From contraction to gene expression:nanojunctions of the sarco/endoplasmic reticulum deliver site-and function-specific calcium signals

Calcium signals determine, for example, smooth muscle contraction and changes in gene expression. How calcium signals select for these processes is enigmatic. We build on the "panjunctional sarcoplasmic reticulum" hypothesis, describing our view that different calcium pumps and release channels, with different kinetics and affinities for calcium, are strategically positioned within nanojunctions of the SR and help demarcate their respective cytoplasmic nanodomains. SERCA2 b and Ry R1 are preferentially targeted to the sarcoplasmic reticulum(SR) proximal to the plasma membrane(PM), i.e., to the superficial buffer barrier formed by PM-SR nanojunctions, and support vasodilation. In marked contrast, SERCA2 a may be entirely restricted to the deep, perinuclear SR and may supply calcium to this sub-compartment in support of vasoconstriction. Ry R3 is also preferentially targeted to the perinuclear SR, where its clusters associate with lysosome-SR nanojunctions. The distribution of Ry R2 is more widespread and extends from this region to the wider cell. Therefore, perinuclear Ry R3 s most likely support the initiation of global calcium waves at L-SR junctions, which subsequently propagate by calcium-induced calcium release via Ry R2 in order to elicit contraction. Data also suggest that unique SERCA and Ry R are preferentially targeted to invaginations of the nuclear membrane. Site- and function-specific calcium signals may thus arise to modulate stimulus-response coupling and transcriptional cascades.

Natural products:Perspectives in the pharmacological treatment of gastrointestinal anisakiasis

Objective: To evaluate the activity of different natural products against L3 larvae of Anisakis type I. Methods: Information on investigations into the activity of natural products against the L3 larvae of Anisakis simplex was collected from scientific journals, books, theses and reports via a library and electronic search(using Pubmed, Scopus, Medline, Web of Science and Science Direct). The search terms included: natural products, medicinal plants, essential oils, terpenic derivatives, Anisakis, antinematodal activity. Results: In the literature reviewed numerous papers were found concerning the in vitro and in vivo activity against Anisakis type I of various natural products(plant extracts, essential oils and their major components). Analysis of the results showed that in vitro tests the Melaleuca alternifolia essential oil and the ar-turmerone isolated from Curcuma longa displayed the greatest activity. In vivo, the most active compound was perillaldehyde, the main component of Perilla frutescens essential oil. Conclusions: This study shows that some natural products exhibited promising antianisakis properties.

Dear Editor, The PIWI proteins （originally P-element-induced wimpy testis in Drosophila） are predominantly present in the germ-line in diverse organisms and are involved in the processing of a class of small RNAs known as piRNAs （see Refs. for review）. The human PIWI protein family consists of four members： PIWIL1-4. Of these, PIWIL4 is known to have essential roles in the first phases ofspermatogenesis： its expression is restricted to gonocytes and it is required for transposon silencing. The lack of this gene in mice causes meiotic arrest in spermatogenesis.

Background The identification of modifiable bleeding risk factors may be of relevance.The aim is to evaluate if aortic stenosis(AS)provides additional information to bleeding risk scores for predicting major bleeding(MB)in non-valvular atrial fibrillation(AF).Methods We designed a retrospective multi-center study including 2880 consecutive non-valvular AF patients initiating oral anticoagulation between January 2013 and December 2016.AS was defined as moderate or severe according to European echocardiography guidelines criteria.HASBLED,ATRIA and ORBIT scores were used to evaluate the bleeding risk.MB was defined according to the International Society on Thrombosis and Haemostasia criteria and registered at 18 months of follow-up.Results 168(5.8%)patients had AS.Patients with AS had higher risk for MB compared to those without AS(HR=2.13,95%CI:1.40-3.23,P<0.001).Patients without AS and low-intermediate bleeding risk(0 points)showed the lowest MB rate,whereas the MB rate observed among patients with AS and high bleeding risk(2 points)was the highest one.Discrimination and reclassification analyses showed that AS provided additional information to bleeding risk scores for predicting MB at 18 months of follow-up.Conclusions In this population,AS was associated with an increased risk for MB at midterm follow-up.The three scoring systems showed a moderate discriminatory ability for MB.Moreover,the addition of AS was associated with a significant improvement in their predictive accuracy.We suggest that the presence of this valvulopathy should be taken into account for bleeding risk assessment.

