Background: Incorporation of HPV tests into cervical cancer screening programs may be advantageous over conventional cytology, especially in developing nations, where the largest burden of cervical cancer is observed. Objectives: To conduct an evaluation of commercially available molecular HPV tests in Brazilian women. Study design: Two groups were recruited: group A was composed of 511 women referred to the clinics because of a previous abnormal Pap test while group B consisted of 2464 subjects under routine screening. Cervical samples were collected using SurePath liquid cytology (LBC) device, and split into aliquots which were submitted to molecular testing by Hybrid Capture and cobas HPV. Colposcopy and biopsies were made according to the standard guidelines, directed by cytological diagnosis. Results: Prevalence of HSIL was 5.97% and 0.7% in Group A and B respectively. High-Risk HPV DNA was found in about 9% of group B women, while in group A this frequency was 24%. Having CIN3+ as the study end-point, the negative predictive values for molecular methods were above 99.8%. All “in-situ” and invasive cervical carcinomas were detected by both HPV nucleic acid assays. Conclusion: Use of HPV DNA testing was feasible and highly sensitive in cancer screening settings of Brazil.

The emerging role of mesenchymal stem cell-derived extracellular vesicles to ameliorate hippocampal NLRP3 inflammation induced by binge-like ethanol treatment in adolescence

Our previous studies have reported that activation of the NLRP3(NOD-,LRR-and pyrin domain-containing protein 3)-inflammasome complex in ethanol-treated astrocytes and chronic alcohol-fed mice could be associated with neuroinflammation and brain *** stem cell-derived extracellular vesicles(MSC-eVs)have been shown to restore the neuroinflammatory response,along with myelin and synaptic structural alterations in the prefrontal cortex,and alleviate cognitive and memory dysfunctions induced by binge-like ethanol treatment in adolescent *** the therapeutic role of the molecules contained in mesenchymal stem cell-derived extracellular vesicles,the present study analyzed whether the administration of mesenchymal stem cell-derived extracellular vesicles isolated from adipose tissue,which inhibited the activation of the NLRP3 inflammasome,was capable of reducing hippocampal neuroinflammation in adolescent mice treated with binge *** demonstrated that the administration of mesenchymal stem cell-derived extracellular vesicles ameliorated the activation of the hippocampal NLRP3 inflammasome complex and other NLRs inflammasomes(e.g.,pyrin domain-containing 1,caspase recruitment domain-containing 4,and absent in melanoma 2,as well as the alterations in inflammatory genes(interleukin-1β,interleukin-18,inducible nitric oxide synthase,nuclear factor-kappa B,monocyte chemoattractant protein-1,and C–X3–C motif chemokine ligand 1)and miRNAs(miR-21a-5p,miR-146a-5p,and miR-141-5p)induced by binge-like ethanol treatment in adolescent *** analysis further revealed the involvement of miR-21a-5p and miR-146a-5p with inflammatory target genes and NOD-like receptor signaling *** together,these findings provide novel evidence of the therapeutic potential of MSC-derived eVs to ameliorate the hippocampal neuroinflammatory response associated with NLRP3 inflammasome activation induced by binge drinking in adolescence.

Structure,stability,catalytic activity,and polarizabilities of small iridium clusters

At the second order Douglas–Kroll–Hess（DKH2） level, the B3 PW91 functional in conjunction with the relativistic all-electron basis set of valence triple zeta quality plus polarization functions are employed to compute bond lengths, dissociation energies, vertical ionization potentials, and the highest occupied molecular orbital-lowest unoccupied molecular orbital energy gaps of the small iridium clusters（Irn, n≤8）. These results are compared with the experimental and theoretical data available in the literature. Our results confirm the theoretical predictions made by Feng et al. about the catalytic activities of the Ir4 and Ir6 clusters. From the optimized geometries, DKH2 calculations of static electric mean dipole polarizabilities and polarizability anisotropies are also carried out. It is the first time that the polarizabilities of small iridium clusters have been studied. For n≤4, the mean dipole polarizabilities per atom present an odd–even oscillatory behavior,whereas from Ir5 to Ir8, they decrease with the cluster size increasing. The dependence of the polarizability anisotropy on the structure symmetry of the iridium cluster is verified.

Mechanisms for electric field induced color change in coupled colloidal quantum dot molecules revealed by low temperatures single particle spectroscopy

Colloidal quantum dots(QDs),the building blocks of modern displays and optoelectronic devices,have reached the highest level of size and shape control,and stability during the last 30 ***,full utilization of their potential requires integration or assembly of more than one nanocrystal as in the case of coupled quantum dots molecules(CQDM),where two core–shell QDs are fused to form two emission centers in close *** CQDMs were recently shown to switch color under an applied electric field at room *** we use cryogenic single particle spectroscopy of single CQDMs under an electric field to show that various mechanisms can contribute to the spectrum change under an applied electric field at cryogenic *** first mechanism is the control of the delocalized electron wave function when the electric field is applied along the dimer *** electric field bends the conduction band and forces the electron wave function to localize in one of the QDs yielding preferential emission of that particular *** addition,we found that QDs and CQDMs could become sensitive to surface traps under an electric *** the case of CQDMs,that can result in decreasing the intensity of one of the QDs while increasing the other QD’s ***,we show that there are surface charges which screen the applied electric field in some of the *** as well can result in electric field-induced color-tuning of *** the underlying mechanisms responsible for spectral shifts under applied electric fields is critical for the development of color-tunable devices utilizing CQDMs,including efficient displays and single photon sources.

