Root hairs(RHs)grow in a polar manner as cylindrical protrusions from the root epidermis.They significantly expand the root volume in the rhizosphere to search for water and nutrients,anchor the plant,and promote interactions with several kinds of microorganisms in the soil.Trichoblasts,the epidermis cell type responsible for RH development,switch from anisotropic cell elongation to tip growth through polarization of the growth machinery to a predefined RH initiation domain(RHID)at the plasma membrane.

Root hairs(RHs)emerge as cylindrical cell protrusions from the root epidermis in a polar manner,and these outgrows increase the rhizosphere space to acquire water and nutrients,anchor the plant,and interact with various soil microorganisms.Auxin is one of the key hormones to promote RH growth,and high levels of auxin are directly promoted by low levels of macronutrients in the soil,such as phosphate(Bhosale et al.,2018)and nitrates(Jia et al.,2023).The nutritional signals for roots,which rapidly change in both time and place within the soil,are tightly linked to signaling pathways that execute swifts on cellular processes to adjust to a challenging environment.Plant rapid alkalinization factors(RALFs)are peptides that are released outside the cell and act as peptide-hormone signals.They attach to the extracellular domains of members of the Catharanthus roseus RECEPTOR-LIKE KINASE 1-LIKE family,specifically FERONIA(FER)in conjunction with its co-receptor LORELEI-LIKE-GPIANCHORED PROTEIN 1.

Plant long noncoding RNAs(lncRNAs)have emerged as important regulators of chromatin dynamics,impacting on transcriptional programs leading to different developmental outputs.The lncRNA AUXIN-REGULATED PROmOTER LOOP(APOLO)directly recognizes multiple independent loci across the Arabidopsis genome and modulates their three-dimensional chromatin conformation,leading to transcriptional shifts.Here,we show that APOLO recognizes the locus encoding the root hair(RH)master regulator ROOT HAIR DEFECTIVE 6(RHD6)and controls RHD6 transcriptional activity,leading to cold-enhanced RH elongation through the consequent activation of the transcription factor gene RHD6-like RSL4.Furthermore,we demonstrate that APOLO interacts with the transcription factor WRKY42 and modulates its binding to the RHD6 promoter.WRKY42 is required for the activation of RHD6 by low temperatures and WRKY42 deregulation impairs cold-induced RH expansion.Collectively,our results indicate that a novel ribonucleoprotein complex with APOLO and WRKY42 forms a regulatory hub to activate RHD6 by shaping its epigenetic environment and integrate signals governing RH growth and development.

Extensins （EXTs） are highly repetitive plant O-glycoproteins that require several post-translational modifi- cations （PTms） to become functional in plant cell walls. First, they are hydroxylated on contiguous proline residues; then they are O-glycosylated on hydroxyproline and serine. After secretion into the apoplast, O-glycosylated EXTs form a tridimensional network organized by inter- and intra-Tyr linkages. Recent studies have made significant progress in the identification of the enzymatic machinery required to process EXTs, which includes prolyl 4-hydroxylases, glycosyltransferases, papain-type cysteine endopeptidases, and peroxidases. EXTs are abundant in plant tissues and are particularly important in rapidly expanding root hairs and pollen tubes, which grow in a polar manner. Small changes in EXT PTms affect fastgrowing cells, although the molecular mechanisms underlying this regulation are unknown. In this review, we highlight recent advances in our understanding of EXT modifications throughout the secretory pathway, EXT assembly in cell walls, and possible sensing mechanisms involving the Catharanthus roseus cell surface sensor receptor-like kinases located at the interface between the apoplast and the cytoplasmic side of the plasma membrane.

Implicit Continuous User Authentication for mobile Devices based on Deep Reinforcement Learning

The predominant method for smart phone accessing is conﬁned to methods directing the authentication by means of Point-of-Entry that heavily depend on physiological biometrics like,ﬁngerprint or face.Implicit continuous authentication initiating to be loftier to conventional authentication mechanisms by continuously conﬁrming users’identities on continuing basis and mark the instant at which an illegitimate hacker grasps dominance of the session.However,divergent issues remain unaddressed.This research aims to investigate the power of Deep Reinforcement Learning technique to implicit continuous authentication for mobile devices using a method called,Gaussian Weighted Cauchy Kriging-based Continuous Czekanowski’s(GWCK-CC).First,a Gaussian Weighted Non-local mean Filter Preprocessing model is applied for reducing the noise pre-sent in the raw input face images.Cauchy Kriging Regression function is employed to reduce the dimensionality.Finally,Continuous Czekanowski’s Clas-siﬁcation is utilized for proﬁcient classiﬁcation between the genuine user and attacker.By this way,the proposed GWCK-CC method achieves accurate authen-tication with minimum error rate and time.Experimental assessment of the pro-posed GWCK-CC method and existing methods are carried out with different factors by using UmDAA-02 Face Dataset.The results conﬁrm that the proposed GWCK-CC method enhances authentication accuracy,by 9%,reduces the authen-tication time,and error rate by 44%,and 43%as compared to the existing methods.

