Cancer is an abnormal state of cells where they undergo uncontrolled proliferation and produce aggressive malignancies that causes millions of deaths every *** the new understanding of the molecular mechanism(s)of disease progression,our knowledge about the disease is snowballing,leading to the evolution of many new therapeutic regimes and their successive *** the past few decades,various combinations of therapies have been pro-posed and are presently employed in the treatment of diverse *** drug therapy,immunotherapy,and personalized medicines are now largely being employed,which were not common a few years *** field of cancer discoveries and therapeutics are evolving fast as cancer type-specific biomarkers are progressively being identified and several types of cancers are nowadays undergoing systematic therapies,extending patients’disease-free survival *** growing evidence shows that a systematic and targeted approach could be the future of cancer medicine,chemotherapy remains a largely opted therapeutic option despite its known side effects on the patient’s physical and psychological ***-apeutic agents/pharmaceuticals served a great purpose over the past few decades and have remained the frontline choice for advanced-stage malignancies where surgery and/or radiation therapy cannot be prescribed due to specific *** present report succinctly reviews the existing and contemporary advancements in chemotherapy and assesses the status of the enrolled drugs/pharmaceuticals;it also comprehensively discusses the emerging role of specific/targeted therapeutic strategies that are presently being employed to achieve better clinical success/survival rate in cancer patients.

Calculation of Two-Tailed Exact probability in the Wald-Wolfowitz One-Sample Runs Test

The objectives of this paper are to demonstrate the algorithms employed by three statistical software programs (R, Real Statistics using Excel, and SpSS) for calculating the exact two-tailed probability of the Wald-Wolfowitz one-sample runs test for randomness, to present a novel approach for computing this probability, and to compare the four procedures by generating samples of 10 and 11 data points, varying the parameters n0 (number of zeros) and n1 (number of ones), as well as the number of runs. Fifty-nine samples are created to replicate the behavior of the distribution of the number of runs with 10 and 11 data points. The exact two-tailed probabilities for the four procedures were compared using Friedman’s test. Given the significant difference in central tendency, post-hoc comparisons were conducted using Conover’s test with Benjamini-Yekutielli correction. It is concluded that the procedures of Real Statistics using Excel and R exhibit some inadequacies in the calculation of the exact two-tailed probability, whereas the new proposal and the SpSS procedure are deemed more suitable. The proposed robust algorithm has a more transparent rationale than the SpSS one, albeit being somewhat more conservative. We recommend its implementation for this test and its application to others, such as the binomial and sign test.

patterns of variation in the size of vulture foraging groups,and their ecological causes and consequences,remain litle explored despite strong links with the carrion recycling service that this key functional group *** documented the group size-frequency pattern of Andean con-dors Vultur gryphus gathered to feed on 42 equine carcasses experimentally placed in Bolivia,between 2012 and 2019,along an elevation range of 1,300-4,500 m *** on the location(altitude and latitude)of the foraging groups,we examined the relationship between their size and species'population parameters(size and trend),habitat conditions,and livestock carcass availability and *** utilized a high frequency(93%)of carcasses forming groups that ranged from 1 to 80 individuals(mean=25,median=18)and shaped a"lazy-J curve"typical pattern of size-frequency distribution whereby few groups(5,12%)were large(>55 individuals)and most(21,50%)were relatively small(p size related to altitude in that most larger groups formed at lower sites(below c.3,000 m asl),likely following an altitudinal gradient whereby larger groups are more likely to form around larger carcasses(i.e.,cattle),which are more likely to occur at lower *** of population size,group size could be an adaptive response of condors via local enhancement for improving individual scavenging *** information gaps on this topic still exist,thus we provide a set of questions to address them,especially amidst the unrestricted impacts of human activities that condition vulture survival globally.

Connexin 43 (CX43) is one of the major components of gap junctions, the structures responsible for the intercellular communication and transmission of the electrical impulse in the left ventricle. There is limited information on the histological changes of CX43 with age and their effect on electrophysiology, especially in humans. Here, we analyzed left ventricular biopsies from living donors starting at midlife to characterize age-related CX43 remodeling. We assessed its quantity, degree of lateralization, and spatial heterogeneity together with fibrotic deposition. We observed no significant age-related remodeling of CX43. Only spatial heterogeneity increased slightly with age, and this increase was better explained by biological age than by chronological age. Importantly, we found that CX43 features varied considerably among individuals in our population with no relevant relationship to age or fibrosis content, in contrast to animal species. We used our experimental results to feed computational models of human ventricular electrophysiology and to assess the effects of interindividual differences in specific features of CX43 and fibrosis on conduction velocity, action potential duration, and arrhythmogenicity. We found that larger amounts of fibrosis were associated with the highest arrhythmic risk, with this risk being increased when fibrosis deposition was combined with a reduction in CX43 amount and/or with an increase in CX43 spatial heterogeneity. These mechanisms underlying high arrhythmic risk in some individuals were not associated with age in our study population. In conclusion, our data rule out CX43 remodeling as an age-related arrhythmic substrate in the population beyond midlife, but highlight its potential as a proarrhythmic factor at the individual level, especially when combined with increased fibrosis.
