BACKGrOUND Coronavirus disease 2019(COvID-19)can be associated with life-threatening organ dysfunction due to septic shock,frequently requiring intensive care unit(ICU)admission,respi-ratory and vasopressor ***-fore,clear clinical criteria are pivotal for early recognition of patients more likely to need prompt organ *** most patients with severe COvID-19 meet the Sepsis-3.0 criteria for septic shock,it has been increasingly recognized that hyperlactatemia is frequently absent,possibly leading to an underestimation of illness severity and mortality *** To identify the proportion of severe COvID-19 patients with vasopressor support requirements,with and without hyperlactatemia,and describe their clinical outcomes and mortality METHODS We performed a single-center prospective cohort *** adult patients admitted to the ICU with COvID-19 were included in the analysis and were further divided into three groups:Sepsis group,without both criteria;vasoplegic Shock group,with persistent hypotension and vasopressor support without hyperlactatemia;and Septic Shock 3.0 group,with both ***-19 was diagnosed using clinical and radiologic criteria with a severe acute respiratory syndrome coronavirus 2(SArS-Cov-2)positive rT-PCr *** 118 patients(mean age 63 years,87%males)were included in the analysis(n=51 Sepsis group,n=26 vasoplegic Shock group,and n=41 Septic Shock 3.0 group).SOFA score at ICU admission and ICU length of stay were different between the groups(Prtality was significantly higher in the vasoplegic Shock and Septic Shock 3.0 groups when compared with the Sepsis group(Prence between the former two groups(P=0.713).The log rank tests of Kaplan-Meier survival curves were also different(P=0.007).ventilator-free days and vasopressor-free days were different between the Sepsis vs vasoplegic Shock and Septic Shock 3.0 groups(both Pr in the last two groups(P=0.128

In recent years,there has been a growing interest in exploring alternative treatments for bone *** tissue engineering has turned its attention to plant extracts containing osteogenic flavonoids as potential promoters of bone *** our study,we specifically investigated the extract of Petroselinum crispum,a plant known for its abundance of osteogenic flavonoids such as apigenin,quercetin,and *** objective was to compare the total flavonoid content(TFC)and their accumulation in different *** obtained hydrolyzed aqueous extracts from the leaves of parsley plants(grown for 12 weeks in the field),weekly embryogenic cultures,and suspension cell *** TFC,measured in mg quercetin equivalent per gram of dry weight(QE mg/g dw),was found to be 53.81±0.97 for 52 g dw of leaves(1 kg plant fw),31.10±1.52 for 420 g dw of embryogenic cultures,and 11.80±0.76 for 210 g dw of cell ***,only the leaves and embryogenic cultures showed significant accumulation of osteogenic flavonoids,with apigenin levels of 1.8±0.3 and 0.32±0.02 mg/g dw,and kaempferol levels of 1.0±0.18 and 0.2±0.01 mg/g dw,*** findings indicate that embryogenic cultures have the potential to produce up to 40%more apigenin in 12 weeks compared to 1 kg of parsley plant,thanks to their superior biomass reproductive *** results present a promising biotechnological approach for generating extracts enriched with bioactive compounds,which can be further utilized in in vitro osteogenic testing.

Dengue is one of the most prominent tropical epidemic diseases present in the rio de Janeiro city and Southeast part of Brazil, due to the widespread conditions of occurrence of the dengue vector, the mosquito Aedesaegypti, such as high-temperature days interlaced with afternoon or nocturnal rainstorms in summer. This work has the objective of investigating the relationships between variabilities of the El Ni?o-South Oscillation (ENSO) and greater epidemics of dengue in rio de Janeiro city. To accomplish this goal, the analysis and signal decomposition by cross-wavelet transform (WT) was applied to obtain the cross variability associated with variations of power and phase of both signals by characteristic periods and along with the time series. Data considered in the analysis are (the decimal logarithm of normalized value) of the monthly available notifications of dengue worsening, provided by the public health system of Brazil, and the Southern Oscillation Index (SOI) Ni?o 3.4 data, provided by the National Oceanic and Atmospheric Administration (NOAA), in the period 2000-2017. A maximum cross-wavelet power close to 0.45 was obtained for the representative period of 1 year and also to periods between 3 and 4 years, associated with the positive phase of the SOI index (i.e. , La Ni?a) or with a transition to the positive phase. The evolution of the combined variability of SOI and dengue can be expressed by progressive differences in phase along the time, eventually resulting in yielding phases (i.e., La Niña-Dengue epidemic).

Grapevine is one of the most economically important crops ***,the previous versions of the grapevine reference genome tipically consist of thousands of fragments with missing centromeres and telomeres,limiting the accessibility of the repetitive sequences,the centromeric and telomeric regions,and the study of inheritance of important agronomic traits in these ***,we assembled a telomere-to-telomere(T2T)gap-free reference genome for the cultivar PN40024 using PacBio HiFi long *** T2T reference genome(PN_T2T)is 69 Mb longer with 9018 more genes identified than the 12X.v0 *** annotated 67%repetitive sequences,19 centromeres and 36 telomeres,and incorporated gene annotations of previous versions into the PN_T2T *** detected a total of 377 gene clusters,which showed associations with complex traits,such as aroma and disease *** though PN40024 derives from nine generations of selfing,we still found nine genomic hotspots of heterozygous sites associated with biological processes,such as the oxidation–reduction process and protein *** fully annotated complete reference genome therefore constitutes an important resource for grapevine genetic studies and breeding programs.

