Engineered neuronal microtissue provides exogenous axons for delayed nerve fusion and rapid neuromuscular recovery in rats

Nerve injury requiring surgical repair often results in poor functional recovery due to the inability of host axons to re-grow long distances and reform meaningful connections with the target muscle. While surgeons can re-route local axon fascicles to the target muscle, there are no technologies to provide an exogenous source of axons without sacrificing healthy nerves. Accordingly, we have developed tissue engineered neuromuscular interfaces (TE-NmIs) as the first injectable microtissue containing motor and sensory neurons in an anatomically-inspired architecture. TE-NmIs provide axon tracts that are intended to integrate with denervated distal structures and preserve regenerative capacity during prolonged periods without host innervation. Following implant, we found that TE-NmI axons promoted Schwann cell maintenance, integrated with distal muscle, and preserved an evoked muscle response out to 20-weeks post nerve transection in absence of innervation from host axons. By repopulating the distal sheath with exogenous axons, TE-NmIs also enabled putative delayed fusion with proximal host axons, a phenomenon previously not achievable in delayed repair scenarios due to distal axon degeneration. Here, we found immediate electrophysiological recovery after fusion with proximal host axons and improved axon maturation and muscle reinnervation at 24-weeks post-transection (4-weeks following delayed nerve fusion). These findings show that TE-NmIs provide the potential to improve functional recovery following delayed nerve repair.

Holographic imaging offers a reliable and fast method to capture the complete three-dimensional （3D） information of the scene from a single perspective. We review our recently proposed single-channel optical system for generating digital Fresnel holograms of 3D real-existing objects illuminated by incoherent light. In this motionless holographic technique, light is reflected, or emitted from a 3D object, propagates through a spatial light modulator （SLm）, and is recorded by a digital camera. The SLm is used as a beamsplitter of the single-channel incoherent interferometer, such that each spherical beam originated from each object point is split into two spherical beams with two different curve radii. Incoherent sum of the entire interferences between all the couples of spherical beams creates the Fresnel hologram of the observed 3D object. When this hologram is reconstructed in the computer, the 3D properties of the object are revealed.

Imaging with an optical incoherent synthetic aperture (SA) means that the incoherent light from observed objects is processed over time from various points of view to obtain a resolution equivalent to single-shot imaging by the SA larger than the actual physical aperture. The operation of such systems has always been based on two-wave interference where the beams propagate through two separate channels. This limitation of two channels at a time is removed in the present study with the proposed SA where the two beams pass through the same single channel at any given time. The system is based on a newly developed self-interference technique named coded aperture correlation holography. At any given time,the recorded intensity is obtained from interference between two waves co-propagating through the same physical channel. One wave oriented in a particular polarization is modulated by a pseudorandom coded phase mask and the other one oriented orthogonally passes through an open subaperture. Both subapertures are multiplexed at the same physical window. The system is calibrated by a point spread hologram synthesized from the responses of a guide star. All the measurements are digitally processed to achieve a final image with a resolution higher than that obtained by the limited physical aperture. This unique configuration can offer alternatives for the current cumbersome systems composed of far apart optical channels in the large optical astronomical interferometers. Furthermore,the proposed concept paves the way to an SA system with a single less-expensive compact light collector in an incoherent optical regime that may be utilized for future ground-based or space telescopes.

Assembly of two-dimensional (2D) nanomaterials into well-organized architectures is pivotal for controlling their function and enhancing performance. As a promising class of 2D nanomaterials, mXenes have attracted significant interest for use in wearable electronics due to their unique electrical and mechanical properties. However, facile approaches for fabricating mXenes into macroscopic fibers with controllable structures are limited. In this study, we present a strategy for easily spinning mXene fibers by incorporating polyanions. The introduction of poly(acrylic acid) (PAA) into mXene colloids has been found to alter mXene aggregation behavior, resulting in a reduced concentration threshold for lyotropic liquid crystal phase. This modification also enhances the viscosity and shear sensitivity of mXene colloids. Consequently, we were able to draw continuous fibers directly from the gel of mXene aggregated with PAA. These fibers exhibit homogeneous diameter and high alignment of mXene nanosheets, attributed to the shear-induced long-range order of the liquid crystal phase. Furthermore, we demonstrate proof-of-concept applications of the ordered mXene fibers, including textile-based supercapacitor, sensor and electrical thermal management, highlighting their great potential applied in wearable electronics. This work provides a guideline for processing 2D materials into controllable hierarchical structures by regulating aggregation behavior through the addition of ionic polymers.
