One-year survival in recipients older than 50 bridged to heart transplant with Impella 5.5 via axillary approach

BACKGROUND Optimizing patients with advanced heart failure before orthotopic heart transplantation(OHT), especially in patients greater than 50 years old, is imperative to achieving successful post-transplant outcomes. Complications are well-described for patients bridged to transplant(BTT) with durable left ventricular assist device(LVAD) support. Given the lack of data available in older recipients after the recent increase in mechanical support use, we felt it crucial to report our center’s one-year outcomes in older recipients after heart transplantation with percutaneously placed Impella 5.5 as a *** Forty-nine OHT patients were supported with the Impella 5.5 intended as a bridge between December 2019 and October 2022 at mayo Clinic in Florida. Data were extracted from the electronic health record at baseline and during their transplant episode of care after Institutional Review Boards approval as exempt for retrospective data *** Thirty-eight patients aged 50 or older were supported with Impella 5.5 as BTT. Ten patients underwent heart and kidney transplantation within this cohort. The median age at OHT was 63(58–68) years, with 32 male(84%) and six female patients(16%). Etiology was divided into ischemic(63%) and non-ischemic cardiomyopathy(37%). The baseline median ejection fraction was 19%(15–24). most patients were in blood group O(60%), and 50% were diabetic. The average duration of support was 27 days(range 6–94). The median duration of follow-up is 488 days(185–693). For patients that have reached the 1-year follow-up timeframe(22 of 38, 58%), the 1-year post-transplant survival is 95%.CONCLUSION Our single-center data provides awareness for using the Impella 5.5 percutaneously placed axillary support device in older heart failure patients in cardiogenic shock as a bridge to transplantation. One-year survival outcomes after heart transplantation are excellent despite the older recipient’s age and prolonged pre-transplant support.

Alternative splicing enhances transcriptome complexity in desiccating seeds

Before being dispersed in the environment, mature seeds need to be dehydrated. The survival of seeds after dispersal depends on their low hydration in combination with high desiccation tolerance. These characteristics are estab- lished during seed maturation. Some key seed maturation genes have been reported to be regulated by alternative splicing （AS）. However, so far AS was described only for single genes and a comprehensive analysis of AS during seed maturation has been lacking. We investigated gene expression and AS during Arabidopsis thaliana seed development at a global level, before and after desiccation. Bioinformatics tools were developed to identify differentially spliced regions within genes. Our data suggest the importance and shows the peculiar features of AS during seed desiccation. We identified AS in 347o of genes that are expressed at both timepoints before and after desiccation. most of these AS transcript variants had not been found before in other tissues. Among the AS genes some seed master regulators could be found. Interestingly, 6% of all expressed transcripts were not transcriptionally regulated during desiccation, but only modified by AS. We propose that AS should be more routinely taken into account in the analysis of transcriptomic data to prevent overlooking potentially important regulators.

An Invasive Succulent Plant (Kalanchoe daigremontiana) Influences Soil Carbon and Nitrogen mineralization in a Neotropical Semiarid Zone

It has long been recognized that plant invasions may alter carbon （C） and nitrogen （N） cycles, but the direction and magnitude of such alterations have been rarely quantified. In this study, we quantified the effects caused by the invasion of a noxious exotic plant, Kalanchoe daigrernontiana （Crassulaceae）, on C and N mineralization and enzymatic and microbial activities in the soil of a semiarid locality in Venezuela. We compared soil parameters associated with these processes （C and N mineralization time and the cumulative values, fluorescein diacetate hydrolytic activity, and activities of dehydrogenase, β-glucosidase, glucosaminidase, and urease） between invaded and adjacent non-invaded sites. In addition, correlations among these parameters and the soil physical-chemical properties were also examined to determine if a positive feedback exists between nutrient availability and K. daigremontiana invasion. Overall, our results showed that C mineralization and transformation of organic compounds to NH4^＋ were favored at sites colonized by K. daigrernontiana. With this species, we found the highest cumulative amounts of NH4^＋-N and C and the lowest mineralization time. These results could be explained by higher activities of urease and glueosaminidase in soils under the influence of K. daigremontiana. In addition, higher amounts of organic matter and moisture content in invaded soils might favor C and N mineralization. In conclusion, invasion of Neotropical semiarid zones by K. daigrernontiana may influence the chemical and biological properties of the soils covered by this species, increasing nutrient bioavailability, which, in time, can facilitate the invasion process.

