Optimization of Parameters for the Production of Lipase from Pseudomonas sp. BUP6 by Solid State Fermentation

Solid-state fermentation (SSF) holds tremendous potentials for the production of industrially significant enzymes. The present study describes the production of lipase by a novel rumen bacterium, Pseudomonas sp. strain BUP6 on agro-industrial residues. Pseudomonas sp. strain BUP6 showed higher lipase production when grown in Basal salt medium (BSM) supplemented with oil cakes. Initially, five different oil cakes (obtained after extracting oil from coconut, groundnut, cotton seed, gingelly or soybean) were screened to find out the most suitable substrate-cum-inducer for the production of lipase. Among them, groundnut cake supported the maximum production of lipase (107.44 U/gds). Box-Behnken design (BBd), followed by response surface methodology (RSM) was employed to optimize the culture parameters for maximizing the production of lipase. Using the software Minitab 14, four different parameters like temperature, pH, moisture content and incubation time were selected for the statistical optimization, which resulted in 0.7 fold increase (i.e., 180.75 U/gds) in production of lipase under the optimum culture conditions (temperature 28°C, pH 5.9, moisture 33% and incubation 2 d). Thus, this study signifies the importance of SSF for the production of industrially-significant lipase using agro-industrial residues as solid support.

Peanut （Arachis hypogaea; 2n = 4x = 40） is a nutritious food and a good source of vitamins, minerals, and healthy fats. Expansion of genetic and genomic resources for genetic enhancement of cultivated peanut has gained momentum from the sequenced genomes of the diploid ancestors of cultivated peanut. To facil- itate high-throughput genotyping of Arachis species, 20 genotypes were re-sequenced and genome-wide single nucleotide poiymorphisms （SNPs） were selected to develop a large-scale SNP genotyping array. For flexibility in genotyping applications, SNPs polymorphic between tetraploid and diploid species were included for use in cultivated and interspecific populations. A set of 384 accessions was used to test the array resulting in 54 564 markers that produced high-quality polymorphic clusters between diploid species, 47 116 polymorphic markers between cultivated and interspecific hybrids, and 15 897 polymorphic markers within A. hypogaea germplasm. An additional 1193 markers were identified that illuminated genomic re- gions exhibiting tetrasomic recombination. Furthermore, a set of elite cultivars that make up the pedigree of US runner germplasm were genotyped and used to identify genomic regions that have undergone pos- itive selection. These observations provide key insights on the inclusion of new genetic diversity in culti- vated peanut and will inform the development of high-resolution mapping populations. due to its efficiency, scope, and flexibility, the newly developed SNP array will be very useful for further genetic and breeding applications in Arachis.

由于投影数据中存在大量的散射线,导致使用CBCT对含有铝、铬镍铁合金和其他金属的物体进行断层扫描时存在巨大的挑战.一次旋转扫描可采集大量的体积数据,还可以减轻物体平移的负担;但是,投影时广域射束会生成大量的散射,如果不进行相应的校正,生成的影像可能就会出现杯状伪影、阴影、条纹、虚假尺寸和量化误差.投影数据散射最常见的一种估算和校正方法就是反卷积法(也被称为基于核的校正),而该方法对工业应用中常见的不均匀、形状不规则的物体,效果往往较差.虽然通过单纯的蒙特卡罗(MC)算法随机解线性玻尔兹曼方程(LBTE),可以对产生的散射线进行精确的估算,但该算法执行时间太长,只适用于学术和实验室应用.3d VSHARP是一款快速的基于有限元的线性玻尔兹曼方程(LBTE)解算器,不仅可以模拟光子传输,而且可以计算投影数据中预期的散-原射线比.使用3d VSHARP时需要创建一个成像系统模型和一个物体模型.成像系统模型旨在描述系统几何结构、X射线源组件(多能谱、光束滤波和硬件准直)和X射线探测器组件(本用例中为平板探测器).物体模型由3d材料密度图构成,而3d材料密度图从第一步重建开始创建,可以通过计算机辅助设计(CAd)文件进行进一步了解.随后,3d VSHARP将使用系统模型和物体模型来估算投影影像的散射分量,并对其进行相应的校正,然后进行第二步重建.对重叠金属块进行蒙特卡罗(MC)模拟的验证结果显示,3d VSHARP和MC计算得出的散射分数都在97% RMS范围内.每次投影的3d VSHARP执行时间都不超过1秒钟(使用单一NVidia TitanXp GPU),而MC模拟花费的时间为1566个CPU小时.

近年来,在全球能源转型的大背景下,高温地热发电得到世界各国极大的重视,前景广阔。要实现对高温地热能源的开采,采用的钻井技术是关键。北美从20世纪50年代开始,对地热能源商业化开采、并用以发电的技术进行了持续性的研究和试验。在高温硬岩地热钻井技术上形成了完整的体系,比如高温地热井设计与井筒完整性技术,高温、超高温坚硬岩石钻头设计与生产技术,高温井底动力钻具,高温井下测量工具,高温泥浆与堵漏技术,高温水泥浆体系以及高温井控技术等。本文结合作者在国内地热井钻完井及北美地热井的工作经验,概要介绍北美的地热钻井技术现状、地热钻井技术难点以及与之相关的关键技术。

背景：膝关节前髁解剖形态存在明显性别差异的观点已使存全膝关节假体设计时专门为女性患者设计了薄型前髁假体。我们通过对自己的患者进行观察，发现前髁的解剖形态变异很大，不仅取决于患者的性别，还取决于患者的身材、身高和人种。鉴于所发现的膝关节前髁解剖存在较大变异，我们试图明确是否真正存在前髁解剖的性别差异。方法：我们对212例随机挑选患者的MRI（男112例，女100例）进行评估。前髁定义为膝关节间隙以上10mm的股骨前侧皮质前方的骨性区域。前髁的内、外侧高度直接从MRI两个层面的标记值读出，以毫米为单位。经计算得出的髁高横径比用来确定患者的身材对前髁的大小是否有影响。

随着177Lu-1,4,7,10-四氮杂环十二烷-1,4,7,10-四乙酸-d-苯丙氨酸1-酪氨酸3-苏氨酸8-奥曲肽(dOTATATE)肽受体放射性核素治疗(PRRT)的日益普及,有关患者合理选择的问题日益增多。该文回顾了有关177Lu-dOTATATE在不同原发性神经内分泌肿瘤(NETs)中的应用文献,讨论了一些有争议的问题,并回顾了治疗的潜在禁忌证。此共识是由北美神经内分泌肿瘤学会(NANETS)和核医学与分子影像学会(SNMMI)共同制定的。NANETS是成立于2005年的北美多学科神经内分泌专家专业学会,目的在于通过增加研究和教育机会改善对NETs的管理。NANETS致力于多学科研究,由NETs涉及的不同专业的医师和科学家组成。SNMMI成立于1954年,旨在促进核医学科学、技术和实践应用的国际性科学专业组织,除了出版期刊、时事通讯和书籍外,还主办提高核医学从业人员能力和促进核医学科学新发展的国际会议及讲习班。
