New prospects in the diagnosis and treatment of immune-mediated inner ear disease

Autoimmune inner ear disease(AIED) represents a very fertile research field and the advancements in the understanding of this disease have a direct application not only in patients affected with this condition but also in other inner ear disorders that share the same injury mechanism, damage to the inner ear hair cells. AIED also presents many challenges that have still to be overcome. Firstly, access to the inner ear is limited, as many interventions such as biopsies can result in great irreversible damage. Secondly, there are no completely specific markers for AIED. Lack of a definitive diagnosis can result in the treatment of patients not affected with the disease and, therefore, no response. Finally, some patients become refractory to glucocorticoids and new therapies are needed. This review offers an overview of the animal models that have contributed to the understanding of AIED pathophysiology, the value of currently available diagnostic tests, and therapeutic options, with a special focus on new therapies for non responders or patients refractory to glucocorticoids. Among these new options for therapy, biological agents have been tested recently, whereas gene and stem celltherapy may have a role in the future. The intratympanic route of administration avoids the systemic side effects associated with currently used drugs, and may become a more frequent approach in the future.

Characterization of fungal pathogens associated with grapevine trunk diseases in Arkansas and Missouri

Grapevine trunk diseases are a major concern to the wine-and table-grape industries worldwide,limiting both vineyard longevity and productivity.Field surveys conducted throughout the grape-growing regions of Arkansas and Missouri revealed the presence of three economically important grapevine trunk diseases including,Botryosphaeria canker,Eutypa dieback and esca.Morphological studies along with multi-gene phylogenetical analyses confirmed the identification of 15 different fungal taxa associated with different vascular symptoms.These include Botryosphaeria dothidea,Diatrypella sp.,Diplodia seriata,Dothiorella americana,Eutypa lata,Eutypella vitis,Lasiodiplodia missouriana,Lasiodiplodia viticola,Neofusicoccum ribis,Neofusicoccum vitifusiforme,Pestalotiopsis sp.,Pestalotiopsis uvicola,Phaeomoniella chlamydospora,Phomopsis viticola,Schyzophyllum commune,and Togninia minima.All of these represent new records on grapevines in Arkansas and Missouri.Dothiorella americana,L.missouriana and L.viticola are described as new species,and both N.ribis and N.vitifusiforme are first reported as grapevine pathogens in North America.Koch’s postulates confirmed the pathogenicity of all fungal species except S.commune in the interspecific hybrids Vignoles,Chambourcin,Norton,and Traminette.Lasiodiplodia spp.,N.ribis,and P.viticola were the most virulent fungi,while D.americana,E.vitis and N.vitifusiforme were considered to be weak pathogens.This research highlights the importance that grapevine trunk diseases have on grapevine health in growing regions where,due to different climatological conditions,interspecific hybrid cultivars are predominantly grown.

In their recent article in Nature,Moiseeva et al.reported that senescent cells are integral components of the skeletal muscle regenerative niche,showing that they repress regeneration in response to different types of injuries in young mice and that this regeneration is even more profound in aged mice.Their observations point to selective links between the conservation of cell identity traits and two universal senescence hallmarks(i.e.,inflammation and matrix remodeling/fibrosis)across cell types,regeneration times,and aging statuses.

Simulation, Power Quality Analysis and Improvements in a Hot Rolling Mill Using a STATCOM

This paper presents an analysis of the electric power flows in a hot rolling mill plant using a power quality analyzer and developing simulation models with Matlab-Simulink. The model inputs have been taken from process data by the process computer of the plant. Data from different rolling campaigns have been analysed. A STATCOM (static synchronous compensator) with a control strategy based on voltage oriented control with d-q coordinates is used as solution to improve the reactive compensation as well as to minimize the voltage fluctuations and sags effect. The overall electric model has been simulated with the STATCOM to check the improvement in the voltage stability and the consumption.

