Metabolic and Phenotypic Responses of Greenhouse-Grown Maize Hybrids to Experimentally Controlled Drought Stress

Adaptation to abiotic stresses like drought is an important acquirement of agriculturally relevant crops like maize. Development of enhanced drought tolerance in crops grown in climatic zones where drought is a very dominant stress factor therefore plays an essential role in plant breeding. Previous studies demonstrated that corn yield potential and enhanced stress tolerance are associated traits. In this study, we analyzed six different maize hybrids for their ability to deal with drought stress in a greenhouse experiment. We were able to combine data from morphophysiological parameters measured under well-watered conditions and under water restriction with metabolic data from different organs. These different organs possessed distinct metabolite compositions, with the leaf blade displaying the most considerable metabolome changes following water deficiency. Whilst we could show a general increase in metabolite levels under drought stress, including changes in amino acids, sugars, sugar alcohols, and intermediates of the TCA cycle, these changes were not differential between maize hybrids that had previously been designated based on field trial data as either drought-tolerant or susceptible. The fact that data described here resulted from a greenhouse experiment with rather different growth conditions compared to natural ones in the field may explain why tolerance groups could not be confirmed in this study. We were, however, able to highlight several metabolites that displayed conserved responses to drought as well as metabolites whose levels correlated well with certain physiological traits.

Grain yield losses in yellow-rusted durum wheat estimated using digital and conventional parameters under field conditions

The biotrophic fungus Puccinia striiformis f. sp. tritici is the causal agent of the yellow rust in wheat. Between the years 2010–2013 a new strain of this pathogen(Warrior/Ambition),against which the present cultivated wheat varieties have no resistance, appeared and spread rapidly. It threatens cereal production in most of Europe. The search for sources of resistance to this strain is proposed as the most efficient and safe solution to ensure high grain production. This will be helped by the development of high performance and low cost techniques for field phenotyping. In this study we analyzed vegetation indices in the Red,Green, Blue(RGB) images of crop canopies under field conditions. We evaluated their accuracy in predicting grain yield and assessing disease severity in comparison to other field measurements including the Normalized Difference Vegetation Index(NDVI), leaf chlorophyll content, stomatal conductance, and canopy temperature. We also discuss yield components and agronomic parameters in relation to grain yield and disease ***-based indices proved to be accurate predictors of grain yield and grain yield losses associated with yellow rust(R2= 0.581 and R2= 0.536, respectively), far surpassing the predictive ability of NDVI(R2= 0.118 and R2= 0.128, respectively). In comparison to potential yield, we found the presence of disease to be correlated with reductions in the number of grains per spike, grains per square meter, kernel weight and harvest index. Grain yield losses in the presence of yellow rust were also greater in later heading varieties. The combination of RGB-based indices and days to heading together explained 70.9% of the variability in grain yield and 62.7% of the yield losses.

Diagnosis and management of bacterial infections in decompensated cirrhosis

TML:a language to specify aerial robotic missions for the framework Aerostack

Depth of closure:New calculation method based on sediment data

Obtaining depth of closure(DoC) in an accurate manner is a fundamental issue for coastal engineering,since good results for coastal structures and beach nourishment depend mainly on DoC. Currently, there are two methods for obtaining the DoC, mathematical formulations and profile surveys. However, these methods can incur important errors if one does not take into account the characteristics and morphology of the area, or if one does not have a sufficiently long time series. In this work the DoC is obtained from the break in the trend of the sediment with the depth, that is, in general with the increase of the depth a decrease in the size of the sediment takes place. However, at one point this tendency changes and the size increases, and then decreases again. When comparing the point where the minimum sediment size occurs before the increase, it is observed that the error incurred is small compared to other methods. If the Standard Deviation of Depth Change(SDDC) method is considered as the most accurate method, the error incurred by the proposed method is less than 7%. In addition, it can be seen that the dispersion of the sediment method always occurs outside the zone of bar movement. Whereas in the methods of profiles survey(using 2 cm precision profiles), sometimes the DoC is obtained within the active zone of bar movement. In addition, where the relative minimum of the median sediment size is found, and the sizes of 0.063 and 0.125 mm predominate in the composition of the sample. Therefore, this new method allows the precise location of the DoC to be obtained in a fast and simple way. Furthermore, this method has the advantage that it is not affected by the modifications that may be experienced by both the study area and the cross-shore beach profile.

Seasonal Effect of Sunlight on COVID-19 among Countries with and without Lock-Downs

Objective: The main aim of the study was to determine whether COVID-19 epidemiological data reported by countries in different hemispheres correlated with the seasons of the year. Since stay-at-home orders could be a main factor affecting the time individuals spent outdoors, the progression of COVID-19 in countries that mandated the most stringent lock-downs and stay-at-home orders was compared to countries in the same hemisphere that did not order their citizens to remain at home. Methods: Infections attributed to COVID-19 per million inhabitants, deaths per infections × 100, and deaths per million inhabitants from different countries were analyzed utilizing national reports registered in the Johns’ Hopkins database together with the most recent world population data. The null hypothesis (no difference between countries with and without lock-downs) was tested (two tailed test, p Results: The shift of highest infection rates from countries in the northern-towards countries in the southern-hemisphere during early 2020 and the reverse in December of the same year correlates with the seasonal variation in the flux of germicidal sunlight. Mortality rate for the same virus among different countries did not show a seasonal component. COVID-19 infection mortality rate was considerably lower in developing countries of South America (11 of the largest countries) than in several (at least 8) developed European countries. Discussion: COVID-19 resulted in higher infections during winter than in summer. The finding of a seasonal component, correlating the progression of the pandemic with local solar flux, demonstrates that infectious virus in the environment plays a role in the pandemic since direct person-to-person transmission would afford little time for solar inactivation. Cetuximab （CTX） is a potent chimeric mouse/human monoclonal antibody （mAb） approved worldwide for treatment of metastatic colorectal cancer. Among the various biological and physical analyses per- formed for full study on this biopharmaceutic, the determination of the concentration preparations throughout manufacturing and subsequent handling in hospital is particularly relevant. In the present work, the study and validation of a method for quantifying intact CTX by reverse-phase high-perfor- mance liquid chromatography with diode array detection （（RP）HPLC/DAD） is presented. With that end, we checked the performance of a chromatographic method for quantifying CTX and conducted a study to validate the method as stability-indicating in accordance with the International Conference on Harmo- nization guidelines （ICH） for biotechnological drugs; therefore, we evaluated linearity, accuracy, preci- sion, detection and quantification limits, robustness and system suitability. The specificity of the method and the robustness of the mAb formulation against external stress factors were estimated by compre- hensive chromatographic analysis by subjecting CTX to several informative stress conditions. As de- monstrated, the method is rapid, accurate, and reproducible for CTX quantification. It was also suc- cessfully used to quantify CTX in a long-term stability study performed under hospital conditions.
