptor(AHR)biology,metabolic phenotyping and liver proteomics were performed in mice following ligand-activation or whole-body genetic ablation of this *** wild type(WT)and Ahr^(-/-) mice(Taconic)were fed a control diet and exposed to 3,3',4,4',5-pentachlorobiphenyl(PCB126)(61 nmol/kg by gavage)or vehicle for two ***126 increased expression of canonical AHR targets(Cyp1 a1 and Cyp1 a2)in WT but not Ahr^(-/-).Knockouts had increased adiposity with decreased glucose tolerance;smaller livers with increased steatosis and perilipin-2;and paradoxically decreased blood ***126 was associated with increased hepatic triglycerides in Ahr^(-/-).The liver proteome was impacted more so by Ahr^(-/-) genotype than ligandactivation,but top gene ontology(GO)processes were *** PCB126-associated liver proteome was *** principally regulated liver metabolism(e.g.,lipids,xenobiotics,organic acids)and bioenergetics,but it also impacted liver endocrine response(e.g.,the insulin receptor)and function,including the production of steroids,hepatokines,and pheromone binding *** effects could have been indirectly mediated by interacting transcription factors or *** biologic roles of the AHR and its ligands warrant more research in liver metabolic health and disease.

Clinician Utilization of Best Practices for HIV Care and Status Disclosure: A Cross-Sectional Study

Determining the baseline utilization of best practices among clinicians, with regard to HIV care and disclosure of status to at-risk partners, is critical to addressing HIV prevention issues and disease transmission. In this study, we examined clinicians' utilization of best practices for HIV/AIDS care and the disclosure of HIV status to sexual and needle-sharing partners (HSSnSP). We conducted a cross-sectional survey of 100 clinicians to assess knowledge, attitude, and behavior towards best practices for HIV care and disclosure of HSSnSP. Multiple logistic regression analysis was used to determine factors associated with the utilization of clinical best practices for HIV care and disclosure of HSSnSP. Forty eight percent of the participants revealed poor behaviors. Fifty six percent showed less positive attitudes towards clinical best practices for HIV care and disclosure of status. Less than half (32%) of the participants had earned at least 1 HIV/AIDS CME credit in the past two years. Our study showed strong associations between the utilization of best practices for HIV care and the disclosure of status, and clinician attitudes and knowledge levels. Further research that aims to improve knowledge and attitudes on clinical best practices for providing HIV/AIDS care and the disclosure of status is needed. This improvement may lead to increase in the utilization of best practices for HIV care and disclosure of status, and contribute to decreases in disease transmission rates.

Replacement of Fish Meal with Graded Levels of Earthworm Meal in the Diet of Fingerlings： Effect on Feed and Growth Parameters

The study was aimed at evaluating growth response of Heteroclarias fingerlings fed diet in which fish meal was substituted with graded levels of earthworm meal. The replacement levels were 0.00%, 25%, 50%, 75% and 100% designated as E0, E25, E50, E75 and E100, respectively. Fingerlings used for the work were obtained through artificial insemination of brood stock (purchased from local farmers) with ovaprim in the laboratory. The resulting fry were reared and fed with cultured plankton for 2 weeks followed by 100% fish meal for another two weeks. Thereafter sixteen fingerlings weighing 4.73 g and measuring 6.512 mm standard length (on the average) were starved over night and reared in five indoors tanks (0.8 × 0.5 × 0.5 m) in duplicate for eight weeks. Fingerlings were fed on graded earthworm meals of 40% protein twice daily at 08:00 and 16:00. The highest mean weight (6.77 g), specific growth rate (0.86) and protein efficiency ratio (0.6) were obtained at 50% replacement level. Also, Food conversion ratio was the highest at 50% substitution level and lowest at 0.00% substitution, indicating that fingerlings use earthworm containing meal more efficiently. Furthermore, cost benefit analysis revealed that the profit index for 50% inclusion of earthworm meal was the highest (1.71) and the highest net profit of n374.32 (circa $2.3) was achieved at this level. Water quality parameters observed were within tolerable values for the fingerlings well being. Consequently, farmers are advised to use 50% earthworm meal in place of fish meal for formulation of Heteroclarias fingerlings diet. This will drastically reduce production cost, increase farmers profit and enhance the biological value of our menu.

Alcoholic liver disease(ALD)comprises a spectrum of liver pathology,including steatosis,steatohepatitis,and *** work from our group and others suggests that dietary fat,both the amount and composition,plays a pivotal role in ALD development and progression;however,the impact of specific dietary fatty acids on ALD pathogenesis is not fully *** rodent models of ALD revealed the deleterious effects of omega-6 polyunsaturated fatty acids(n-6 PUFAs),specifically linoleic acid(LA),and this may be partially attributed to the increased levels of pro-inflammatory oxidized LA *** is limited understanding regarding the role of omega-3 polyunsaturated fatty acids(n-3 PUFAs,such as alpha-linolenic acid,eicosapentaenoic acid,and docosahexaenoic acid),and bioactive n-3 PUFAderived lipid molecules in *** that majority of n-6 and n-3 PUFAs-derived metabolites are potent endogenous signaling molecules,knowledge regarding the changes in these lipid mediators may shed new light on the mechanisms contributing to ALD pathogenesis and reveal novel therapeutic targets and biomarkers of this *** current review summarizes relevant scientific literature regarding the role of dietary fat,distinct fatty acids,and bioactive fatty acid metabolites in ALD,and highlights recent advances in the field.
