Association between the CYBA and NOX4 genes of nadPH oxidase and its relationship with metabolic syndrome in non-alcoholic fatty liver disease in Brazilian population

Background: Oxidative stress has been implicated in the progression of severe forms of non-alcoholic fatty liver disease(naFld). nadPH oxidase produces reactive oxygen species. In the present study, we investigated for the first time two single nucleotide polymorphisms(SNPs) in the regulatory region of genes encoding nadPH oxidase 4(NOX4) and p22 phox(CYBA) in ***: A total of 207 biopsy-proven naFld patients [simple steatosis(n = 27); nonalcoholic steatohepatitis(naSH)(n = 180)] were evaluated. Genomic dna was extracted from peripheral blood cells, and polymorphisms in CYBA(unregistered) and NOX4(rs3017887) were determined by direct sequencing of ***: Associations of CYBA-675 T/A with high-density lipoprotein(Hdl)(TT vs TA vs AA; P d triglycerides(TGl)(TT vs XA; P d only in naSH patients. For polymorphisms in the NOX4 gene, NOX4(rs3017887) CA + AA genotypes was significant associated with alanine aminotransferase(AlT)(CA + AA vs CC; P = 0.02). However, there was no association of SNPs in the CYBA and NOX4 genes encoding the nadPH oxidase system proteins and the presence of naSH. Regarding the clinical results, it was observed that the most advanced degrees of fibrosis occurred in patients diagnosed with type 2 diabetes mellitus(66.9% vs 37.5%, P d those who were more obese(32.2 vs 29.0 kg/m2,P ddition, serum glucose and insulin levels increased significantly in the presence of ***: There were associations between the presence of the allele A in the NOX4 SNP and a higher concentration of AlT in the naFld population; between the presence of the AA genotype in the polymorphism of the CYBA-675 T/A CYBA gene and a higher level of TGl and lower Hdl in naSH patients. The presence of metabolic syndrome was associated with advanced degrees of fibrosis in naFld patients.

With the introduction of antiretroviral therapy (ART),a dramatic reduction in HIV-related morbidity and mortality has been ***,it is now becoming increasingly clear that liver-related complications,particularly rapid fibrosis development from ART as well as from the chronic HIV infection itself,are of serious concern to HIV *** pathophysiology of liver fibrosis in patients with HIV is a multifactorial process whereby persistent viral replication,and bacterial translocation lead to chronic immune activation and inflammation,which ART is unable to fully suppress,promoting production of fibrinogenic mediators and *** addition,mitochondrial toxicity,triggered by both ART and HIV,contributes to intrahepatic damage,which is even more severe in patients co-infected with viral *** recent years,new insights into the mechanisms of accelerated fibrosis and liver disease progression in HIV has been obtained,and these are detailed and discussed in this review.

Morphometric and kinematic sperm subpopulations in split ejaculates of normozoospermic men

This study was designed to analyze the sperm kinematic and morphometric subpopulations in the different fractions of the ejaculate in normozoospermic men. Ejaculates from eight normozoospermic men were collected by masturbation in three fractions after 3-5 days of sexual abstinence. Analyses of sperm motility by computer-assisted sperm analysis （CASA-Mot）, and of sperm morphometry by computer-assisted sperm morphometry analysis （CASA-Morph） using fluorescence were performed. Clustering and discriminant procedures were performed to identify sperm subpopulations in the kinematic and morphometric data obtained. Clustering procedures resulted in the classification of spermatozoa into three kinematic subpopulations （slow with low AlH [35.6% of all motile spermatozoa], with circular trajectories [32.0%], and rapid with high AlH [32.4%]）, and three morphometric subpopulations （large-round [33.9% of all spermatozoa], elongated [32.0%], and small [34.10%]）. The distribution of kinematic sperm subpopulations was different among ejaculate fractions （P 〈 0.001）, with higher percentages of spermatozoa exhibiting slow movements with low AlH in the second and third portions, and with a more homogeneous distribution of kinematic sperm subpopulations in the first portion. The distribution of morphometric sperm subpopulations was also different among ejaculate fractions （P〈 0.001）, with more elongated spermatozoa in the first, and of small spermatozoa in the third, portion. It is concluded that important variations in the distribution of kinematic and morphometric sperm subpopulations exist between ejaculate fractions, with possible functional implications.

Allophane as a nanotechnological Additive in Masonry Materials to Increase Resistance and Waterproofing

The constant need for high-strength materials in the construction industry promotes the research of additives that improve the properties of masonry materials. The use of allophane as an additive in concrete and mortar mixtures was implemented to improve their strength and waterproofing, respectively, using compression and water absorption tests according to their corresponding standards (ASTM C1231, ASTM d2938, and ASTM C1585). The samples were evaluated at different concentrations and curing ages. In addition, different sand/cement ratios were considered for the mortar. The results revealed that there was a 9.4% increase in compressive strength in concrete and a 23.7% reduction in water absorption in mortar for the 5:1 ratio. These changes would be the result of the interaction of the nanoporous additive in the atomic crystal structure of the material demonstrating the nanotechnological nature of allophane.

