AIM: To elucidate the mechanism(s) by which S-adenosyl-l-methionine(SAM) decreases hepatitis C virus(hCV) ***: We examined the effects of SAM on viral expression using an hCV subgenomic replicon cell culture system. huh7 hCV-replicon cells were treated with 1 mmol/l SAM for different times(24-72 h), then total RNA and proteins were isolated. c DNA was synthesized and real time-PCR was achieved to quantify hCV-RNA, superoxide dismutase 1 and 2(SOD-1, SOD-2) catalase, thioredoxin 1, methionine adenosyltransferase 1A and 2A(MAT1A, MAT2A) expression, and GAPDh and RPS18 as endogenous genes. Expression of cellular and viral protein was evaluated by western-blot analysis using antibodies vs hCV-NS5 A, SOD-1, SOD-2, catalase, thioredoxin-1, MAT1 A, MAT2 A, GAPDh and actin. Total glutathione levels were measured at different times by Ellman's recycling method(0-24 h). Reactive oxidative species(ROS) levels were quantified by the dichlorofluorescein assay(0-48 h); Pyrrolidin dithiocarbamate(PDTC) was tested as an antioxidant control and h2O2 as a positive oxidant ***: SAM exposition decreased hCV-RNA levels 50%-70% compared to non-treated controls(24-72 h). SAM induced a synergic antiviral effect with standard IFN treatment but it was independent of IFN signaling. In addition, 1 mmol/l SAM exposition did not modify viral RNA stability, but it needs cellular translation machinery in order to decrease hCV expression. Total glutathione levels increased upon SAM treatment in hCV-replicon cells. Transcriptional antioxidant enzyme expression(SOD-1, SOD-2 and thioredoxin-1) was increased at different times but interestingly, there was no significant change in ROS levels upon SAM treatment, contrary to what was detected with PDTC treatment, where an average 40% reduction was observed in exposed cells. Severity of leaf Rust and Brown Eyespot in Genotypes of Coffea arabica *** with high Plant Density

This study investigated the severity of leaf rust and brown eyespot in genotypes of Coffea arabica l. cultivated with high plant density in the region of Caparaó-ES. The experiment was conducted in a competition field, cultivated with high plant density (8333 plants per hectare), following a randomized block design, with 16 genotypes and four replications. The plants were evaluated during consecutive harvests to study two complete reproductive cycles (from 2010 to 2012). Data were obtained for the phenologicalstages of flowering, graining, maturation and vegetative rest of each cycle. The severity of leaf rust (hemileia vastratrix) and brown eyespot (Cercospora coffeicola) was evaluated using descriptive scales. It was observed that the genotypes are able to keep a considerable level of resistance to the leaf rust and brown eyespot when cultivated with increased density. The genotypes presented variability regarding the severity of the leaf rust and brown eyespot, indicating the existence of differential levels of resistance between them. For cultivation with high plant density, the genotypes Katipó, Paraíso MG h419-1, h419-3-3-7-16-4-1-1, Araponga MG1, Catucaí Amarelo 24/137, Catiguá MG2, Sacramento MG1, Pau-Brasil MG1, Catiguá MG3, Oeiras MG 6851 and Tupi present higher level of resistance for leaf rust. In addition, the genotypes Paraíso MG h419-1, Catiguá MG2, Pau-Brasil MG1, Catiguá MG3, Oeiras MG 6851, Tupi, Catuaí IAC 44, Catuaí IAC 81 and Catuaí IAC 144 present higher level of resistance for brown eyespot.

A Pharmacist-led Medication Use Evaluation of Proton-Pump Inhibitors

Objective： To determine whether patients maintain symptom control after discontinuation of PPI （proton-pump inhibitor） therapy through intervention of a pharmacist-led medication utilization program and determine annual clinic cost-savings per prescription. Design： The study was conducted as a prospective, medication use evaluation. Using an electronic medical record system at a clinic, reports generated all patients receiving PP1 therapy from January 2013-June 2015. Patients were encouraged to participate in a PPI discontinuation trial through a pharmacist-led educational session. Participants were followed-up three months post enrollment to assess their quality of life through a survey assessing severity and frequency of symptoms. Participants unable to maintain symptom control after the intervention were referred back to their primary care physician for further evaluation. Results： Of one hundred participants, 25% （n = 25） were able to discontinue the use of PPI by lifestyle modifications, 43% （n = 43） refused to discontinue PP1 therapy and lifestyle changes due to the severity of their symptoms, 17% （n = 17） switched to over-the-counter h2 receptor antagonist daily to control their symptoms, 9% （n = 9） used PPIs only as needed, and 6% （n = 6） of participants were dropped from the study after three failed communication attempts. The clinic saved approximately $11 thousand annually for every one prescription of PPI＇s. Conclusions： Discontinuation of PPI or step-down therapy was possible for patients with mild-moderate GERD （Gastroesophageal Reflux Disease） symptoms when mediated by pharmacist counseling and follow-up. Also, the annual PPI expenditure at this clinic will decrease due to participant＇s discontinuation of therapy.