A new lighting and enlargement on phase spectrogram(PS)and frequency spectrogram(FS)is presented in this paper.These representations result from the coupling of power spectrogram and short time Fourier transform(STFT).The main con-tribution is the construction of the 3D phase spectrogram(3DPS)and the 3D frequency spectrogram(3DFS).These new tools allow such specific test signals as small slope linear chirp,phase jump and small frequency jump to be analyzed.An application case of musical signal analysis is reported.The main objective is to detect small frequency and phase variations in order to characterize each type of sound attack without losing the amplitude information given by power spectrogram.

Many orthopedic surgeons require that their patients obtain dental clearance before elective total joint arthroplasty(TJA).However,there is no consensus substantiating the practice.To this end,a systematic review on the prevalence of dental pathology in TJA patients,risk factors for failing dental screening,and impact of dental evaluations was performed.Literature was sourced from PubMed and Scopus databases.Six papers were sourced from the initial search,one study was extracted from the references of the original six manuscripts,and one new publication was retrieved from a second search conducted after the first.The prevalence of dental pathology ranged from 8.8%to 29.4%across studies.Two of four papers reported lower than average or improvements in postoperative infection with pre-operative dental evaluations while two found no such association.There is insufficient evidence to support universal dental clearance before TJA.

Yersinia type Ⅲ effectors perturb host innate immune responses

The innate immune system is the first line of defense against invading pathogens. Innate immune cells recognize molecular patterns from the pathogen and mount a response to resolve the infection. The production of proinflammatory cytokines and reactive oxygen species, phagocytosis, and induced programmed cell death are processes initiated by innate immune cells in order to combat invading pathogens. However, pathogens have evolved various virulence mechanisms to subvert these responses. One strategy utilized by Gram-negative bacterial pathogens is the deployment of a complex machine termed the type Ⅲ secretion system(T3SS). The T3SS is composed of a syringe-like needle structure and the effector proteins that are injected directly into a target host cell to disrupt a cellular response. The three human pathogenic Yersinia spp.(Y. pestis, Y. enterocolitica, and Y. pseudotuberculosis) are Gramnegative bacteria that share in common a 70 kb virulence plasmid which encodes the T3 SS. Translocation of the Yersinia effector proteins(YopE, YopH, YopT, YopM, YpkA/YopO, and YopP/J) into the target host cell results in disruption of the actin cytoskeleton to inhibit phagocytosis, downregulation of proinflammatory cytokine/chemokine production, and induction of cellular apoptosis of the target cell. Over the past 25 years, studies on the Yersinia effector proteins have unveiled tremendous knowledge of how the effectors enhance Yersinia virulence. Recently, the long awaited crystal structure of YpkA has been solved providing further insights into the activation of the YpkA kinase domain. Multisite autophosphorylation by YpkA to activate its kinase domain was also shown and postulated to serve as a mechanism to bypass regulation by host phosphatases. In addition, novel Yersinia effector protein targets, such as caspase-1, and signaling pathways including activation of the inflammasome were identified. Tumor-secreted extracellular vesicles(EVs)participate in the metastasis process through different mechanisms,including the preparation of the pre-metastatic niche to grant circulating tumor cells(CTCs)implantation and growth.The study of the metastasis process through the analysis of CTCs and tumor-derived EVs is difficult because of the dilution grade of these elements in peripheral blood.In early-stage lung cancer patients,the tumor-secreted products are even more diluted.An attractive strategy in surgical lung cancer patients is to purify them from a pulmonary tumor-draining vein where they are enriched.The information obtained from the analysis of EVs and CTCs purified from this source could give more accurate information about tumor biology and could be an important source of biomarkers to identify patients at high risk of relapse after curative surgery.

Effect of malathion on the male reproductive organs of earthworms, Eisenia foetida

Aim: To observe the cytotoxic effect of the organophosphate insecticide malathion in the reproductive tissues of the earthworms, Eisenia foetida. Methods: Worms were nourished in soil treated with malathion at single sub-lethal doses of 0, 80, 150, 300 and 600 mg-kg^(-1) soil. (LD_(50) = 880 mg kg^(-1) soil) and evaluated on days 1, 5, 15 and 30 after exposure. The body weights were recorded and male reproductive organs evaluated. Results: Malathion-treated animals showed a significant reduction in body weight in a dose-dependent manner. Malathion treatment modified the disposition of spermatozoa in the basal epithelium of the spermatheca. The Br-deoxyuridine test showed a significant rise in cells in phase S on days 5 and 15. Also, a higher percentage of spermatogonia with fragmented DNA were observed by means of the TdT-mediated dUTP nick-end labeling (TUNEL) technique in the spermatheca of treated animals. Conclusion: Treatment with malathion decreased the body weight and the spermatic viability in spermatheca, altering the cell proliferation and modifying the DNA structure of spermatogonia.
