Penicillin Resistance in Nasopharyngeal Streptococcus pneumoniae among Children with Sickle Cell Disease Immunized with 7-Valent Pneumococcal Conjugate Vaccine

To assess the prevalence, the antimicrobials resistance and to identify risk factors of nasopharyngeal colonization by Streptococcus pneumoniae in children with sickle cell disease (SCD) using prophylactic penicillin and immunized with 7-valent pneumococcal conjugate vaccine. A total of 424 swabs were collected from the nasopharynx in 216 children with SCD and 109 samples from the control group, both from 2 to 60 months age range. Isolation and identification of pneumococci followed standard procedures. Minimum inhibitory concentration (MIC) for penicillin was determined by the E-test method. Prevalence of nasopharyngeal colonization by pneumococci in children with SCD was 17%, and 11% in the control group. The risk factors for increased colonization in children with SCD were the presence of more than five people at home, daycare/school attendance and low prophylaxis compliance. The prevalence of strains with penicillin resistance was 57.5% in patients with SCD and 25% in the control group. Conclusions: Prevalence of pneumococci nasopharyngeal colonization was similar among the study groups. Prophylactic use of penicillin may have increased the prevalence of resistant strains. The vaccine did not decrease the colonization with penicillin resistant strains.

Sida pilosa Retz. (Malvaceae) is a medicinal plant used in Africa for the treatment of dysmenorrhea, lower abdominal pains and intestinal helminthiasis. S. pilosa aqueous extract and derived fractions were investigated for their bioactivity against Schistosoma mansoni. The aqueous extract from S. pilosa aerial parts (1.25 - 40 mg/mL) and derived fractions (n-hexane, DCM, EtOAc and n-BuOH: 0.25 - 8 mg/mL) were tested on adult S. mansoni maintained in a GMEN culture medium. Praziquantel was used as the reference drug. After 24 h of incubation, worms were monitored for their viability and egg output. The antioxidant activity of S. pilosa was evaluated by the ability to scavenge the 2,2-diphenyl-1-picrylhydrazyl free radicals. The chemical composition of the n-BuOH fraction was investigated by HPLC-MS analysis. S. pilosa aqueous extract and fractions significantly increased worm mortality in a concentration-dependent manner. The n-BuOH fraction was the most active with a LC50 of 1.25 mg/mL. Significant reduction of motor activity (25% to 100%) was recorded for surviving worms incubated in different concentrations of the extract and fractions. Incubation of S. mansoni in different concentrations of S. pilosa extract and fractions led to significant reduction of egg laying (52% to 100%). The aqueous extract and derived fractions exhibited antioxidant activity in a concentration-dependent manner. The highest antioxidant activity was found with the EtOAc fraction, followed by the DCM and n-BuOH fractions. HPLC-MS analysis of the n-butanol fraction revealed the presence of two indoloquinoline alkaloids. This study disclosed the schistosomicidal activity of the n-butanol fraction from S. pilosa aqueous extract. This activity is probably related to the indoloquinoline alkaloids identified in the fraction.

The possibility of commercialization of Moro blood oranges in tropical countries such as Brazil was evaluated to verify whether post-harvest management through storage at low temperatures for a period of 60 days can improve the bioactive properties and quality parameters. Moro blood oranges cultivated in Brazil did not contain significant amounts of anthocyanins at the time of harvesting, but these compounds were activated by post-harvest management through storage at low temperatures (4°C and 8°C) for a period of 60 days. The emergence of the anthocyanins in the juices occurred within a few weeks of storage, but the maximum levels were attained after 60 days and at the temperature of 8°C. Cold storage positively influenced other bioactive compounds such as total phenolic compounds, individual phenolic compounds, β-carotene and the antioxidant activity determined by the sequestration of DPPH free radicals. It did not influence the vitamin C content. In addition, storage significantly altered the color, total acidity and pH of the fruits, but it did not prevent its commercial use. The remaining quality parameters were not influenced. It is possible to commercialize these oranges in Brazil through post-harvest management.

