Treatment-related motor complications in Parkinson’s disease have been previously linked to disease-induced pre-synaptic alterations: dopaminergic denervation and changes in dopamine (DA) release patterns. The occurrence of such complications is also known to be partly dependent on the age of disease onset, occurring more frequently in patients with disease onset at a younger age. Using positron emission tomography (PET) and 4-h-long 18F-fluorodopa (FD) scans we have investigated in vivo an age dependence of disease-induced changes in DA turnover as a possible contributing factor to the agerelated differences in treatment-related motor complications. We evaluated the relative changes in DA turnover (measured by its direct inverse, effective DA distribution volume -EDV) and DA synthesis and vesicular storage capacity (quantified by the plasma input uptake rate constant Ki) in Parkinson’s disease patients as a function of age (n = 27, age range 38-79 years). After correcting for disease severity, a significant negative correlation was found between age and magnitude of disease-induced decrease in EDV and in Ki in the putamen (P decrease in EDV and that in Ki also exhibited an age dependence (P latively higher disease-induced increase in DA turnover (inverse of EDV) compared with the decrease in DA synthesis and storage rate in patients of younger age compared with older patients. This finding implies that DA turnover in younger-onset patients undergoes a relatively greater alteration and thus likely contributes to a greater imbalance between DA synthesis, storage and release, which could lead to larger swings in synaptic DA levels. It has indeed been suggested on theoretical grounds that such imbalance may contribute to the greater propensity for motor fluctuations. Diffuse large B-cell lymphoma(DLBCL)and follicular lymphoma(FL)are the most common forms of aggressive and indolent lymphoma,respectively.The majority of patients are cured by standard R-CHOP immunochemotherapy,but 30%–40%of DLBCL and 20%of FL patients relapse or are refractory(R/R).DLBCL and FL are phenotypically and genetically hereterogenous B-cell neoplasms.To date,the diagnosis of DLBCL and FL has been based on morphology,immunophenotyping and cytogenetics.However,next-generation sequencing(NGS)is widening our understanding of the genetic basis of the B-cell lymphomas.In this review we will discuss how integrating the NGS-based characterization of somatic gene mutations with diagnostic or prognostic value in DLBCL and FL could help refine B-cell lymphoma classification as part of a multidisciplinary pathology work-up.We will also discuss how molecular testing can identify candidates for clinical trials with targeted therapies and help predict therapeutic outcome to currently available treatments,including chimeric antigen receptor T-cell,as well as explore the application of circulating cell-free DNA,a non-invasive method for patient monitoring.We conclude that molecular analyses can drive improvements in patient outcomes due to an increased understanding of the different pathogenic pathways affected by each DLBCL subtype and indolent FL vs R/R FL.

The 2004 discovery of EGFR mutations,followed by ALK rearrangements,ushered in a targeted therapy era for advanced non-small cell lung cancer(NSCLC).Tyrosine kinase inhibitors targeting gene alterations have substantially improved survival and quality of life for patients with NSCLC.In the last decade,rearrangements of the ROS1 oncogene have been incorporated into healthcare practice that are applicable to another small subgroup of patients who benefit from similar targeted strategies.Recent genome studies of lung adenocarcinoma have identified other possible therapeutic targets,including RET,NTRK fusions,c-MET alterations,and activating mutations in KRAS,BRAF,and HER2,all with frequencies greater than 1%.Lung cancers harbouring these genome changes can potentially be treated with agents approved for other indications or under clinical development.This review updates the therapeutic arsenal that especially targets those genes.

El Soplao outcrop, an Early Cretaceous amber deposit recently discovered in northern Spain （Cantabria）, has been shown to be the largest site of amber with arthropod inclusions that has been found in Spain so far. Relevant data provided herein for biogeochemistry of the amber, palynology, taphonomy and arthropod bioinclusions complement those previously published. This set of data suggests at least two botanical sources for the amber of El Soplao deposit. The first （type A amber） strongly supports a source related to Cheirolepidiaceae, and the second （type B amber） shows non-specific conifer biomarkers. Comparison of molecular composition of type A amber with Frenelopsis leaves （Cheirolepidiaceae） strongly suggests a biochemical affinity and a common botanical origin. A preliminary palynological study indicates a regional high taxonomical diversity, mainly of pteridophyte spores and gymnosperm pollen grains. According to the preliminary palynological data, the region was inhabited by conifer forests adapted to a dry season under a subtropical climate. The abundant charcoalified wood associated with the amber in the same beds is evidence of paleofires that most likely promoted both the resin production and an intensive erosion of the litter, and subsequent great accumulation of amber plus plant cuticles. In addition, for the first time in the fossil record, charcoalified plant fibers as bioinclnsions in amber are reported. Other relevant taphonomic data are the exceptional presence of serpulids and bryozoans on the surfaces of some amber pieces indicating both a long exposure on marine or brackish-water and a mixed assemblage of amber. lastly, new findings of insect bioiuclusions, some of them uncommon in the fossil record or showing remarkable adaptations, are reported. In conclusion, a documented scenario for the origin of the El Soplao amber outcrop is provided.