Microstructural Morphologies and experimental Growth Laws during Solidification of Monotectic and Hypermonotectic Al-Pb Alloys

AI-Pb alloys with monotectic and hypermonotectic compositions were directionally solidified under unsteady- state heat flow conditions. The cooling curves recorded during solidification allowed solidification thermal parameters such as the cooling rate （T）, growth rate （v） and thermal gradient （G） to be experimentally determined. Different microstructural patterns have been associated with the alloy solute content, i.e., AI-1.2 and 2.1 wt% Pb. A sequence of morphologies from the bottom to the top of the A1-1.2 wt% Pb alloy casting （monotectic） can be observed： Pb-rich droplets in the aluminum-rich matrix, followed by a region of microstructural transition formed by droplets and fibers and finally by a mixture of fibers and strings of pearls. A completely fibrous structure （without transition） has been observed along the entire AI-2.1 wt% Pb alloy casting （hypermonotectic）. The interphase spacing （λ） was measured along the casting length, and experimental correlations between λand experimental solidification thermal parameters have been established. Power laws with a -2.2 exponent expressing λ as a function of the growth rate, v, were found to better represent the fibrous growth of both AI-Pb alloys. Moreover, a single experimental law expressing λ as a function of both G and v was found to describe the fibrous growth of both the monotectic and the hypermonotectic alloys experimentally examined.

Olfactory ensheathing glia promote axonal regeneration in the mammalian central nervous system,including retinal ganglion cell axonal growth through the injured optic ***,it is unknown whether olfactory ensheathing glia also have neuroprotective *** ensheathing glia express brain-derived neurotrophic factor,one of the best neuroprotectants for axotomized retinal ganglion ***,we aimed to investigate the neuroprotective capacity of olfactory ensheating glia after optic nerve *** ensheathing glia cells from an established rat immortalized clonal cell line,TeG3,were intravitreally injected in intact and axotomized retinas in syngeneic and allogeneic mode with or without microglial inhibition or immunosuppressive *** and gene expression analyses were *** bulb-derived primary olfactory ensheathing glia and TeG3 express major histocompatibility complex classⅡ*** and syngenically transplanted TeG3 cells survived in the vitreous for up to 21 days,forming an *** axotomized retinas,only the allogeneic TeG3 transplant rescued retinal ganglion cells at 7 days but not at 21 *** these retinas,microglial anatomical activation was higher than after optic nerve crush *** intact retinas,both transplants activated microglial cells and caused retinal ganglion cell death at 21 days,a loss that was higher after allotransplantation,triggered by pyroptosis and partially rescued by microglial inhibition or ***,neuroprotection of axotomized retinal ganglion cells did not improve with these *** different neuroprotective properties,different toxic effects,and different responses to microglial inhibitory treatments of olfactory ensheathing glia in the retina depending on the type of transplant highlight the importance of thorough preclinical studies to explore these variables.

Chasing the mechanisms of ecologically adaptive salinity tolerance

Plants adapted to challenging environments offer fascinating models of evolutionary change. Importantly,they also give information to meet our pressing need to develop resilient, low-input *** mounting environmental fluctuation—including temperature, rainfall, and soil salinity and degradation—this is more urgent than ever. Happily, solutions are hiding in plain sight: the adaptive mechanismsfrom natural adapted populations, once understood, can then be leveraged. Much recent insighthas come from the study of salinity, a widespread factor limiting productivity, with estimates of 20% ofall cultivated lands affected. This is an expanding problem, given increasing climate volatility, rising sealevels, and poor irrigation practices. We therefore highlight recent benchmark studies of ecologicallyadaptive salt tolerance in plants, assessing macro- and microevolutionary mechanisms, and therecently recognized role of ploidy and the microbiome on salinity adaptation. We synthesize insightspecifically on naturally evolved adaptive salt-tolerance mechanisms, as these works move substantiallybeyond traditional mutant or knockout studies, to show how evolution can nimbly ‘‘tweak’’ plantphysiology to optimize function. We then point to future directions to advance this field that intersectevolutionary biology, abiotic-stress tolerance, breeding, and molecular plant physiology.

Multiple primary colorectal cancer: Individual or familial predisposition?

Colorectal carcinoma(CRC) is one of the most frequent cancers. Along the surface of the large bowel, several foci of CRC may appear simultaneously or over the time. The development of at least two different tumours has been defined as multiple primary CRC(MPCRC):When more than one tumour is diagnosed at the same time, it is known as synchronous CRC(SCRC), while when a second neoplasm is diagnosed some time after the resection and/or diagnosis of the first lesion, it is called metachronous CRC(MCRC). Multiple issues can promote the development of MPCRC, ranging from different personal factors, such as environmental exposure, to familial predisposition due to hereditary factors. However, most studies do not distinguish this dichotomy. High- and low-pentrance genetic variants are involved in MPCRC. An increased risk for MPCRC has been described in Lynch syndrome, familial adenomatous polyposis, and serrated polyposis. Non-syndromic familial CRCs should also be considered as risk factors for MPCRC. environmental factors can promote damage to colon mucosae that enable the concurrence of MPCRC. epigenetics are thought to play a major role in the carcinogenesis of sporadic MPCRC. The methylation state of the DNA depends on multiple environmental factors(e.g., smoking and eating foods cooked at high temperatures), and this can contribute to increasing the MPCRC rate. Certain clinical features may also suggest individual predisposition for MPCRC. Different etiopathogenic factors are suspected to be involved in SCRC and MCRC, and different familial vs individual factors may be implicated. MCRC seems to follow a familial pattern, whereas individual factors are more important in SCRC. Further studies must be carried out to know the molecular basis of risks for MPCRC in order to modify, if necessary, its clinical management, especially from a preventive point of view.