Microglia are the resident macrophages of the brain,originally described by Pio del Rio-Hortega(a student of Santiago ramon y Cajal)in a series of studies in 1919.Since those pioneering studies,many others have followed to describe microglia as complex and multitasking cells with many diverse roles under physiological(e.***.their key role in synapse pruning during development)or diseased conditions.Microglia exist as sentinels or surveyors of the environment that surround neurons,becoming reactive upon a wide array of stimuli and consequently developing an appropriate inflammatory response.In fact,a neuroinflammatory response driven by microglia is found in virtually every disease process that occurs within the central nervous system.Therefore,a better understanding of the mechanisms governing the microglia response is the key to improving the outcome of neurodegenerative conditions.

This paper presents an application of the simulation of discrete events (SED) using ARENATM in the management of large-scale breeding farms. The main objective of the simulation model is to find a policy of replacement, to ensure the best economic performance of a farm. The only variant analyzed of replacement policy was the number of cycles set in permanency for a sow in the herd. Considered incomes come from the sale of piglets and unproductive sows, and costs are due to the feeding of animals, replacement sows purchases, and the operation expenses of the farm. For this analysis, the production process was divided into three major stages called: mating, pregnancy or gestation and lactation. The sow’s movement from one stage to other was modeled by cycle-dependent transition probabilities. Considering the daily utility, as response variable, the model shows the best number of cycles to maintain the sows.

OBJECTIVES Elderly patients show a higher incidence of ischemic and bleeding events after percutaneous transluminal coron-ary intervention(PCI).We sought to investigate outcomes in elderly patients treated with antithrombotic strategy guided by bleeding and ischemic risks after revascularization with last generation everolimus-eluting stent(EES).METHODS Prospective multicenter registry including patients over 75 years revascularized with EES and antithrombotic ther-apy guided by clinical presentation,PCI complexity and PRECISE DAPT score.Co-primary safety endpoints were:(1)composite of cardiac death,myocardial infarction and stent thrombosis and;(2)bleeding(BARC 2-5).Primary efficacy endpoint was target lesion revascularization.A matched group of patients revascularized with current drug-eluting stents and no such tailored antith-rombotic therapy was used as control.RESULTS Finally,1064 patients were included in SIERRA-75 cohort,80.8±4.2 years,36.6%women,71%acute coronary syn-dromes(ACS)and 53.6%complex PCI.Co-primary safety endpoint of major adverse cardiovascular events was met in 6.2%,co-primary safety endpoint of bleeding in 7.8%and primary efficacy endpoint of TKLR in 1.5%.The multivariable adjusted model showed no significant association of the prescribed short/long dual antiplatelet therapy(DAPT)durations with any endpoint suggesting a well tailored therapy.No stent thrombosis reported in the subgroup with 1-3 months DAPT duration.As compared to control group,bleeding BARC 2-5 was significantly lower in SIERRA-75 group(7.4%vs.10.2%,P=0.04)as well as the net safety-efficacy endpoint(14.3%vs.18.5%,P=0.02).CONCLUSIONS In elderly population,the application of this risks-adjusted antithrombotic protocol after revascularization with last generation EES seems to be associated with an improved prognosis in terms of ischemic and bleeding outcomes.

The global fuel management problem in BWRs(Boiling Water Reactors) can be understood as a very complex optimization problem,where the variables represent design decisions and the quality assessment of each solution is done through a complex and computational expensive simulation.This last aspect is the major impediment to perform an extensive exploration of the design space,mainly due to the time lost evaluating non promising solutions.In this work,we show how we can train a Multi-Layer Perceptron(MLP) to predict the reactor behavior for a given configuration.The trained MLP is able to evaluate the configurations immediately,thus allowing performing an exhaustive evaluation of the possible configurations derived from a stock of fuel lattices,fuel reload patterns and control rods patterns.For our particular problem,the number of configurations is approximately 7.7×10^(10);the evaluation with the core simulator would need above 200 years,while only 100hours were required with our approach to discern between bad and good configurations.The later were then evaluated by the simulator and we confirm the MLP usefulness.The good core configurations reached the energy requirements,satisfied the safety parameter constrains and they could reduce uranium enrichment costs.