The conventional processes for recovery of silver from cyanide leach solutions are the carbon adsorption, the Merrill-Crowe zinc dust cementation, the Ion Exchange, and Solvent Extraction processes;among other available options for recovery of precious metals from cyanide solutions, Electrocoagulation (EC) is a very promising electrochemical process that does not require high concentrations of silver in cyanide solutions to yield excellent results and neither pretreatment of cyanide solutions like Merrill-Crowe process (deoxygenating and clarification). The present study has been done for the recovery of silver contained in pregnant solution from the cyanidation process using the electrocoagulation technology with iron electrodes, and therefore develops an alternative technology for Merril-Crowe process. The average silver content in pregnant solution was of 52 ppm, recovery was obtained of 99% of silver, with this optimum operating parameters, pH = 8, residence time = 20 minutes and conductivity by addition of sodium chloride = 4 grs/L. Finally the characterization of the solid products formed during the EC process with X-ray Diffraction and Scanning Electronic Microscope was performed, results suggest that magnetite particles and amorphous iron oxyhydroxides are present (Lepidocrocite).

ObjectiveTo characterize effects of the GP Iba mutation(A156v)on its interaction with von Willebrand factor(vWf)under high fluid shear stress.MethodsThe residue A156 of GP Iba was converted to a valine and the mutant expressed in CHO cells expressing wild-type GP Ibβ and GPIX. The transfected cells were tested for their interaction with a panel of GP Iba antibodies and for rolling on immobilized vWf under high shear.resultsThe mutation led to surface expression of a GP Iba polypeptide that adopted a different conformation at its N-terminus because binding of the GP Iba antibody AN51, which has a binding epitope it the N-terminal 35 residues, was eliminated, whereas binding of the others(AK2, MB45, and SZ2, all of which bind to regions C-terminal to the AN51 epitope)was normal. Mutant-expressing cells could adhere and roll on immobilized vWf under high fluid shear stress and rolled significantly faster than wild-type cells.ConclusionThese studies demonstrate that the mutation A156v results in a conformation change at the N-terminus of GP Ibα, which leads to an increase in the dissociation rate of the bond between the GP Ibα mutant and vWf. In this work we introduce a modified version of the simple genetic algorithm (MGA) and will show the results of its application to two GMA power law models (a general theoretical branched pathway system and a mathematical model of the amplification and responsiveness of the JAK2/STAT5 pathway representing an actual, experimentally studied system). The two case studies serve to illustrate the utility and potentialities of the MGA method for concerning parameter estimation in complex models of biological significance. The analysis of the results obtained from the application of the MGA algorithm allows an evaluation of the potentialities and shortcomings of the proposed algorithm when compared with other parameter estimation algorithm such as the simple genetic algorithm (SGA) and the simulated annealing (SA). MGA shows better performance in both studied cases than SGA and SA, either in the presence or absence of noise. It is suggested that these advantages are due to the fact that the objective function definition in the MGA could include the experimental error as a weight factor, thus minimizing the distance between the data and the predicted value. Actually, MGA is slightly slower that the SGA and the SA, but this limitation is compensated by its greater efficiency in finding objective values closer to the global optimum. Finally, MGA can lead to an early local optimum, but this shortcoming may be prevented by providing a great population diversity through the insertion of different selection processes.

Background:To assess the clinical outcomes of the visian Implantable Collamer Lens(ICL)with a central port throughout 7 years of ***:Eighty-four eyes of 52 patients were evaluated over a follow-up period of 7 years after v4c ICL ***(UDvA)and corrected(CDvA)distance visual acuities,refraction,intraocular pressure(IOP),endothelial cell density(ECD)and vault were ***:The mean postoperative UDvA(logMAr)was 0.04±0.11,0.13±0.19 and 0.17±0.23 at 1-,5-and 7-years,respectively(PvA(logMAr)remains unchanged throughout a 7-year follow-up period(0.02±0.08 and 0.02±0.08,at 5-and 7-years,respectively,P=0.2).At all follow-up visits,more than 95%of the eyes achieved a CDvA of 20/25 or better and more than 85%a CDvA of 20/*** the end of the follow-up(7 years),no eye lost more lines of CDvA,56 eyes(66.7%)and 28 eyes(33.3%)gained lines of *** 7-years,the spherical equivalent was−0.62±0.62 *** significant increase in IOP(>20 mmHg or an increase higher than 5 mmHg)occurred in any case throughout the 7-year of *** loss in ECD from the preoperative baseline at the last follow-up visit was 2.6%.No intraoperative or postoperative complications or adverse events occurred during the follow-up ***:The outcomes of this study show the long-term viability of the v4c ICL implantation as a surgical option for the correction of myopia.