One of the clear signals of the ongoing climate change is sea level rise (SlR). Normal oceanic tides superimposed on a rising sea level and coastal flooding will affect many coastal communities. An international collaboration among Brazil, the United Kingdom, and the United States was designed to evaluate local decision making processes and to open space for local urban managers to reflect on possible actions toward adaption to sea level rise given the historical constraints imposed by administrative and institutional structures. This project focused on the processes that shape adaptation of three coastal communities in three countries. It worked jointly with these communities in defining the problem, examining risks, and understanding the benefits and obstacles that may hinder implementation of adaptation options. The framework was co-designed by an interdisciplinary team that incorporated social and natural scientists from the three countries, including local government officials. The study addressed 1) evaluation of adaptive capacity through participant surveys and 2) physical and cost impact simulations using geospatial models of the built infrastructure and implementation of adaptation options under different hazard scenarios, including 50 and 100-year sea level rise projections and severe storms. Based on the surveys’ results, there is a clear sense of the awareness of each community of the risk of floods due to intense storms, and of the usefulness of engaging early in a process that promotes the understanding of risks, impacts, and costs. A majority of workshop participants prioritized pursuing physical and green infrastructure actions now or within coming years or decades. A positive common aspect of the three sites was the commitment shown by the stakeholders in taking part in the process and evaluating which adaptation measures could be more effective in their cities. The paricá [Schizolobium amazonicum huber ex Ducke] is a native species of the Amazon region, belonging to the Fabaceae family and is a legume that has great social and economic importance, raw material for the manufacture of wood panels. heliophilous, with wood of light density and good workability. Amazonian soils are highly weathered and characterized by high acidity, so Al harms the growth and development of sensitive plants, as well as the presence of resistant and/or tolerant plants. Thus, in this work, we sought to study the biochemical metabolism alterations generated in young plants of Schizolobium amazonicum huber ex Ducke affected at different dosages of AlCl3. The research was conducted in a greenhouse of the Federal Rural University of Amazonia, using pre-scarified paricá seeds, which were seeded in plastic containers of 3.5 l capacity, with black soil fertilized with NPK 15-15-20. On the 8th day after sowing (DAS), thinning of 5 seedlings was performed for 2, and on day 12 of 2 for 1 seedling. The treatments started after 1 month and 22 days of sowing and the plants were collected 24 days later. A completely randomized experimental design (RED) was used, with 6 treatments (Control, Al 15 mg/l-1, Al 30 mg/l-1, Al 45 mg/l-1, Al 60 mg/l-1 and Al 75 mg/l-1) with 5 replicates each (one plant/pot) totaling 30 experimental units. The AlCl3 dosages resulted in considerable changes in the biochemical variables evaluated, especially when the 75 mg·l-1 dosage was applied. The considerable reduction in some variables such as nitrate reductase, glycine betaine and total soluble carbohydrates of these compounds to the applied AlCl3·6h2O dosages, is a way to create resistance mechanisms to try to overcome stress or as a way of showing limitations in the applied dosages. Background:The development and spread of artemisinin-resistant Plasmodium falciparum malaria in Greater Mekong Subregion has created impetus for continuing global monitoring of efficacy of artemisinin-based combination therapies(ACTs). This post analyses is aimed to evaluate changes in early treatment response markers 10 years after the adoption of ACTs as first-line treatments of uncomplicated falciparum malaria in ***: At 14 sentinel sites in six geographical areas of Nigeria, we evaluated treatment responses in 1341 children under 5 years and in additional 360 children under 16 years with uncomplicated malaria enrolled in randomized trials of artemether-lumefantrine versus artesunate-amodiaquine at 5-year interval in 2009-2010 and 2014-2015 and at 2-year interval in 2009-2010 and 2012-2015, respectively after deployment in ***: Asexual parasite positivity 1 day after treatment initiation (APPD1) rose from 54 to 62% and 2 days after treatment initiation from 5 to 26% in 2009-2010 to 2014-2015(P=0.002 and Ply).Parasite clearance time increased significantly from 1.6 days (95% confidence interval [CI]: 1.55-1.64) to 1.9 days (95% C,1.9-2.0) and geometric mean parasite reduction ratio 2 days after treatment initiation decreased significantly from 11 000 to 4700 within the same time period (Ph). Enrolment parasitaemia > 75 000 μl^-1, haematocrit > 27% 1 day post-treatment initiatiortreatment with artemether-lumefantrine and enrolment in 2014-2015 independently predicted APPD1. In paralle , Kaplan-Meier estimated risk of recurrent infections by day 28 rose from 8 to 14% (P=0005) and from 9 to 15%(P=0.02) with artemether-lumefantrine and artesunate-amodiaquine, respectively. 背景本研究中，我们探讨了微创髋关节成形术后创口持续输注0．3％罗哌卡因对疼痛和吗啡用量的影响。方法在这项双盲研究中，前瞻性纳入连续67例在蛛网膜下隙麻醉下择期行微创髋关节成形术的患者。外科医师将15cm长的有孔导管放入关节囊外。患者随机分为两组：在关闭切口前，创口内单次注入20ml的0．3％罗哌卡因（R-组）或0．9％生理盐水（P-组）。随后用弹性泵以8ml／h的速度分别持续泵入0．3％的罗哌卡因或者安慰剂至术后48小时。所有患者均给予静脉吗啡自控镇痛。记录48小时内患者吗啡用量、静息和运动时疼痛以及血浆罗哌卡因总浓度和游离浓度。术后随访3个月。结果两组-般资料和手术数据相似。术后第一个48小时期间，R-组平均吗啡用量显著低于P组：45．4±9．5VS69．7±9．6（P〈0．001）。R-组吗啡用量在术后第一个24小时期间平均减少14．4mg[95％可信区间（confidenceinterval，CI）12．6～16．1]，在术后第二个24小时期间平均减少20．8mg（95％CI为19．1～22．4）。R组患者静息和运动时疼痛评分均较低（P〈0．0001）。R-组患者平均满意度较基线增加了22．7％（95％CI为15．9～16．1）。R-组血浆罗哌卡因总浓度和游离浓度均低于中毒浓度。R-组在第24小时和第48小时时血浆罗哌卡因游离浓度分别是0．141μmol／ml和0．11μmol／ml。在术后3个月时，两组臀部疼痛和镇痛药用量相似，但是R．组患者创口接触（31．2；95％CI27．7～34．7）不适感和压迫不适感（24；95％CI为20．1-27．9）明显减轻（P〈0．0001）。结论微创髋关节置换术后关节囊外创口持续输注0．3％罗哌卡因是-种减少吗啡用量，改善术后镇痛质量的有效方法，术后3个月时该方法依然具有-定优势。