This study was designed to analyze the sperm kinematic and morphometric subpopulations in the different fractions of the ejaculate in normozoospermic men. Ejaculates from eight normozoospermic men were collected by masturbation in three fractions after 3-5 days of sexual abstinence. Analyses of sperm motility by computer-assisted sperm analysis （CASA-mot）, and of sperm morphometry by computer-assisted sperm morphometry analysis （CASA-morph） using fluorescence were performed. Clustering and discriminant procedures were performed to identify sperm subpopulations in the kinematic and morphometric data obtained. Clustering procedures resulted in the classification of spermatozoa into three kinematic subpopulations （slow with low ALH [35.6% of all motile spermatozoa], with circular trajectories [32.0%], and rapid with high ALH [32.4%]）, and three morphometric subpopulations （large-round [33.9% of all spermatozoa], elongated [32.0%], and small [34.10%]）. The distribution of kinematic sperm subpopulations was different among ejaculate fractions （P 〈 0.001）, with higher percentages of spermatozoa exhibiting slow movements with low ALH in the second and third portions, and with a more homogeneous distribution of kinematic sperm subpopulations in the first portion. The distribution of morphometric sperm subpopulations was also different among ejaculate fractions （P〈 0.001）, with more elongated spermatozoa in the first, and of small spermatozoa in the third, portion. It is concluded that important variations in the distribution of kinematic and morphometric sperm subpopulations exist between ejaculate fractions, with possible functional implications.

molecular mechanisms of Ethylene-Auxin Interaction

During the century-long history of plant hormone research, the focus in this area has shifted from the original physi- ological experiments to molecular, genetic, biochemical, and, more recently, genomic approaches. During this jour- ney, we have learned about the many effects these natural compounds have on plant growth and development at the morphological and molecular levels. We have also uncov- ered how these molecules are sensed by the plant cell and how they trigger signaling cascades that relay information to the nucleus, ultimately culminating in a transcriptional cas- cade. With this fundamental knowledge at hand, the main efforts in the field have turned towards a more integrative systems approach, in which the function of a hormone is considered in the context of the complex and dynamic net of interactions with other hormones or, more broadly, with other developmental and environmental signals. Initially, due to the complexity of the problem, most crosstalk studies have focused on pairwise interactions between two or few hormones. Special attention has been given to the interac- tions between ethylene and auxin, perhaps in part because of the experimental tractability of some of the processes in which the interaction between these two hormones is most manifested. An excellent review has been recently written describing the many different processes in which these two hormones are known to interact, either synergistically, such as root growth inhibition, or antagonistically, as is the case for lateral root formation （muday et al., 2012）. Rather than following this process-centered approach, herein we decided to focus on the molecular mechanisms involved in the inter- action between ethylene and auxin. We take a closer look at the best-known transcriptional and posttranscriptional points of interaction, as well as revisit the most recent experimental findings that suggested additional levels of ethylene-auxin crosstalk.

Occult hepatitis B virus(HBV) infection(OBI) is characterized by the persistence of HBV DNA in the liver tissue in individuals negative for the HBV surface *** prevalence of OBI is quite variable depending on the level of endemic disease in different parts of the world,the different assays utilized in the studies,and the different populations *** studies have been carried out on OBI prevalence in different areas of the world and categories of *** studies show that OBI prevalence seems to be higher among subjects at high risk for HBV infection and with liver disease than among individuals at low risk of infection and without liver disease.

