Phenolic profile and biological potential of Endopleura uchi extracts

Objective:To improve the knowledge on the metabolite composition of Endopleura uchi bark,concerning phenolic compounds,and to evaluate some of its biological capacities for further possible exploitation in food and pharmaceutical industries associated to their healthpromoting ***:Two different extracts(infusion and hydroethanolic) were studied concerning phenolic composition and biological ***:Five compounds were determined by HPLC-DAD,being bergenin the major *** general way,infusion presents a greater richness in these *** antioxidant,***,α-glucosidase and antibacterial activity were checked by in vitro assays.A dose-dependent response was noticed against DPPH,superoxide and nitric oxide radicals,acetylcholinesterase,butyrylcholinesterase and against α-glucosidase inhibitory *** capacity of both extracts was investigated against Gram-positive and Gramnegative bacteria,being more effective against the first *** concentrations of infusion extract tested here revealed that is non-toxic for intestinal(Caco-2) cells ***:The results suggest that the extracts of Endopleura uchi may be interesting for incorporating in pharmaceutical preparations for human health,since it can suppress hyperglycaemia and inhibit cholinesteases,and or as food additive due to its antiradical and antibacterial activities.

Rheological Characterization of a Mixed Fruit/Vegetable Puree Feedstock for Hydrogen Production by Dark Fermentation

Bio-hydrogen (Bio-H2) production from the organic fraction of solid waste, as fruit and vegetable wastes, constitutes an interesting and feasible technology to obtain clean energy. In spite of the feasibility to produce Bio-H2 from fruit/vegetable wastes (FVW), data about its rheological characterization are scarce. This information is useful to establish the hydrodynamic behavior, which controls the overall mixing process when the feedstock for Bio-H2 production process is a mixture of FVW. In this work, the rheological behavior of a vegetable/fruit waste mixture was characterized. The effect of the solids content (%, w/w), temperature, time (tyxotropy effects) and shear rate over the apparent viscosity of the mixture was evaluated. Most of the mixtures showed non-Newtonian behavior. The curves are typical rheofluidizing fluids. The rheological curves were different at increasing solids contents (80%, 60%, 40% and 30%), independent from the temperature. Rheological data were fitted to the power law model. Correlation factors R2 for the different mixtures were 0.991-0.995 for 80%, 0.961 -0.986 for 60%, 0.890 -0.925 for 40%. In the case of 30% of solids, the R2 value was not acceptable, and it was also found that this mixture was very near to the Newtonian behavior. Calculated activation energies (Ea) values were 15.98, 14.89 and 20.96 kJ/mol for the 80%, 60%, 40% mixtures, respectively. FVW purees rheological behavior was well characterized by Carbopol solutions at given concentrations and pH values. This fluid can be used as a model for other studies, e.g. LDA (Laser Doppler Anemometry) and PIV (Particle Image Velocimetry).

Dear Editor Abscisic acid （ABA） is the main phytohormone involved in adaptive crop responses to drought. ABA signaling relies on the family of pyrabactin resistance 1/PYRl-like/regulatory components of ABA receptors （PYR/PYL/RCAR）, which upon ABA binding form high-affinity ternary complexes with clade A protein phosphatases type 2C （PP2Cs） and inhibit them （Cutler et al., 2010）. This results in the activation of an SnRK2-dependent phosphorylation cascade affecting a high number of targets in the plant cells, regulating stomatal aper- ture and the expression of stress-responsive genes （Cutler et al., 2010）.

Objective:To investigate the vasorelaxant effect of organic extracts from Apium graveolens(***)which is a part of a group of plants subjected to pharmacological and phytochemical study with the purpose of offering it as an ideal source for obtaining lead compounds for designing new therapeutic agents with potential vasorelaxant and antihypertensive ***:An ex vivo method was employed to assess the vasorelaxant *** consisted of using rat aortic rings with and without endothelium precontracted with ***:All extracts caused concentration-dependent relaxation in precontracted aortic rings with and without endothelium;the most active extracts were Dichloromethane and Ethyl Acetate extracts from *** results suggested that secondary metabolites responsible for the vasorelaxant activity belong to a group of compounds of medium ***,our evidence showed that effect induced by dichloromethane and ethyl acetate extracts from *** is mediated probably by calcium ***:*** represents an ideal source for obtaining lead compounds for designing new therapeutic agents with potential vasorelaxant and antihypertensive effects.

Embrionary way to create a fatty liver in portal hypertension

Portal hypertension in the rat by triple partial portal vein ligation produces an array of splanchnic and systemic disorders, including hepatic steatosis. In the current review these alterations are considered components of a systemic inflammatory response that would develop through three overlapping phenotypes: The neurogenic, the immune and the endocrine. These three inflammatory phenotypes could resemble the functions expressed during embryonic development of mammals. In turn, the inflammatory phenotypes would be represented in the embryo by two functional axes, that is, a coelomic-amniotic axis and a trophoblastic yolk-sac or vitelline axis. In this sense, the inflammatory response developed after triple partial portal vein ligation in the rat would integrate both functional embryonic axes on the liver interstitial space of Disse. If so, this fact would favor the successive development of steatosis, steatohepatitis and fibrosis. Firstly, these recapitulated embryonic functions would produce the evolution of liver steatosis. In this way, this fat liver could represent a yolk-sac-like in portal hypertensive rats. After that, the systemic recapitulation of these embryonic functions in experimental prehepatic portal hypertension would consequently induce a gastrulationlike response in which a hepatic wound healing reaction or fibrosis occur. In conclusion, studying the mechanisms involved in embryonic development could provide key results for a better understanding of the nonalcoholic fatty liver disease etiopathogeny.

