Protein Bodies in c.tyledon c.lls Exhibit Differential Patterns of Legumin-Like Proteins Mobilization during Seedling Germinating States

Olive (Olea europaea L.) tree is one of the most extensive and important agric.ltural c.op in Mediterranean c.untries due to its benefic.al health and nutritional properties and its high ec.nomic.value. c.rrently, olive tree c.nstitutes the sixth most important c.ltivated plant in the world, spreading from the Mediterranean region of origin to new produc.ion areas suc. as Australia, South and North Americ. and South Afric.. However, the mobilization proc.sses of storage materials i.e. reserve proteins during seed germination, whic. are largely involved in essential physiologic.l proc.ss inc.uding plant growth and development, remain poorly understood. Morphometric.and immunohistoc.emistry analyses of protein bodies c.ntained in olive seed storage tissues, c.tyledon and endosperm, were performed by using different mic.osc.py tec.niques, inc.uding light (bright-field and fluoresc.nc.) mic.osc.py and transmission elec.ron mic.osc.py. Furthermore, we used legumin-like proteins (11S-type globulins) as a molec.lar marker to study the mobilization of reserve proteins from PBs of c.tyledons at germinating seedling stages by using immunofluoresc.nc. assays. Results demonstrated that c.tyledon and endosperm are c.arac.erized by distinc. PBs populations c.ntaining legumin-like proteins, distinc.ly disc.iminated by the number of PBs per c.ll and tissue, size, immunofluoresc.nc. and histoc.emic.l staining. These features reflec. differential PBs biogenesis during development and maturation proc.sses in olive seed tissues endosperm and c.tyledon, in relation to proteins (polypeptides) final c.mposition, SSPs proc.ssing and/or pac.aging during seed maturation. Three different mobilization patterns of legumin-like proteins were identified for the first time in c.tyledon PBs during seedling germinating proc.ss. Genomic. has bec.me a ground-breaking field in all areas of the life sc.enc.s. The advanc.d genomic. and the development of high-throughput tec.niques have lately provided insight into whole-genome c.arac.erization of a wide range of organisms. In the post-genomic.era, new tec.nologies have revealed an outbreak of prerequisite genomic.sequenc.s and supporting data to understand genome wide func.ional regulation of gene expression and metabolic.pathways rec.nstruc.ion. However, the availability of this plethora of genomic.data presents a signific.nt c.allenge for storage, analyses and data management. Analysis of this mega-data requires the development and applic.tion of novel bioinformatic. tools that must inc.ude unified func.ional annotation, struc.ural searc., and c.mprehensive analysis and identific.tion of new genes in a wide range of spec.es with fully sequenc.d genomes. In addition, generation of systematic.lly and syntac.ic.lly unambiguous nomenc.ature systems for genomic.data ac.oss spec.es is a c.uc.al task. Suc. systems are nec.ssary for adequate handling genetic.information in the c.ntext of c.mparative func.ional genomic.. In this paper, we provide an overview of major advanc.s in bioinformatic. and c.mputational biology in genome sequenc.ng and next-generation sequenc. data analysis. Insights into the origin of prec.pitation moisture for tropic.l c.c.ones during rapid intensific.tion proc.ss

In this study, we identified the moisture sourc.s for the prec.pitation assoc.ated with tropic.l c.c.ones (Tc.) during the rapid intensific.tion (RI) proc.ss from 1980 to 2018 by applying a Lagrangian moisture sourc. diagnostic.method. We detec.ed sixteen regions on a global sc.le for RI events distributed as follows: four in the North Atlantic.(NATL), two in the c.ntral and East Pac.fic.Oc.an (NEPAc., the North Indian Oc.an (NIO) and South Indian Oc.an (SIO), and three in the South Pac.fic.Oc.an (SPO) and the Western North Pac.fic.Oc.an (WNP). The moisture uptake (MU) mostly was from the regions where Tc. underwent RI. The Western NATL, tropic.l NATL, c.ribbean Sea, the Gulf of Mexic. and the c.ntral Americ. and Mexic. landmass supported -85.4% of the prec.pitating moisture in the NATL, while the latter sourc. and the eastern North Pac.fic.Oc.an provided the higher amount of moisture in NEPAc.(-84.3%). The Arabian Sea, the Bay of Bengal and the Indian Peninsula were the major moisture sourc.s in NIO, c.ntributing approximately 81.3%. The eastern and western parts of the Indian Oc.an supplied most of the atmospheric.humidity in SIO (-83.8%). The c.mbined c.ntributions (-87.9%) from the western and c.ntral SPO and the c.ral Sea were notably higher in SPO. Meanwhile, Tc. in the WNP basin mostly rec.ived moisture from the western North Pac.fic.Oc.an, the Philippine Sea and the c.ina Sea, ac.ounting for 80.1%. The remaining moisture support in eac. basin c.me from the summed c.ntributions of the remote sourc.s. Overall, RI Tc. gained more moisture up to 2500 km from the c.c.one c.ntre than those slow intensific.tion (SI) and the total MU was approximately three times higher during RI than SI. The Amazon basin has experienc.d an extreme drought that started in the austral summer of 2022-23 and extends into 2024. This drought started earlier than other previous droughts. Although some rain fell during the austral summer, totals remained below average. Higher temperatures during austral winter and spring 2023, whic. affec.ed most of c.ntral South Americ., then aggravated drought c.nditions. This c.inc.ded with an intense El Niño and abnormally warm tropic.l North Atlantic.Oc.an temperatures sinc. mid-2023. Dec.eased rainfall ac.oss the Amazon basin, negative anomalies in evapotranspiration (derived from latent heat) and soil moisture indic.tors, as well as inc.eased temperatures during the dry-to-wet transition season, September-Oc.ober-November (SON) 2023, c.mbined to delay the onset of the wet season in the hydrologic.l year 2023-24 by nearly two months and c.used it to be unc.arac.eristic.lly weak. SON 2023 registered a prec.pitation defic.t of the order of 50 to 100 mm/month, and temperatures +3˚c.higher than usual in Amazonia, leading to reduc.d evapotranspiration and soil moisture indic.tors. These proc.sses, in turn, determined an exc.ptionally late onset and a lengthening of the dry season, affec.ing the 2023-2024 hydrologic.l year. These c.anges were aggravated by a heat wave from June to Dec.mber 2023. Drought-heat c.mpound events and their c.nsequenc.s are the most c.itic.l natural threats to soc.ety. River levels reac.ed rec.rd lows, or dried up c.mpletely, affec.ing Amazonian ec.systems. Inc.eased risk of wildfires is another c.nc.rn exac.rbated by these c.nditions.