Precision-cut tissue slices are an important in vitro system to study organ function because they preserve most of the native cellular microenvironments of organs,including complex intercellular connections.However,during sample manipulation or slicing,some of the natural surface topology and structure of these tissues is lost or damaged.Here,we introduce a microfluidic platform to perform multiple assays on the surface of a tissue section,unhindered by surface topography.The device consists of a valve on one side and eight open microchannels located on the opposite side,with the tissue section sandwiched between these two structures.When the valve is actuated,eight independent microfluidic channels are formed over a tissue section.This strategy prevents cross-contamination when performing assays and enables parallelization.Using irregular tissues such as an aorta,we conducted multiple in vitro and ex vivo assays on tissue sections,including short-term culturing,a drug toxicity assay,a fluorescence immunohistochemistry staining assay,and an immune cell assay,in which we observed the interaction of neutrophils with lipopolysaccharide(LPS)-stimulated endothelium.Our microfluidic platform can be employed in other disciplines,such as tissue physiology and pathophysiology,morphogenesis,drug toxicity and efficiency,metabolism studies,and diagnostics,enabling the conduction of several assays with a single biopsy sample.

In their recent article in Nature,moiseeva et al.reported that senescent cells are integral components of the skeletal muscle regenerative niche,showing that they repress regeneration in response to different types of injuries in young mice and that this regeneration is even more profound in aged mice.Their observations point to selective links between the conservation of cell identity traits and two universal senescence hallmarks(i.e.,inflammation and matrix remodeling/fibrosis)across cell types,regeneration times,and aging statuses.

Aim:To describe findings when comparing lympho-SPECT-CT images before and after lymphovenous anastomosis(LVA)surgeries and to correlate these results with pre-and post-operative volume changes in the limbs of patients.methods:An observational,prospective,longitudinal study was designed.20 consecutive patients were treated for lymphedema by means of LVA between 2015 and 2018.All were affected by secondary lymphedema(ISGⅡ-Ⅲ)following lymphadenectomy,radiation or both.All patients received preoperative rehabilitation as well as radiotherapy after oncological surgery.Limb volume was measured before surgery and at one year later.LVA was performed under general anesthesia with ICG guidance.ICG was also used to evaluate postoperative outcomes.Lympho-SPECT-CT was performed in all subjects at their first consultation and at one year after every surgical intervention.Description of findings included an absence of lymph nodes,new lymph node activity in anatomical areas and new lymphatic activity in extra-anatomical areas.Results:Limb volume decreased in 19 patients after LVA.Six patients showed preoperative linear ICG patterns,combined with areas presenting with another type of pattern.After LVA,the linear pattern was observed in 11 patients.SPECT-CT/lymphoscintigraphy before surgery showed a total absence of lymph nodes,except in two cases,in whom small nodes in anatomical locations were described.After LVA,we observed new landmarks in 16 patients corresponding to lymphatic circulation that was not present in preoperative studies.In six cases,newlymphatic activity compatible with lymph nodes was detectable after LVA.The Spearman correlation coefficient was negative when circumferences and lympho-SPECT-CT were tested(P=0.02).Conclusion:Results showed a postoperative decrease in volume that correlated inversely with lympho-SPECT/CT findings.Lympho-SPECT/CT provided additional information related to accurate identification and the anatomical location of lymphatic structures that were not observed before reconstructive surgery

As photovoltaic energy increasingly penetrates in power systems,transmission system operators have started to request its participation in providing ancillary services.One of the demanded services is the power ramp-rate control(PRRC),which attempts to limit the power ramps produced by intermittent irradiance conditions.In order to achieve the desired objective,solutions based on storage systems or modifying the maximum power point tracking(mPPT)in perturb and observe(P&O)algorithms are commonly adopted.The starting point in PRRC is the determination of the instantaneous power ramprate,and different methods have been proposed in the literature for its calculation.However,the accuracy and computational speed of existing procedures can be improved,which may be critical in situations with rapid irradiance fluctuations.In this paper,a decoupled photovoltaic power ramp-rate calculation method is presented,in which the effect of variable irradiance and the P&O algorithm are computed separately.The proposed method has been theoretically demonstrated and tested through simulation and experimental tests.Simulation results show that it can improve the previous methods in terms of accuracy and computation time.Experimental validation with hardware-inthe-loop demonstrates the suitability of the proposed method for real-time applications,even in presence of noisy measurements.
