Production of transgenic cashmere goat embryos expressing red fluorescent protein and containing IGF1 hair-follicle-cell specific expression cassette by somatic cell nuclear transfer

In the present study, cashmere goat fetal fibroblasts were transfected with pCDsR-KI, a hair-follicle-cell specific expression vector for insulin-like growth factor 1 (IGF1) that contains two markers for selection (red fluorescent protein gene and neomycin resistant gene). The transgenic fibroblasts cell lines were obtained after G418 selection. Prior to the somatic cell nuclear transfer (SCNT), the maturation rate of caprine cumulus oocytes complexes (COCs) was optimized to an in vitro maturation time of 18 h. Parthenogenetic ooctyes were used as a model to investigate the effect of two activation methods, one with calcium ionophore IA23187 plus 6-DMAP and the other with ethanol plus 6-DMAP. The cleavage rates after 48 h were respectively 88.7% and 86.4%, with no significant difference (P>0.05). There was no significant difference between the cleavage rate and the blastocyst rate in two different media (SO- Faa and CR1aa; 86.3% vs 83.9%, P>0.05 and 23.1% vs 17.2%,P>0.05). The fusion rate of a 190 V/mm group (62.4%) was significantly higher than 130 V/mm (32.8%) and 200 V/mm (42.9%), groups (P<0.05). After transgenic somatic cell nuclear transfer (TSCNT) manipulation, 203 reconstructed embryos were obtained in which the cleavage rate after in vitro development (IVD) for 48 h was 79.3% (161/203). The blastocyst rate after IVD for 7 to 9 d was 15.3% (31/203). There were 17 embryos out of 31 strongly ex- pressing red fluorescence. Two of the red fluorescent blastocysts were randomly selected to identify transgene by polymerase chain reaction. Both were positive. These results showed that: (i) RFP and Neor genes were correctly expressed indicating that transgenic somatic cell lines and positive trans- genic embryos were obtained; (ii) one more selection at the blastocyst stage was necessary although the donor cells were transgenic positive, because only partially transgenic embryos expressing red fluorescence were obtained; and (iii) through TSCNT manipulation and optim

Prospect of creating trans-genic animals by using sper-matogonial transplantation

Transgenic animal technology is a powerful tool for researching bioscience, biomedicine, bioreactor, and ag- riculture. There are various ways to produce transgenic animals. The most common ways currently available are pronuclear microinjection and nuclear transfer techniques. However, these methods usually result in low efficiency, causing mosaic (in pronuclear microinjection), or develop- mental abnormalities (in nuclear transfer). In 1994, Brinster and his colleagues reported an original method to transfer spermatogonial stem cells from donor to recipient mice. The donor spermatogonia were able to form spermatozoa in re- cipient testes, and to produce progeny carrying the donor’s genetic characters. Since then, a series of novel methods were invented by using spermatogonia transplantation. These new methods facilitate the research and application of sper- matogonia. Some of these methods, when combining with genetic modification methods, will form a novel methodology for creating transgenic animals. The present paper reviews the achievements of research on spermatogonia transplanta- tion related to creating transgenic animal. Such as, trans- plantation techniques, cryopreservation of spermatogonia, preparation of recipients, long-term proliferation of sper- matogonia in culture, genetic modification of spermatogonia, and characterization of germ line transmission of the modi- fied gene, etc. Furthermore the methodologies for creating transgenic animals by using spermatogonia transplantation were described. Based on the difference between donors and recipients used, the methodology is categorized into two groups: allogeneic transplantation, and autologous trans- plantation. Although progress in this research area has been swift, potential difficulties remain to be overcome in each approach. The advantages and existing problems in the methodology are discussed.

Adverse reactions to mesalamine,a treatment used to induce and maintain remission in inflammatory bowel diseases,particularly ulcerative colitis,have been described in the literature as case *** case illustrates an unusual adverse *** patient developed an isolated fever of unexplained etiology,which was found to be related to mesalamine treatment.A 22-year-old patient diagnosed with ulcerative colitis developed a fever with rigors and anorexia 10 d after starting oral mesalamine while his colitis was clinically *** revealed no infection.A mesalamineinduced fever was considered,and treatment was stopped,which led to spontaneous resolution of the *** diagnosis was confirmed by reintroducing the *** year later,this side effect was noticed again in the same patient after he was administered topical *** reaction to mesalamine seems to be idiosyncratic,and the mechanism that induces fever remains *** encountered in the course of a mesalamine treatment in ulcerative colitis must be considered a mesalamine-induced fever when it cannot be explained by the disease activity,an associated extraintestinal manifestation,or an infectious etiology.

