Fermented chile pepper ma.h （Ca.sicum a.nuum cv. Mesilla.Ca.enne） is a.ma.or industria. food product in New Mexico. The fermenta.ion of chile pepper ma.h depends on tempera.ure, a.idity, sa.t concentra.ion, dissolved a.r, a.a.la.le ca.bohydra.es a.d enzymes. The objective of this study wa. to eva.ua.e the effects of the a.dition of ca.cium chloride （Ca.I2） on the microbia. cha.a.teristics of the pepper ma.h fermenta.ion. Nine five ga.lon buckets were prepa.ed with pepper tha. ha. been wa.hed a.d ground by the ma.ufa.turer, ea.h with 15% sodium chloride a.ded. The buckets were a.lotted ra.domly to 1 of 3 trea.ments. The trea.ments included no Ca.l2, 0.2% Ca.l2 a.d 0.4% Ca.l2 a.ded to the pepper ma.h. Sa.ples were stored a. room tempera.ure a.d sa.pled over a.3 months period to exa.ine cha.ges in the microflora. Chile ma.h sa.ples were seria.ly diluted in butterfield＇s phospha.e buffer for microbia. enumera.ion. a.robic pla.e counts were conducted by pour pla.ing with a. overla.. Petrifilms were used for E. coil/coliform counts a.d Enteroba.teria.ea.. Yea.ts a.d molds were surfa.e pla.ed on Rose Benga. Chlora.phenicol a.a. while streptococci a.d la.toba.illi were pla.ed onto M17 a.d a.idified MRS respectively. a.robic pla.e counts, coliforms, E. coli, Enteroba.teria.ea., yea.t, mold, la.toba.illi, a.d streptococci were not a.fected by Ca.l2 level. The a.dition of 0.4% Ca.l2 wa. found to ha.e no effect on the chile pepper ma.h fermenta.ion.

Fermented chile pepper ma.h is a.ma.or food product in New Mexico. There a.e few reports on the fermenta.ion process or on methods to monitor it, In the current study we exa.ined a.pour pla.e procedure with a. overla. using pla.e count a.a. a.d 3 MTM PetrifilmTM a.robic Count （a.） pla.es for determina.ion of tota. a.robic ba.teria. counts during the fermenta.ion of chile ma.h. Fifty chile ma.h sa.ples were obta.ned directly from commercia. fermenta.ion va.s a.d exa.ined within 2 h of collection. Seria. dilutions of the chile ma.h were prepa.ed in Butterfield＇s Phospha.e Buffer. 1 mL portions of the diluted sa.ples were a.iquoted in duplica.e onto the a. pla.es a.d into empty Petri dishes. Pla.e count a.a. wa. poured a.d once the pla.es ha. solidified, they were overla.d with a.out 10 mL of PCa.to minimize sprea.ers. Pla.es were incuba.ed a. 30 ℃ for 48 h a.d enumera.ed. Pa.red difference tests were conducted on log tra.sformed da.a.to compa.e the results of the two pla.ing procedures. For commercia. chile ma.h sa.ples, we did not show a.y significa.t differences between the a. pla.e counts a.d the pour pla.e counts （α = 0.05）, 3 MTM PetrifilmTM a. pla.es a.e a.good a.terna.ive to pour pla.es for the determina.ion of the tota. a.robic counts in fermented chile ma.hes.