Transmission of cryptococcosis by liver transplantation:A case report and review of literature

BACKGROUND Cryptococcosis is a fungal infection caused by the yeast-like encapsulated basidiomycetous fungus of the Cryptococcus neoformans(***)species *** fungi are ubiquitous in soil and bird droppings,and infection by them is an important global health concern,particularly in immunosuppressed patients,such as organ transplant recipients and those infected by the human immunodeficiency *** fungus usually enters the body through the respiratory tract,but extremely rare cases of infection acquired by transplantation of solid organs have been *** SUMMARY We report a case of disseminated cryptococcosis in a liver transplant recipient,diagnosed 2 wk after the *** patient initially presented with fever,hyponatremia and elevated transaminase levels,manifesting intense headache after a few *** cultures were positive for *** biopsy showed numerous fungal elements surrounded by gelatinous matrix and sparse granulomatous *** resonance imaging of the brain showed multiple small lesions with low signal in T2,peripheric enhancement and edematous halo,diffuse through the parenchyma but more concentrated in the subcortical *** with amphotericin B for 3 wk,followed by maintenance therapy with fluconazole,led to complete resolution of the *** recipients of both kidneys from the same donor also developed disseminated cryptococcosis,confirming the transplant as the source of *** organ donor lived in a rural area,surrounded by tropical rainforest,and had negative blood cultures prior to organ *** This case highlights the risk of transmission of fungal diseases,specifically of ***,through liver graft during liver transplantation.

Amyloid-β (Aβ) can induce a chronic inflammatory immune response that is associated, amongst many others, to abnormal glycosylation, inducible nitric oxide synthase (iNOS) and nitric oxide (NO). The relation between development of Mild Cognitive Impairment (MCI) and Alzheimer’s disease progression and these serum markers has not been evaluated. Serum levels of iNOs, NO, TNF-α, IL-1β, IL-6, IL-8, IL-10, and IL-12 are determined with commercially available kits. Sialylation of albumin-free serum patterns is determined by Western blot analysis with Sambucus nigra (specific for sialic acid attached to terminal galactose in α2,6 linkage) lectin. Apolipoprotein E (ApoE) haplotype is determined by Western blot using specific anti-ApoE 2, 3 or 4 antibodies. A mini-mental state examination (MMSE) test is also performed in the 10 MCI patients, 19 Alzheimer’s disease (AD) patients and 46 healthy age-matched controls evaluated. The results show an increase of iNOS in MCI and AD but significantly higher NO concentrations are only found in MCI patients. TNF-α and IL-1β concentrations are the only significantly increased cytokines in MCI patients;no differences between control and MCI or AD patients are found in regard to the other cytokines. An abnormal MMSE test result only correlates with a decrease in serum NO concentration in MCI patients. The terminal sialic acid linkage pattern of serum proteins also shows highly significant differences between MCI and AD patient. ApoE3/4 or 4/4 haplotypes are characteristic of MCI and AD patients. Our results imply that increased serum TNF-α, IL-1β, iNOS, NO and alterations of serum proteins glycosylation patterns in adult individuals with an abnormal MMSE test may serve as an early biomarker of MCI and AD development.

Corticosteroids for severe influenza pneumonia: A critical appraisal

Influenza pneumonia is associated with high number of severe cases requiring hospital and intensive care unit(ICU) admissions with high mortality. Systemic steroids are proposed as a valid therapeutic option even though its effects are still controversial. Heterogeneity of published data regarding study design, population demographics, severity of illness, dosing, type and timing of corticosteroids administered constitute an important limitation for drawing robust conclusions. However, it is reasonable to admit that, as it was not found any advantage of corticosteroid therapy in so diverse conditions, such beneficial effects do not exist at all. Its administration is likely to increase overall mortality and such trend is consistent regardless of the quality as well as the sample size of studies. Moreover it was shown that corticosteroids might be associated with higher incidence of hospital-acquired pneumonia and longer duration of mechanical ventilation and ICU stay. Finally, it is reasonable to conclude that corticosteroids failed to demonstrate any beneficial effects in the treatment of patients with severe influenza infection. Thus its current use in s evere influenza pneumonia should be restricted to very selected cases and in the setting of clinical trials.

A systematic literature review of the management of chronic venous ulcers with autologous fibrin matrix with or without growth factors

Aim: To evaluate available evidence for the effective management of venous leg ulcers with fibrin matrix with or without growth factors. Methods: A systematic review of the literature was performed to evaluate the use of fibrin matrices with or without growth factors for the management of chronic venous ulcers in lower limbs. Article searches were performed in MEDLINE, EMBASE, COCHRANE, LILACS and ongoing clinical trials at ***. Results: The search in MEDLINE and EMBASE identified three articles;one was a pilot study evaluating the use of fibrin matrix and autologous growth factors that included patients with chronic ulcers of different etiology. The second article was a description of the product used in the previous study, and the third consisted of a series case reports of patient treated with cultured keratinocytes in a fibrin matrix. A COCHRANE searched resulted in one study assessing the cost effectiveness of using different fibrin matrices. The search in *** and LILACS did not result in any findings. Conclusion: The study did not provide a conclusive evidence for the use of fibrin matrices in patients with venous leg ulcers.