Simultaneous optimization of even flow and land and timber value in forest planning:a continuous approach

Background:Forest management planning involves deciding which silvicultural treatment should be applied to each stand and at what time to best meet the objectives established for the *** this,many mathematical formulations have been proposed,both within the linear and non-linear programming frameworks,in the latter case generally considering integer variables in a combinatorial *** present a novel approach for planning the management of forests comprising single-species,even-aged stands,using a continuous,multi-objective formulation(considering economic and even flow)which can be solved with gradient-type ***:The continuous formulation has proved robust in forest with different structures and different number of *** results obtained show a clear advantage of the gradient-type methods over heuristics to solve the problems,both in terms of computational time(efficiency)and in the solution obtained(effectiveness).Their improvement increases drastically with the dimension of the problem(number of stands).Conclusions:It is advisable to rigorously analyze the mathematical properties of the objective functions involved in forest management planning *** continuous bi-objective model proposed in this paper works with smooth enough functions and can be efficiently solved by using gradient-type *** advantages of the new methodology are summarized as:it does not require to set management prescriptions in advance,it avoids the division of the planning horizon into periods,and it provides better solutions than the traditional combinatorial ***,the graphical display of trade-off information allows an a posteriori articulation of preferences in an intuitive way,therefore being a very interesting tool for the decision-making process in forest planning.

Bioleaching of zinc from gold ores using Acidithiobacillus ferrooxidans

The present work investigated microbial leaching of zinc from ore using Acidithiobacillus ferrooxidans （ATCC 14859）. The ore samples, consisted of 13wt% zinc, were obtained from a gold mine in north-eastern Thailand. A shake flask study was performed on the ore samples using a rotary shaker under the following fixed conditions （250 r·min^-1, 30℃ for 16 d）. The influence of various conditions, namely medium type （with and without iron）, particle ore size （〈20, 20-40, 40-60, 60-100, and 〉100 mesh）, ore density （20 kg·m^-3, 50 kg·m^-3, and 100 kg·m^-3）, and pH of the medium （2, 2.5, 2.8, and 3）, were investigated. The microbial leaching was assessed by determining the concentration of zinc in the medium and compared with the initial sample concentration. The results show that Acidithiobacillus ferrooxidans can successfully leach zinc by as much as 6 times compared with the control experiment （without Acidithiobacillus ferrooxidans ferrooxidans）. The maximum efficiency （92.3%） for microbial leaching is obtained in iron-containing medium, 20-40 mesh ore sizes, 20 kg·m^-3 ore density at pH 2.8, and the zinc content is found in the medium at about 120 mg·m^-1.

Determining indoor air quality and identifying the origin of odour episodes in indoor environments

A methodology for identifying volatile organic compounds （VOCs） and determining air quality of indoor air has been developed. The air samples are collected using pump samplers by the inhabitants when they perceive odorous and/or discomfort episodes. Glass multi-sorbent tubes are connected to the pump samplers for the retention of VOC. The analysis is performed by automatic thermal desorption （ATD） coupled with gas chromatography-mass spectrometry （GC/mS）. This methodology can be applied in cases of sick building syndrome （SBS） evaluation, in which building occupants experience a series of varied symptoms that appear to be linked to time spent in the building. Chemical pollutants concentrations （e.g., VOC） have been described to contribute to SBS. To exemplify the methodology, a qualitative determination and an evaluation of existing VOC were performed in a dwelling where the occupants experienced the SBS symptoms. Higher total VOC （TVOC） levels were detected during episodes in indoor air （1.33 ±1.53 mg/m^3） compared to outdoor air （0.71± 0.46 mg/m^3）. The concentrations of individual VOCs, such as ethanol, acetone, isopropanol, 1-butanol, acetic acid, acetonitrile and 1-methoxy-2-propanol, were also higher than the expected for a standard dwelling. The external source of VOC was found to be an undeclared activity of storage and manipulation of solvents located at the bottom of a contiguous building.
