Irbesartan (IBS) is a tetrazole derivative and antihypertensive drug that has two interconvertible structures, 1H- and 2H-tautomers. The difference between them lies in the protonation of the tetrazole ring. In the solid-state, both tautomers can be isolated as crystal forms A (1H-tautomer) and B (2H-tautomer). Studies have reported that IBS is a polymorphic system and its forms A and B are related monotropically. These reports indicate form B as the most stable and less soluble form. Therefore, the goal of this contribution is to demonstrate through a complete solid-state characterization, thermodynamic study and dissolution properties that the IBS forms are desmotropes that are not related monotropically. However, the intention is also to call attention to the importance of conducting strict chemical and in solid-state quality controls on the IBS raw materials. Hence, powder X-ray diffraction (PXRD) and Raman spectroscopy (RS) at ambient and non-ambient conditions, differential scanning calorimetry (DSC), hot stage microscopy (HSm), Fourier transform infrared (FT-IR) and scanning electron microscopy (SEm) techniques were applied. Furthermore, intrinsic dissolution rate (IDR) and structural stability studies at 98% relative humidity (RH), 25℃ and 40 ℃ were conducted as well. The results show that in fact, form A is approximately four-fold more soluble than form B. In addition, both IBS forms are stable at ambient conditions. Nevertheless, structural and/or chemical instability was observed in form B at 40℃ and 98% RH. IBS has been confirmed as a desmotropic system rather than a polymorphic one. Consequently, forms A and B are not related monotropically.

AIm: To evaluate the technical failures of the Bravo pH test in a population with nonerosive gastroesophageal reflux disease. mETHODS: Over the course of a year, we prospectively studied a population of 66 nonerosive reflux disease patients who received a Bravo pH test. The number and frequency of all technical failures were documented, quantified and analyzed. RESULTS: A total of 66 patients, with a mean age of 41.7 years, were studied. Technical failures occurred in 15.15% of the sample. The most frequent failures were due to poor data reception (4.5%), early dislodgement (4.5%) and capsule removal (6.1%). CONCLUSION: The Bravo capsule pH test involves a low but non-negligible rate of technical problems, a fact that must always be considered by physicians.

Capillary hydrodynamic fractionation（CHDF） with turbidity detection at a single wavelength is an analytical technique that is often used for sizing the sub-micrometric particles of hydrophobic *** article investigates three sources of errors that affect the particle size distribution（PSD） estimated by CHDF：diameter calibration errors,uncertainties in the particle refractive index（PRI）,and instrumental broadening（IB）.The study is based on simulated and experimental examples that involve unimodal and bimodal *** errors in the diameter calibration curve can produce important deviations in the number average diameter due to systematic shifts suffered by the PSD *** uncertainties in the PRI are unimportant in the analysis of unimodal PSDs,but in the specific case of bimodal PSDs,errors in the PRI can strongly affect the estimated number concentration of each *** typical IB correction（based on the IB function estimated from narrow standards） produces slightly erroneous average diameters but can lead to PSDs with underestimated widths and distorted *** practice,the three investigated sources of errors can be present simultaneously,and uncertainties in the average diameters,the shape and width of the PSD,and the number concentration of the PSD m.des are unavoidable.