Sessile oak forest plant community changes on the NE Iberian Peninsula over recent decades

Aims Our aims were 3-fold:(i)to determine whether global change has altered the composition and structure of the plant community found in the sessile oak forests on the NE Iberian Peninsula over the last decades,(ii)to establish whether the decline in forest exploitation activities that has taken place since the mid-20th century has had any effect on the forests and(iii)to ascertain whether there is any evidence of impact from climate *** We assess changes in the plant community by comparing a cur-rent survey of sessile oak forest with a historical data set obtained from previous regional studies dating from 1962 to *** ana-lyse the regional changes in the community in terms of biodiversity variables,species composition and plant ***,plants traits such as plant life forms and chorological groups are used to discern any effects from land-use changes and climate warming on the plant *** Findingsthere has been a loss of diversity in the community and,in the hottest region,there is also a loss of species *** composition of the community suggests that,although significant changes have taken place over recent decades,these changes differ between regions as a result of the low impact global change has had in the western *** instance,while the tree canopy cover in the western sessile oak forests remains stable,the eastern sessile oak forests are still recovering from the former exploitation that led to a loss of their rich and abundant herbaceous *** fact,the recovery process in the catalan Pre-coastal Range has constituted an increase in the Euro-Siberian plants typical to this ***,in the eastern forests,there is evidence that climate warming has impacted the thermophilization of the sessile oak forests found on the coastal Range.

Antioxidant and anti-diabetic activities in commercial and homemade pomegranate molasses in Lebanon

Pomegranate fruit and its derived products are rich sources of bioactive *** have many biological *** molasses(PM)is a thick,traditional Middle Eastern syrup used in many recipes for Lebanese and international *** is a highly nutritious product which makes it of great *** purpose of this study was to assess the phytochemical composition,antioxidant and anti-diabetic properties of homemade and commercial PM consumed in Lebanon using samples collected from households in rural areas(n=4)and the market(n=28).The objectives of this study were to determine their total phenolic content using the Follin-Ciocalteu method,their total flavonoid content using aluminum chloride method,their antioxidant activity using DPPH radical scavenging as well as ferrous ion chelating assays and their anti-diabetic activity usingα-amylase andα-glucosidase inhibitory activities with acarbose,a known anti-diabetic drug,used as the standard *** PM samples exhibited a higher antioxidant activity than commercial samples,with the most active homemade PM sample having IC_(50)values of 0.09 mg/mL(DPPH radical scavenging assay)and 46.78%ferrous ion chelating assay).Homemade PM samples also showed higher anti-diabetic activity than commercial samples,with the most active homemade PM sample having an IC_(50)values of 0.63 mg/mL(α-amylase inhibitory activity)and of 0.41 mg/mL(α-glucosidase inhibitory activity)and being almost as active as acarbose with both enzymes(1.5 times).Significant inverse strong/moderate correlations were observed between total phenolic content/total flavonoid content and the IC_(50)value of DPPH radical scavenging assay,indicating positive associations between total phenolic and flavonoid contents and antioxidant *** addition,significant negative moderate correlations were observed between total phenolic/flavonoid contents andα-amylase inhibitory activity,indicating positive associations between total phenoli

A CFD modeling of oil-water flow in pipeline: Interaction analysis and identification of boundary separation

The aim of this study is to simulate the flow of emulsion (immiscible liquids) which is a complex phenomenon in the petroleum industry. In this study, three types of emulsions (O/W) were simulated usingby CFD which has been applied to predict the interaction behavior of oil with water for differentemulsions (25%Oe75%W, 50%Oe50%W and 75%Oe25%W). Numerical modeling illustrated that the obtained results of the velocity profiles were very good agreement with the experimental data for threetype of emulsions. In addition, the analytical model of interaction analysis and identification of boundaryseparation for two-phase flow approve with the experimental data available, despite the emulsionsremain to be more difficulty due to the complex chemical reactions. The results of this study can providethe best solutions for corrosion during hydrocarbons transportation. Thus, CFD approach is recommended to use for numerical investigates due to the high accuracy.

Two-point compression ultrasonography: Enough to rule out lower extremity deep venous thrombosis?

BACKGROUND: Deep venous thrombosis(DVT) is a major cause of morbidity and is a common presenting complaint to the emergency department(ED). Point-of-care two-point compression ultrasonography has evolved as a quick and effective way of diagnosing DVT. The purpose of this study is to validate the prevalence and distribution of venous thrombi isolated to proximal lower extremity veins, other than common femoral and popliteal veins in patients with ***: This is a single-center retrospective study that looked at patients presenting to the ED of a tertiary care hospital between January 2014 and August 2018. The clinical presentation and laboratory and imaging results were obtained using the hospital's electronic medical ***: A total of 2,507 patients underwent a lower extremity duplex ultrasound during the study period. Among them, 379(15%) were included in the study. The percentages of isolated thrombi to the femoral vein and deep femoral vein were 7.92% and 0.53%, respectively. When the patients were stratified into the two groups of isolated DVT and two-point compression DVT, there were no statistically significant differences in the laboratory results between both groups. However, immobilized patients and patients with recent surgeries were more likely to have an isolated ***: Thrombi isolated to proximal lower extremity veins other than the common femoral and popliteal veins make up 8.45% of DVTs. Given this significant number of missed DVTs, the authors recommend the addition of the femoral and deep femoral veins to the two-point compression exam.