Background: This study aimed to determine the litterfall production, accumulation, decomposition rate and nutrient stocks, and to estimate the soil carbon(C) and nitrogen(N) stocks in three palm swamp ecosystems with different land use intensities in the Southeast of ***: Three palm swamp ecosystems with different land use intensities were evaluated: Agua Doce(AD),conserved area;Capivara(CV), area with small agricultural practices;and Buriti Grosso(BG), area with an intensive land use. Produced and accumulated litterfall from the Cerrado areas surrounding the palm swamps were collected from October 2014 to September 2015, and the Ca, Mg, K, and P concentrations were determined in the dry and rainy seasons. Soil samples were collected in the surrounding Cerrado and within the palm swamps until 100 cm soil depth to determine bulk density and soil C and N contents and ***: Annual litterfall production in the Cerrado surrounding palm swamps was similar in AD(3.58 Mg·ha^-1·year^-1) and CV(3.79 Mg·ha^-1·year^-1), and was lower in BG(2.84 Mg·ha^-1·year^-1), and was more intense during the dry season. Furthermore, litterfall accumulation was higher in CV(7.12 Mg·ha^-1·year^-1) and BG(6.75 Mg·ha^-1·year^-1), culminating in lower decomposition rates. AD showed the highest decomposition rate(0.60) due to its vegetation structure and conservation. The macronutrient contents from the production and accumulated litterfall had a low influence of the land use, decreasing in the following order: Ca > K > Mg > P. Soils in CV and BG palm swamp areas showed higher values of C and N contents, due to the use of land for agricultural crops and cattle raising in an extensive(rangeland) system. This work describes the chemical composition of the essential oils extracted from fresh leaves of Lippia origanoides Kunth. and Mentha spicata L. and their antioxidant and insecticidal activities. The essential oils were extracted by steam distillation using a modified Clevenger apparatus and subsequently analyzed by gas chromatography-flame ionization detector and gas chromatography-mass spectrometry. The antioxidant activities were determined by the β-carotene-linoleic acid method and by sequestration of the 2,2-diphenyl-1-picryl-hidrazila radical. The concentrations of the essential oils and the synthetic standard, butylated hydroxyltoluene, were 25, 50, 100, 150, 200, 250, 300 and 500 μg mL-1. Insecticidal activity was analyzed by non-preference with choice and no choice against the aphid Myzus persicae Sulzer. Gas chromatography analysis of the essential oil from Lippia origanoides Kunth. revealed carvacrol (41.51%), p-cymene (18.36%), γ-terpinene (17.03%) and thymol (4.86%) as major constituents, and the essential oil from Mentha spicata L. contained piperitona (81.18%), piperitenone (14.57%) and limonene (1.47%) as the principal components. The essential oils and the standard exhibited dose-dependent antioxidant activities at the concentrations tested. The essential oils were shown to be potential agents in the integrated management of the aphid Myzus persicae Sulzer.

The coagulating potential of the crude extract from the barbatimao bark for the treatment of dairy effluent was assessed and compared with industrial coagulants such as aluminum sulfate (inorganic coagulant), the crude extract of black wattle bark (used in the manufacture of natural organic coagulants), and gallic acid (the main constituent of condensed tannins). The aqueous extract of barbatimao was obtained by refluxing for 12 h, and the black wattle was provided by a producer of natural coagulants. The determinations of total phenolic compounds and tannins in the extracts were performed by the Folin-Ciocalteu method and precipitation with casein, respectively. The treatment of the effluent with coagulant was performed by the Jar-Test. After testing, the effluent was evaluated for turbidity, total solids, and biochemical oxygen demand (BOD). A higher content of phenolic compounds (19.81% mg of gallic acid/g extract) was observed in the barbatimao extract than in the black wattle extract (14.24% mg of gallic acid/g extract). However, the extracts contained the same amount of tannins, 13.95% for the barbatimao extract and 12.89% for the black wattle extract. A 63%, reduction in BOD was observed for the effluent treated with barbatimao extract, 35% for the black wattle extract, 58% for gallic acid and 60.59% for the treatment with aluminum sulfate. Treatment with gallic acid caused a 98% reduction in the turbidity of the effluent;with aluminum sulfate, a 97% reduction was obtained;with barbatimao extract, 96%, and with black wattle extract, 93%. The barbatimao extract was shown to be a potential coagulant for dairy effluents.

The present work sought to characterize the cyanogenic glycoside dhurrin in five sugarcane varieties and to determine its possible relationship with the formation of ethyl carbamate in cachaca. For each variety, methanol, ethyl acetate and hexane extracts were prepared and submitted to thin layer chromatography. Chromatographic spots were revealed with iodine vapors. The physical and chemical parameters of the spirits produced from the five different varieties of sugar cane and by different production processes were within the legal limits. Although not exceeding the limit for ethyl carbamate, a significant variation in the final concentration of this compound was observed for each sample analyzed. The Rf values for the dhurrin standard and for the sugarcane variety SP 83-2847 were equal, similar to those for SP 80-3280 and CTC 11 and different from the Rf values for the RB 86-7515 and IAC 86-2480 samples.

Obesity is a difficult disease to control;bariatric surgery is one of the tools used to treat obesity and its comorbidities. The present case was chosen because it concerns a very obese patient whose proposed surgery—duodenal switch—resulted in traoperatively difficult to the medical team because the esophagogastric junction could not be reached due to the presence of hepatomegaly. Due to this unfavorable condition, we decided to perform a duodeno-ileal anastomosis with gastric preservation and without duodenal exclusion. The patient has been under follow-up for 14 years. She has lost 55 kg and maintains the comorbidities controlled to date.
