Long-term treatment with PP2 after spinal cord injury resulted in functional locomotor recovery and increased spared tissue

The spinal cord has the ability to regenerate but the microenvironment generated after trauma reduces that capacity. An increase in Src family kinase （SFK） activity has been implicated in neuropathological conditions associated with central nervous system trauma. Therefore, we hypothesized that a decrease in SFK activation by a long-term treatment with 4-amino-5-（4- chlorophenyl）-7-（t-butyl）pyrazolo[3,4-d]pyramidine （PP2）, a selective SFK inhibitor, after spinal cord contusion with the New York University （NYU） impactor device would generate a permissive environment that improves axonal sprouting and/or behavioral activity. Results demonstrated that long-term blockade of SFK activation with PP2 increases locomotor activity at 7, 14, 21 and 28 days post-iniury in the Basso, Beattie, and Bresnahan open field test, round and square beam crossing tests. In addition, an increase in white matter spared tissue and serotonin fiber density was observed in animals treated with PP2. However, blockade of SFK activity did not change the astrocytic response or infiltration of cells from the immune system at 28 days post-injury. Moreover, a reduced SFK activity with PP2 diminished Ephexin （a guanine nudeotide exchange factor） phosphorylation in the acute phase （4 days post-injury） after trauma. Together, these findings suggest a potential role of SFK in the regulation of spared tissue and/or axonal outgrowth that may result in functional locomotor recovery during the pathophysiology generated after spinal cord injury. Our study also points out that ephexinl phosphorylation （activation） by SFK action may be involved in the repulsive microenvironment generated after spinal cord injury.

Areas of research and clinical approaches to the study of liver abscess

AIMTo analyze the research activity on liver abscess (LA) and identify the main topic clusters in the *** identified all documents using the medical subject heading “LA” indexed in the MEdLINE database between 2001 and 2015. We performed a descriptive bibliometric analysis, characterizing the evolution of scientific activity, the publication types of the documents, the document categories of clinical interest (case reports, clinical trials, evaluation studies, meta-analysis, observational studies, practice guidelines and validation studies) and the geographic distribution of the research. We also carried out an analysis of networks and research clusters in order to identify the main topic areas of *** search yielded a total of 1278 documents, showing a stable scientific production over the study period and a marked multidisciplinary nature. The research was dominated by case reports (65.9% of the documents analyzed). In terms of geographic distribution, researchers from the United States led in the number of signatures (n = 229), followed by those from Taiwan (n = 185), India (n = 145), Japan (n = 144), South Korea (n = 100), and China (n = 84). With regard to amebic LA, the top-producing countries were India and Mexico (n = 69 each), followed by the United States (n = 29). In the case of pyogenic LA, Taiwan Residents researchers led scientific production (n = 71), followed by the United States (n = 39) and China (n = 29). The most active areas of research in the field are diagnosis via computerized tomography scan, differential diagnosis with regard to liver cancer, treatment with antimicrobial agents, and Klebsiella infections (including bacteremia).CONCLUSIONClinical case reports associated with diagnosis and treatment are the main topic of study, highlighting the importance of this document type in advancing knowledge.

Objective： To evaluate the treatment effect of acupuncture on patients with idiopathic trigeminal neuralgia（ITN） by case-control longitudinal blinded study. Methods： Sixty ITN patients and 30 healthy subjects were included. The ITN patients were randomly assigned to acupuncture group（15 cases）, sham-acupuncture group（15 cases） and carbamazepine group（30 cases）, respectively. Clinical orofacial evaluation（including pain intensity and medication doses）, research diagnostic criteria for temporomandibular disorders（RdC/TMd） and Helkimo indexes（for functional evaluation of the masticatory system）, and quantitative sensory testing for sensory thresholds（gustative, olfactory, cold, warm, touch, vibration and superficial and deep pain） were evaluated before treatment, immediately after treatment, and 6 months after treatment. Results： The mean pain intensity by the Visual Analogue Scale only decreased in the acupuncture group at the last evaluation（P=0.012）. Patients in the sham-acupuncture group had an increase in carbamazepine doses according to the prescriptions（P〈0.01）. There was a reduction in secondary myofascial pain and mandibular limitations at the acupuncture and sham-acupuncture groups, however only the acupuncture group kept the changes after 6 months（P〈0.01, P=0.023）. There was a decrease in mechanical thresholds in the acupuncture group（tactile, P〈0.01; vibration, P=0.027） and an increase in deep pain thresholds in both acupuncture and sham-acupuncture groups（P=0.013）. Conclusion： Acupuncture can be an option in the treatment of ITN due to its analgesic effect in both ITN and secondary myofascial pain associated with it.

