Circulating CD133+ ste. ce.ls from the.pe.iphe.al blood have.be.n shown to be.able.to diffe.e.tiate.into nume.ous ce.l line.ge.. Howe.e., adults have.only a small numbe. of the.e.circulating ste. ce.ls. The.aim of the.pre.e.t study was to asse.s a ne. isolation and e.richme.t te.hnique.for CD133+ ste. ce.ls from pe.iphe.al blood with the.use.of Pe.coll de.sity gradie.ts. Our re.ults de.onstrate. the.pre.e.ce.of two large.mononucle.r bands whe. whole.blood was ce.trifuge. with 48% and 50% Pe.coll conce.trations. Flow cytome.ric analysis (FACS) re.e.le. a major CD133+ e.richme.t at the.48% Pe.coll conce.tration in one.of the.two bands. Furthe. culture.of the.e.ce.ls re.ulte. in the.formation of multiple.colony-forming units. Our re.ults sugge.t an advantage.from using a simple.Pe.coll gradie.t for succe.sful CD133+ ce.l re.ove.y, which could aid in diffe.e.tiation and transplantation protocols.