This paper is the second in a series focused on providing a stable platform for the taxonomy of phytopathogenic *** focuses on 25 phytopathogenic genera:Alternaria,Bipolaris,Boeremia,Botryosphaeria,Calonectria,Coniella,Corticiaceae,Curvularia,Elsinoe,Entyloma,Erythricium,Fomitiporia,Fulviformes,laetisaria,limonomyces,Neofabraea,Neofusicoccum,Phaeoacremonium,Phellinotus,Phyllosticta,Plenodomus,Pseudopyricularia,Tilletia,Venturia and Waitea,using recent molecular data,up to date names and the latest taxonomic *** each genus a taxonomic background,diversity aspects,species identification and classification based on molecular phylogeny and recommended genetic markers are *** this study,varieties of the genus Boeremia have been elevated to species ***,Bipolaris,Curvularia,Neofusicoccum and Phyllosticta that were included in the One Stop Shop 1 paper are provided with updated entries,as many new species have been introduced to these genera.

The cryptic lifestyle of most fungi necessitates molecular identification of the guild in environmental *** the past decades,rapid development and affordability of molecular tools have tremendously improved insights of the fungal diversity in all ecosystems and ***,in spite of the progress of molecular methods,knowledge about functional properties of the fungal taxa is vague and interpretation of environmental studies in an ecologically meaningful manner remains *** order to facilitate functional assignments and ecological interpretation of environmental studies we introduce a user friendly traits and character database FungalTraits operating at genus and species hypothesis *** the information from previous efforts such as FUNGuild and FunFun together with involvement of expert knowledge,we reannotated 10,210 and 151 fungal and Stramenopila genera,*** resulted in a stand-alone spreadsheet dataset covering 17 lifestyle related traits of fungal and Stramenopila genera,designed for rapid functional assignments of environmental *** order to assign the trait states to fungal species hypotheses,the scientific community of experts manually categorised and assigned available trait information to 697,413 fungal ITS *** the basis of those sequences we were able to summarise trait and host information into 92,623 fungal species hypotheses at 1%dissimilarity threshold.