Magma chamber growth models in the upper crust: A review of the hydraulic and inertial constraints

Finite volumes of magma moving in confinement, store hydraulic potential energy for the generation,control and transmission of power. The Pascal's principle in a hydraulic jack arrangement is used to model the vertical and lateral growth of sills. The small input piston of the hydraulic jack is equivalent to the feeder dike, the upper large expansible piston equivalent to the magmatic chamber and the inertial force of the magma in the dike is the input force. This arrangement is particularly relevant to the case of sills expanding with blunt tips, for which rapid fracture propagation is inhibited. Hydraulic models concur with experimental data that show that lateral expansion of magma into a sill is promoted when the vertical ascent of magma through a feeder dike reaches the bottom contact with an overlying, flat rigid-layer. At this point, the magma is forced to decelerate, triggering a pressure wave through the conduit caused by the continued ascent of magma further down(fluid-hammer effect). This pressure wave can provide overpressure enough to trigger the initial hydraulic lateral expansion of magma into an incipient sill, and still have enough input inertial force left to continue feeding the hydraulic system. The lateral expansion underneath the strong impeding layer, causes an area increase and thus, further hydraulic amplification of the input inertial force on the sides and roof of the incipient sill, triggering further expansion in a self-reinforcing process. Initially, the lateral pressure increase is larger than that in the roof allowing the sill to expand. However, expansion eventually increases the total integrated force on the roof allowing its uplift into either a laccolith, if the roof preserves continuity, or into a piston bounded by a circular set of fractures. Hydraulic models for shallow magmatic chambers, also suggest that laccolith-like intrusions require the existence of a self-supported chamber roof. In contrast, if the roof of magmatic cham

Evaluating methods to quantify sediment volumes trapped behind check dams,Saldana badlands(Spain)

The determination of sediment yield in catchments based on the sediment trapped by check dams is becoming a subject of *** fact,several methods have been developed in recent years to estimate the sediment retained by check *** complexity,precision and accuracy of each method vary *** this study,we evaluate the sediment trapped by check dams comparing the Sections method respect to the Prism,Pyramid,dTMs and Trapezoid *** analyzed a sample of 25 check dams(α-1=90%;ε=10%) in the Saldana badlands(Spain).The results showed that the Sections method offered a volume of retained sediment between the others,which gave an absolute variation from 22%to40%.The high variability of the check dam and sediment wedge sizes made necessary to compare methods in groups combining both *** significant differences in sediment volumes could be found between the methods for very small(height(h):1.8-2.3 m;trapped volume(V):6-102 m3) or large check dams(h > 2.3 m;V:165-387 m3),while significant differences are found for small(h ddle-sized check dams(h:2.2-3.2 m;V:65-235 m3).Nevertheless,volume differences between groups ranged up to + 25%.For these reason,the size of the check dam,the shape of the sediment wedge and the accuracy of the measure methods must considered when selecting an appropriate method to obtain the volume of retained sediment by check dams.A correct estimation of the sediment retention is needed to evaluate the role and efficiency of check dams in restoration projects or to estimate sediment yields.

Over the past few years,the development of nonfullerene acceptors(NFAs)has become a prominent focus in both organic and perovskite solar cell(OSCs and PSCs,respectively)research *** this context,porphyrinoids,compounds structurally related to porphyrins,have emerged as promising solar cell *** contrast to the widely used fullerene acceptors,porphyrinoids exhibit strong,broad absorption properties across the UV–vis/NIR spectrum,which can be easily tuned through chemical ***,they can be prepared and derivatized using cost-effective and straightforward methodologies,allowing for convenient adjustments in thin-film morphology,processability,supramolecular organization,and energy ***,these compounds offer higher thermal and photochemical stability,resulting in longer device lifetimes compared to their fullerene-based *** this review,we outline the utilization of porphyrinoids as NFAs in OSCs and PSCs,discussing essential aspects such as design guidelines,molecular properties,and device *** goal is to inspire and further promote the development of n-type porphyrinoids,which have not yet fully unleashed their potential.

Stem cell therapy in the management of shoulder rotator cuff disorders

Rotator cuff tears are frequent shoulder problems that are usually dealt with surgical repair. despite improved surgical techniques, the tendon-to-bone healing rate is unsatisfactory due to difficulties in restoring the delicate transitional tissue between bone and tendon. It is essential to understand the molecular mechanisms that determine this failure. The study of the molecular environment during embryogenesis and during normal healing after injury is key in devising strategies to get a successful repair. Mesenchymal stem cells(MSC) can differentiate into different mesodermal tissues and have a strong paracrine, anti-inflammatory, immunoregulatory and angiogenic potential. Stem cell therapy is thus a potentially effective therapy to enhance rotator cuff healing. Promising results have been reported with the use of autologous MSC of different origins in animal studies: they have shown to have better healing properties, increasing the amount of fibrocartilage formation and improving the orientation of fibrocartilage fibers with less immunologic response and reduced lymphocyte infiltration. All these changes lead to an increase in biomechanical strength. However, animal research is still inconclusive and more experimental studies are needed before human application. Future directions include expanded stem cell therapy in combination with growth factors or different scaffolds as well as new stem cell types and gene therapy.