Black,pinto and white beans lower hepatic lipids in hamsters fed high fat diets by excretion of bile acids

Hypolipidemic and anti-obesity properties of extruded black,pinto and white beans were examined in male Syrian hamsters fed high fat diets for 3 *** efficiency and effectiveness of extruder processing to eliminate heatlabile antinutrients were also *** fed a high fat diet containing 40%extruded black beans gained the same amount of weight as animals on a low-fat chow diet(based on AIN-93G).Total plasma cholesterol of the hamsters fed bean flour-based diets tended to be lower,ranging between 298 and 356 mg/dl,compared to 365 mg/dl for the control group and total and free liver cholesterol was about 50 and 33%lower,respectively,in the livers(freeze-dried)of hamsters fed the black turtle bean(BB)and pinto bean(PB)*** nitrogen content of the feces from hamsters fed the PB and white bean(WB)diets were 2.6x higher than the control and the excretion of deoxycholic acid(dCA)and lithocholic acid(lCA)was higher in all legume fed hamsters The results show that extrusion efficiently and economically reduces anti-nutritive factors that inhibit the digestion and absorption of proteins and carbohydrates.

Counter-regulatory phosphatases TnaP and NPP1 temporally regulate tooth root cementogenesis

Cementum is critical for anchoring the insertion of periodontal ligament fibers to the tooth root. Several aspects of cementogenesis remain unclear, including differences between acellular cementum and cellular cementum, and between cementum and bone. Biomineralization is regulated by the ratio of inorganic phosphate （Pi） to mineral inhibitor pyrophosphate （PPi）, where local Pi and PPi concentrations are controlled by phosphatases including tissue-nonspecific alkaline phosphatase （TnaP） and ectonucleotide pyrophosphatase/phosphodiesterase 1 （NPP1）. The focus of this study was to define the roles of these phosphatases in cementogenesis. TnaP was associated with earliest cementoblasts near forming acellular and cellular cementum. With loss of TnaP in the Alpl null mouse, acellular cementum was inhibited, while cellular cementum production increased, albeit as hypomineralized cementoid. In contrast, NPP1 was detected in cementoblasts after acellular cementum formation, and at low levels around cellular cementum. loss of NPP1 in the Enppl null mouse increased acellular cementum, with little effect on cellular cementum. developmental patterns were recapitulated in a mouse model for acellular cementum regeneration, with early TnaP expression and later NPP1 expression. In vitro, cementoblasts expressed Alpl gene/protein early, whereas Enppl gene/protein expression was significantly induced only under mineralization conditions. These patterns were confirmed in human teeth, including widespread TnaP, and NPP1 restricted to cementoblasts lining acellular cementum. These studies suggest that early TnaP expression creates a low PPi environment promoting acellular cementum initiation, while later NPP1 expression increases PPi, restricting acellular cementum apposition. Alterations in PPi have little effect on cellular cementum formation, though matrix mineralization is affected.

Background Schistosomiasis is highly endemic in sub-Saharan Africa and frequently imported to Europe. Male urogenital manifestations are often neglected. We aimed to ascertain the prevalence of genitourinary clinical signs and symptoms among long-term African migrants in a non-endemic European country using a serology *** We carried out a prospective, community-based cross-sectional study of adult male migrants from sub-Saharan Africa living in *** serology tests and microscopic urine examinations were carried out, and clinical data were obtained from an electronic medical record search and a structured *** We included 388 adult males, mean age 43.5 years [Standard deviation (Sd) = 12.0, range: 18–76]. The median time since migration to the European Union was 17 [Interquartile range (IQR): 11–21] years. The most frequent country of origin was Senegal (N = 179, 46.1%). Of the 338, 147 (37.6%) tested positive forSchistosoma. Parasite eggs were present in the urine of only 1.3%. Nine genitourinary clinical items were significantly associated with positiveSchistosoma serology results: pelvic pain (45.2%;OR = 1.57, 95%CI: 1.0–2.4), pain on ejaculation (14.5%;OR = 1.85, 95%CI: 1.0–3.5), dyspareunia (12.4%;OR = 2.45, 95%CI: 1.2–5.2), erectile dysfunction (9.5%;OR = 3.10, 95%CI: 1.3–7.6), self-reported episodes of infertility (32.1%;OR = 1.69, 95%CI: 1.0–2.8), haematuria (55.2%;OR = 2.37, 95%CI: 1.5–3.6), dysuria (52.1%;OR = 2.01, 95%CI: 1.3–3.1), undiagnosed syndromic STIs (5.4%), and orchitis (20.7%;OR = 1.81, 95%CI: 1.0–3.1).
