Imprinting in plants

Genomic imprinting leads to the differential expression of parental alleles after fertilization. Imprinting appears to have evolved independently in mammals and flowering plants to regulate the development of nutrient-transfer placental tissues. In addition, the regulation of imprinting in both mammals and flowering plants involves changes in DNA methylation and histone methylation, thus suggesting that the epigenetic signals that regulate imprinting have been co-opted in these distantly related species.

Objective： To examine the effect of metanol extract of Petiveria alliacea （PM） on airway inflammation in a murine model of chronic asthma. Methods： Two-month-old male BALB/c mice （n=6-8/group） were sensitized on days 0 and 14 by intraperitoneal injection of 20μg ovalbumin （OVA）. On day 25, the mice received an airway challenge with OVA （3%, w/v, in phosphate buffered saline）. PM was administered orally by oral gavage to mice at doses of 100, 200 and 400 mg/kg body weight once daily from days 18 to 23. Control mice were orally administered phosphate buffered saline （PBS） to induce a model of asthma. At the end of the test, respiratory reactivity was assayed, the total cell number, interleukin-4 （IL-4）, IL-5, IL-13, tumor necrosis factor-alpha （TNF-α） and reactive oxygen species （ROS） in the bronchoalveolar lavage fluid （BALF） were determined and the levels of serum IgE, intercellular cell adhesion molecule 1 （ICAM-1） and eotoxin were measured. In addition, lung tissue was used to qualify the IL-4, IL-5, IL-13, TNF-α and transforming growth factor beta 1 （TGF-β1）. Histologic examination was performed to observe inflammatory cellular infiltration. Results： The administration of PM in comparison with the OVA-only treated group significantly attenuated the infiltration of eosinophUs and other inflammatory cells （P〈0.01）. Airway resistance （RI） in the OVA-only induced group was significantly higher than that of the PBS control group （P〈0.01） when methacholine was added. TNF-α, IgE, TGF-β1 and cytokine levels IL-4, IL-5, IL-β in the BALF decreased compared to control mice （P〈0.01 or P〈0.05）. PM treatment also inhibited the production of chemokines, eotaxin and ICAM-1 in BALF （P〈0.01）, which improved lung function. Histopathological examination revealed that the sensitized treated PM groups had significant lower in inflammatory scores similar to dexamethasone treatments and the untreated group. In this study, we evaluated the effects of genistein supplementation of the thawing extender on frozen-thawed human semen parameters. We analyzed the effect of supplementation on sperm motility, capacitation （membrane lipid disorder）, reactive oxygen species （ROS） generation, chromatin condensation and DNA damage. Using this preliminary information, it maybe possible to improve the cryopreservation process and reduce the cellular damage. We have confirmed that the isoflavone genistein （10 μmol L-1） has antioxidant properties on the frozen-thawed spermatozoa. This results in a decreased ROS production that shows a slight improvement in the sperm motility, and decreases the membrane lipid disorder and DNA damage caused by cryopreservation. These results suggest an effect of genistein on sperm functionality that could be of interest for assisted reproduction treatments using frozen- thawed human spermatozoa, but further studies will be necessary to confirm our findings and to evaluate the possible clinical applications.

Nitrogen （N） is an essential macronutrient that affects plant growth and development. N is an important component of chlorophyll, amino acids, nucleic acids, and secondary metabolites. Nitrate is one of the most abundant N sources in the soil. Because nitrate and other N nutrients are often limiting, plants have developed sophisticated mechanisms to ensure adequate supply of nutrients in a variable environment. Nitrate is absorbed in the root and mobilized to other organs by nitrate transporters. Nitrate sensing activates signaling pathways that impinge upon molecular, metabolic, physiological, and developmental responses locally and at the whole plant level. With the advent of genomics technologies and genetic tools, important advances in our understanding of nitrate and other N nutrient responses have been achieved in the past decade. Furthermore, techniques that take advantage of natural polymor- phisms present in divergent individuals from a single species have been essential in uncovering new components. However, there are still gaps in our understanding of how nitrate signaling affects biolog- ical processes in plants. Moreover, we still lack an integrated view of how all the regulatory factors iden- tified interact or crosstalk to orchestrate the myriad N responses plants typically exhibit. In this review, we provide an updated overview of mechanisms by which nitrate is sensed and transported throughout the plant. We discuss signaling components and how nitrate sensing crosstalks with hormonal pathways for developmental responses locally and globally in the plant. Understanding how nitrate impacts on plant metabolism, physiology, and growth and development in plants is key to improving crops for sustainable agriculture.

Diabetic retinopathy (DR) is a complication of diabetesmellitus thatappears in the retina. Clinitians use retina images to detect DR pathologicalsigns related to the occlusion of tiny blood vessels. Such occlusion brings adegenerative cycle between the breaking off and the new generation of thinnerand weaker blood vessels. This research aims to develop a suitable retinalvasculature segmentation method for improving retinal screening proceduresby means of computer-aided diagnosis systems. The blood vessel segmentationmethodology relies on an effective feature selection based on SequentialForward Selection, using the error rate of a decision tree classifier in theevaluation function. Subsequently, the classification process is performed bythree alternative approaches: artificial neural networks, decision trees andsupport vector machines. The proposed methodology is validated on threepublicly accessible datasets and a private one provided by Hospital Sant Joanof Reus. In all cases we obtain an average accuracy above 96% with a sensitivityof 72% in the blood vessel segmentation process. Compared with the state-ofthe-art, our approach achieves the same performance as other methods thatneed more computational *** method significantly reduces the numberof features used in the segmentation process from 20 to 5 dimensions. Theimplementation of the three classifiers confirmed that the five selected featureshave a good effectiveness, independently of the classification algorithm.

The inter-cellular translocation of nutrients in plant is mediated by highly specialized transfer cells （TCs）. TCs share similar functional and structural features across a wide range of plant species, including location at plant exchange surfaces, rich in secondary wall ingrowths, facilitation of nutrient flow, and passage of select molecules. The fate of endosperm TCs is determined in the TC fate acquisition stage （TCF）, before the structure features are formed in the TC differentiation stage （TCD）. At present, the molecular basis of TC development in plants remains largely unknown. In this review, we summarize the important roles of the signaling molecules in different development phases, such as sugars in TCF and phytohormones in TCD, and discuss the genetic and epigenetic factors, including TC-specific genes and endogenous plant peptides, and their crosstalk with these signaling molecules as a complex regulatory network in regulation of TC develonment in olants.