Effect of rare-earth oxides on properties of silicon nitride obtained by normal sintering and sinter-HIP

This paper presented the microstructure, mechanical properties and oxidation behavior of silicon nitride obtained by both conventional sintering and sintering followed by hot isostatic pressing （HIP）. Silicon nitride with additives such as 5 wt.% Al2O3 and 5 wt.% Ln203 （Ln= la, la concentrate, Gd or la＋Gd） were studied. The results revealed that Gd203 additions increased the formation of elongated grains of β-Si3N4, the fracture toughness and oxidation resistance. la2O3 additions led to higher densification and hardness values, while addition of la2O3 concentrate promoted the formation of materials with intermediate properties, compared to the other studied compositions. Hot isostatic pressing increased the hardness but decreased the fracture toughness of the material, mainly because it allowed residual pores to close and also reduced the average aspect ratio of β-Si3N4 grains.

This article describes MSW(Municipal Solid Waste)management with APG(Arc Plasma Gasification)technology.First,a brief description of the process and reactor configurations is presented,listing some advantages and disadvantages.Then,alternatives for producing electrical energy with the residue of synthesis gas are addressed.Finally,this technology is proposed as an alternative to face the significant problem of eliminating MSW on a large scale.

Metadoxine improves the three- and six-month survival rates in patients with severe alcoholic hepatitis

AIM:To evaluate the impact of metadoxine(MTD) on the 3- and 6-mo survival of patients with severe alcoholic hepatitis(AH).METHODS:This study was an open-label clinical trial,performed at the"Hospital General de México,Dr.Eduardo Liceaga".We randomized 135 patients who met the criteria for severe AH into the following groups:35 patients received prednisone(PDN)40 mg/d,35patients received PDN+MTD 500 mg three times daily,33 patients received pentoxifylline(PTX)400 mg three times daily,and 32 patients received PTX+MTD 500 mg three times daily.The duration of the treatment for all of the groups was 30 d.RESULTS:In the groups treated with the MTD,thesurvival rate was higher at 3 mo(PTX+MTD 59.4%vs PTX 33.3%,P=0.04;PDN+MTD 68.6%vs PDN20%,P=0.0001)and at 6 mo(PTX+MTD 50%vs PTX18.2%,P=0.01;PDN+MTD 48.6%vs PDN 20%,P=0.003)than in the groups not treated with MTD.A relapse in alcohol intake was the primary independent factor predicting mortality at 6 mo.The patients receiving MTD maintained greater abstinence than those who did not receive it(74.5%vs 59.4%,P=0.02).CONCLUSION:MTD improves the 3-and 6-mo survival rates in patients with severe AH.Alcohol abstinence is a key factor for survival in these patients.The patients who received the combination therapy with MTD were more likely to maintain abstinence than those who received monotherapy with either PDN or PTX.

Unpaved rural roads as source areas of sediment in a watershed of the Brazilian semi-arid region

Approximately 80% of the road network in Brazil is unpaved and shows evidences of a high erosion potential. In the semi-arid Caatinga Biome in the northeast of the country, a monitoring programme has been done for two years in order to analyze runoff and sediment production from unpaved rural roadways and from embankments. Sediment production ranged from 0.30 to 0.92 Mg/ha yr, higher than in undisturbed areas, but generally lower than that reported for unpaved roads in other regions. However,this is a semi-arid area with low rainfall and runoff and, hence, with a limited hydrological connectivity and sediment production. Sediment production on an embankment with no vegetation was around ten times higher than on an embankment with vegetation. On the road surface, annual sediment production(normalized for gradient) in a section with traffic was three times higher than for a road surface without traffic. In addition, events that occurred after roadway maintenance activities generated peaks of sediment concentration of over 5000 mg/L. These results suggest that sediment production from roads and embankments with bare surfaces is at least one order of magnitude higher than in undisturbed catchment areas. Maintenance activity and vehicle traffic contribute to an increase in sediment availability and impact on the sediment concentration, but less intensely on sediment loads, which depend on the runoff magnitude of the events occurring after roadway maintenance. It was also found that the natural vegetation of the semi-arid region potentially captures sediment on roadway embankments; thereby playing an important role in breaking connectivity between the sediment flow from unpaved roads and the natural drainage system of the catchment.

Hereditary spastic paraplegia(HSP)is a clinically and genetically heterogeneous neurodegenerative disorder,characterized primarily by progressive spasticity and weakness in the lower limbs.Pat ients can also experience peripheral neuropathy,cognitive impairment,and other neurological symptoms.To date,more than 80 genes have been implicated in HSP,encompassing various cellular components,although mutations in genes encoding endoplasmic reticulum(ER)-shaping proteins are the most prevalent(Parodi et al.,2017).ER-shaping proteins are generally known for regulating the tubulation and curvation of the ER,but most of them show additional functions,including fusion of ER tubules,microtubule-severing,ER autophagy,lipid droplet synthesis,contact sites with other organelles(Öztürk et al.,2020).This highlights the complexity of studying the role of these proteins and the link between ER function and HSP.
