PAR1 is an atypical basic-helix-loop-helix （bHLH） protein that negatively regulates the shade avoidance syndrome in Arabidopsis thaliana acting as a transcriptional cofactor. Consistently with this function, PAR1 has to be in the nucleus to display biological activity. Previous structure-function analyses revealed that the N-terminal region of PAR1 drives the protein to the nucleus. However, truncated forms of PAR1 lacking this region still display biological activity, implying that PAR1 has additional mechanisms to localize into the nucleus. In this work, we compared the primary structure of PAR1 and various related and unrelated plant bHLH proteins, which led us to suggest that PAR1 contains a non-canonical nuclear localization signal （NLS） in the N-terminal region. By overexpressing truncated and mutated derivatives of PAR1, we have also investigated the importance of other regions of PAR1, such as the acidic and the extended HLH dimerization domains, for its nuclear localization. We found that, in the absence of the N-terminal region, a functional HLH domain is required for nuclear localization. Our results suggest the existence of a dual mechanism for PAR1 nuclear localization：（1） one mediated by the N-terminal non-consensus NLS and （2） a second one that involves interaction with other proteins via the dimerization domain,

Computed tomography (CT)-guided lung biopsy is a common diagnostic procedure that is associated with various complications, including pneumothorax, hemoptysis and parenchymal hemorrhage. Systemic air embolism is a very rare (0.07%) but potentially lifethreatening complication. We report a fatal case of air embolism to the cerebral and coronary arteries confirmed by head and chest CT, followed by a review of the literature.

Since the birth of the first cloned sheep,somatic cell nuclear transfer technology has been successfully used to clone a variety of *** livestock have no apparent health risks,and the quality and safety of the cloned animal products are similar to non-cloned *** social behavior and environmental adaptability of postnatal cloned animals,especially when used for grassland farm production purposes,is *** the present study,the cloned Dorper sheep equipped with GPS location devices were free-grazed in a harsh natural environment similar to conditions commonly experienced by Mongolian *** main findings of this research were as follows.(1)Under free-grazing conditions,the cloned sheep showed excellent climatic and ecological *** extreme temperature conditions ranging from–30 to 40°C,the cloned sheep maintained acceptable body condition and behaved as other sheep.(2)The cloned sheep quickly adapted from a herd feeding strategy to the harsh environment and quickly exhibited a grazing regimen as other free-grazing sheep.(3)The cloned sheep exhibited free-grazing patterns and social behavior as other sheep.(4)The cloned sheep in the harsh environment thrived and produced healthy ***,the cloned Dorper sheep exhibited excellent ecological adaptation,which is an important consideration for breeding meat sheep by *** Dorper sheep readily adapted to the free-grazing conditions on the Mongolian plateau grassland,which attests to their ability to withstand harsh environmental conditions.

A right-hand motor imagery based brain-computer interface is proposed in this work. Such a system requires the identification of different brain states and their classification. Brain signals recorded by electroencephalography are naturally contaminated by various noises and interferences. Ocular artifact removal is performed by implementing an auto-matic method “Kmeans-ICA” which does not require a reference channel. This method starts by decomposing EEG signals into Independent Components;artefactual ones are then identified using Kmeans clustering, a non-supervised machine learning technique. After signal preprocessing, a Brain computer interface system is implemented;physiologically interpretable features extracting the wavelet-coherence, the wavelet-phase locking value and band power are computed and introduced into a statistical test to check for a significant difference between relaxed and motor imagery states. Features which pass the test are conserved and used for classification. Leave One Out Cross Validation is performed to evaluate the performance of the classifier. Two types of classifiers are compared: a Linear Discriminant Analysis and a Support Vector Machine. Using a Linear Discriminant Analysis, classification accuracy improved from 66% to 88.10% after ocular artifacts removal using Kmeans-ICA. The proposed methodology outperformed state of art feature extraction methods, namely, the mu rhythm band power.

采用热模拟试验机、电子显微镜、扫描电镜等手段,研究了冷却速度对低合金高强钢显微组织的影响。结果表明:在0.2~3.0℃/s低冷却速度条件下,低合金高强试验钢的显微组织变化为从铁素体/珠光体+下贝氏体到铁素体+下贝氏体,再到铁素体+针状铁素体,最后在3℃/s冷却冷却速度下,试验钢为单一的针状铁素体组织。随着冷却速度的提高,试验钢中铁素体体积分数逐渐降低。在10~60℃/s高冷却速度条件下,试验钢从状铁素体+马氏体混合组织逐渐转变为单一马氏体组织,随着冷却速度提高,针状铁素体含量逐渐减少,马氏体含量逐渐增加。

Colorectal cancer(CRC) is a devastating disease, mainly because of metastasis. As a result, there is a need to better understand the molecular basis of invasion and metastasis and to identify new biomarkers and therapeutic targets to aid in managing these tumors. The actin cytoskeleton and actin-binding proteins are known to play an important role in the process of cancer metastasis because they control and execute essential steps in cell motility and contractility as well as cell division. Caldesmon(CaD) is an actin-binding protein encoded by the CALD1 gene as multiple transcripts that mainly encode two protein isoforms: High-molecular-weight CaD, expressed in smooth muscle, and low-molecular weight CaD(l-CaD), expressed in nonsmooth muscle cells. According to our comprehensive review of the literature, CaD, particularly l-CaD, plays a key role in the development, metastasis, and resistance to chemoradiotherapy in colorectal, breast, and urinary bladder cancers and gliomas, among other malignancies. CaD is involved in many aspects of the carcinogenic hallmarks, including epithelial mesenchymal transition via transforming growth factor-beta signaling, angiogenesis, resistance to hormonal therapy, and immune evasion. Recent data show that CaD is expressed in tumor cells as well as in stromal cells, such as cancerassociated fibroblasts, where it modulates the tumor microenvironment to favor the tumor. Interestingly, CaD undergoes selective tumor-specific splicing, and the resulting isoforms are generally not expressed in normal tissues, making these transcripts ideal targets for drug design. In this review, we will analyze these features of CaD with a focus on CRC and show how the currently available data qualify CaD as a potential candidate for targeted therapy in addition to its role in the diagnosis and prognosis of cancer.
