BACKGROUND The gut microbiota plays a key role in the maintenance of intestinal homeostasis and the development and activation of the host immune system. It has been shown that commensal bacterial species can regulate the expression of host genes. 16 S rRNA gene sequencing has shown that the microbiota in inflammatory bowel disease(IBD) is abnormal and characterized by reduced diversity. Micro RNAs(miRNAs) have been explored as biomarkers and therapeutic targets, since they are able to regulate specific genes associated with Crohn's disease(CD). In this work, we aim to investigate the composition of gut microbiota of active treatment-na?ve adult CD patients, with miRNA profile from gut microbiota.AIM To investigate the composition of gut microbiota of active treatment-na?ve adult CD patients, with miRNA profile from gut microbiota.METHODS Patients attending the outpatient clinics at Valme University Hospital without relevant co-morbidities were matched according to age and gender. Faecal samples of newonset CD patients, free of treatment, and healthy controls were collected. Faecal samples were homogenized, and DNA was amplified by PCR using primers directed to the 16 S bacterial rRNA gene. Pyrosequencing was performed using GS-Junior platform. For sequence analysis, MGRAST server with the database Ribosomal Project was used. MiRNA profile and their relative abundance were analyzed by quantitative PCR.RESULTS Microbial community was characterized using 16 S rRNA gene sequencing in 29 samples(n = 13 CD patients, and n = 16 healthy controls). The mean Shannon diversity was higher in the healthy control population compared to CD group(5.5 vs 3.7). A reduction in Firmicutes and an increase in Bacteroidetes were found. Clostridia class was also significantly reduced in CD. Principal components analysis showed a grouping pattern, identified in most of the subjects in both groups, showing a marked difference between control and CD groups. Microglia are the resident macrophages of the brain,originally described by Pio del Rio-Hortega(a student of Santiago Ramon y Cajal)in a series of studies in 1919.Since those pioneering studies,many others have followed to describe microglia as complex and multitasking cells with many diverse roles under physiological(e.***.their key role in synapse pruning during development)or diseased conditions.Microglia exist as sentinels or surveyors of the environment that surround neurons,becoming reactive upon a wide array of stimuli and consequently developing an appropriate inflammatory response.In fact,a neuroinflammatory response driven by microglia is found in virtually every disease process that occurs within the central nervous system.Therefore,a better understanding of the mechanisms governing the microglia response is the key to improving the outcome of neurodegenerative conditions.

Abscisic acid(ABA)is an important hormone that regulates both plant development and stress responses(Chen et al.,2020).In the absence of ABA,group A PROTEIN PHOSPHATASE 2Cs(PP2Cs)interact with and inhibit SUCROSE-NON-FERMENTING 1-RELATED KINASES 2(SnRK2s).When ABA is present,ABA receptor PYRABACTIN RESISTANCE 1 and PYRABACTIN RESISTANCE 1-LIKE PROTEINS bind to and inhibit PP2Cs,allowing phosphorylation and activation of SnRK2s that,in turn,phosphorylate downstream targets,such as transcription factors,ion channels,and transporters(Figure 1).

Photoelectrochemical CO_(2) electrolyzers: From photoelectrode fabrication to reactor configuration

The photoelectrochemical conversion of CO_(2) into value-added products emerges as an attractive approach to alleviate climate change. One of the main challenges in deploying this technology is, however, the development and optimization of(photo)electrodes and photoelectrolyzers. This review focuses on the fabrication processes, structure, and characterization of(photo)electrodes, covering a wide range of fabrication techniques, from rudimentary to automated fabrication processes. The work also highlights the most relevant features of(photo)electrodes, with special emphasis on how to measure and optimize them. Finally, the review analyses the integration of(photo)electrodes in different photoelectrolyzer architectures, analyzing the most recent research work that comprises photocathode, photoanode,photocathode-photoanode, and tandem photoelectrolyzer configurations to ideally achieve self-sustained CO_(2) conversion systems. Overall, comprehensive guidelines are provided for future advancements in developing effective devices for CO_(2) conversion, bridging the gap towards the use of sunlight as the unique energy input and practical applications.

Fuzzy C-means Rule Generation for Fuzzy Entry Temperature Prediction in a Hot Strip Mill

Variable estimation for finishing mill set-up in hot rolling is greatly affected by measurement uncertainties, variations in the incoming bar conditions and product changes. The fuzzy C-means algorithm was evaluated for rule base generation for fuzzy and fuzzy grey-box temperature estimation. Experimental data were collected from a real- life mill and three different sets were randomly drawn. The first set was used for rule-generation, the second set was used for training those systems with learning capabilities, while the third one was used for validation. The perform- ance of the developed systems was evaluated by five performance measures applied over the prediction error with the validation set and was compared with that of the empirical rule-base fuzzy systems and the physical model used in plant. The results show that the fuzzy C-means generated rule-bases improve temperature estimation; however, the best results are obtained when fuzzy C-means algorithm, grey-box modeling and learning functions are combined. Application of fuzzy C-means rule generation brings improvement on performance of up to 72%.

Immune checkpoint inhibitors(and more specifically programmed cell death 1/programmed cell death ligand 1 inhibitors as Pembrolizumab)initiated a revolution in the field of melanoma and have now expanded to several tumor subtypes and in increasingly broader clinical contexts,including the adjuvant and neoadjuvant setting,with potentially curable patients and prolonged survival.The side effects related to these drugs include a wide spectrum of manifestations,with endocrinological adverse events being some of the most frequent.Pembrolizumab-induced type 1 diabetes mellitus is an infrequent but potentially serious and not clearly reversible side effect that possesses characteristic clinical features and has high morbidity and mortality,with a chronic impact on quality of life.The etiopathogenesis of this phenomenom needs to be further investigated and a collaborative effort through the involvement of oncologists and other medical specialists is necessary for the correct identification and management of patients at risk.

Biological weathering of phlogopite during enriched vermicomposting

As the dominant macrofauna in many soils,earthworms play a significant role in mineral weathering and nutrient release from mineral structures into available forms for root uptake.Phlogopite is a trioctahedral micaceous mineral that can be weathered to other minerals through release of its interlayer potassium(K).In this study,we investigated the effect of earthworm activities and associated microbial communities on phlogopite weathering during the production of phlogopite-enriched vermicompost with different quantities of phlogopite added.An experiment was run under laboratory conditions using three levels of phlogopite(0,20%and 40%by weight)in triplicates.Each Petri dish contained 20 g of a mixture of cow manure and phlogopite,and vermicomposting process was performed by inoculation of eight Eisenia foetida earthworms.Changes in clay mineralogy and C/N ratio in each dish were assessed at days 0,45,90,135,and 180 during vermicomposting.The results indicated that phlogopite was partially weathered to vermiculite and interstratified mica-vermiculite during the vermicomposting process.The rate of weathering was higher during the early stages of vermicomposting(the first 90 d).The weathering of phlogopite was influenced considerably by the activity of earthworms and associated microbial communities during the organic matter transformation process.At the early stages of the process,the weathering rate for different levels of phlogopite was not significantly different,but over time it accelerated steadily,and the difference increased.

Task Offloading in Edge Computing Using GNNs and DQN

In a network environment composed of different types of computing centers that can be divided into different layers(clod,edge layer,and others),the interconnection between them offers the possibility of peer-to-peer task offloading.For many resource-constrained devices,the computation of many types of tasks is not feasible because they cannot support such computations as they do not have enough available memory and processing capacity.In this scenario,it is worth considering transferring these tasks to resource-rich platforms,such as Edge Data Centers or remote cloud servers.For different reasons,it is more exciting and appropriate to download various tasks to specific download destinations depending on the properties and state of the environment and the nature of the functions.At the same time,establishing an optimal offloading policy,which ensures that all tasks are executed within the required latency and avoids excessive workload on specific computing centers is not easy.This study presents two alternatives to solve the offloading decision paradigm by introducing two well-known algorithms,Graph Neural Networks(GNN)and Deep Q-Network(DQN).It applies the alternatives on a well-known Edge Computing simulator called PureEdgeSimand compares them with the two defaultmethods,Trade-Off and Round Robin.Experiments showed that variants offer a slight improvement in task success rate and workload distribution.In terms of energy efficiency,they provided similar results.Finally,the success rates of different computing centers are tested,and the lack of capacity of remote cloud servers to respond to applications in real-time is demonstrated.These novel ways of finding a download strategy in a local networking environment are unique as they emulate the state and structure of the environment innovatively,considering the quality of its connections and constant updates.The download score defined in this research is a crucial feature for determining the quality of a download path in the GNN training process and has not previous

Bronchiectasis(BE)has been linked to past viral infections such as influenza,measles,or adenovirus.Two years ago,a new pandemic viral infection severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)broke out and it still persists today,and a significant proportion of surviving patients have radiological and clinical sequelae,including BE.Our aim was to thoroughly review the information available in the literature on the bidirectional relationship between SARS-CoV-2 infection and the development of BE,as well as the impact of this infection on patients already suffering from BE.Available information indicates that only a small percentage of patients in the acute phase of coronavirus disease 2019(COVID-19)pneumonia develop BE,although the latter is recognized as one of the radiological sequelae of COVID-19 pneumonia,especially when it is caused by traction.The severity of the initial pneumonia is the main risk factor for the development of future BE,but during the COVID-19 pandemic,exacerbations in BE patients were reduced by approximately 50%.Finally,the impact of BE on the prognosis of patients with COVID-19 pneumonia is not yet known.

Beyond their function as structural barriers,plant cell walls are essential elements for the adaptation of plants to environmental conditions.Cell walls are dynamic structures whose composition and integrity can be altered in response to environmental challenges and developmental cues.These wall changes are perceived by plant sensors/receptors to trigger adaptative responses during development and upon stress perception.Plant cell wall damage caused by pathogen infection,wounding,or other stresses leads to the release of wall molecules,such as carbohydrates(glycans),that function as damage-associated molecular patterns(DAMPs).DAMPs are perceived by the extracellular ectodomains(ECDs)of pattern recognition receptors(PRRs)to activate pattern-triggered immunity(PTI)and disease resistance.Similarly,glycans released from the walls and extracellular layers of microorganisms interacting with plants are recognized as microbe-associated molecular patterns(MAMPs)by specific ECD-PRRs triggering PTI responses.The number of oligosaccharides DAMPs/MAMPs identified that are perceived by plants has increased in recent years.However,the structural mechanisms underlying glycan recognition by plant PRRs remain limited.Currently,this knowledge is mainly focused on receptors of the LysM-PRR family,which are involved in the perception of various molecules,such as chitooligosaccharides from fungi and lipo-chitooligosaccharides(i.e.,Nod/MYC factors from bacteria and mycorrhiza,respectively)that trigger differential physiological responses.Nevertheless,additional families of plant PRRs have recently been implicated in oligosaccharide/polysaccharide recognition.These include receptor kinases(RKs)with leucine-rich repeat and Malectin domains in their ECDs(LRR-MAL RKs),Catharanthus roseus RECEPTOR-LIKE KINASE 1-LIKE group(CrRLK1L)with Malectin-like domains in their ECDs,as well as wall-associated kinases,lectin-RKs,and LRR-extensins.The characterization of structural basis of glycans recognition by these new plant receptors will shed light
