In plants, ca^2＋ signals occur in response to both environmental and developmental stimuli. These signals involve increases in [ca^2＋] in compartments such as the cytoplasm, nucleus, and mitochondria, and can display multiple shapes ranging from sin- gle transients to repetitive ca^2＋ oscillations. Generation and shaping of stimulus specific [ca^2＋] signatures depends on ca^2＋ influxes and effluxes occurring at both the plasma membrane （PM） and membranes of the different subcellular compartments.

Aim: Although epidermal growth factor receptors are expressed in the testes, whether they signal through epidermal growth factor receptor pathway substrate 8 (Eps8) is unknown. Here we evaluated the expression pattern of Eps8 in the maturing testis. Methods: The expression of Eps8 was analysed by Northern blotting, immunocy-tochemistry and Western blotting in primary Sertoli cell cultures and in testicular tissue of rodents. Results: Eps8 is specifically expressed in gonocytes, Leydig and Sertoli cells of the neonatal rats and in Leydig and Sertoli cells of the adult rats and mice. Although gonocytes express Eps8, no signal was found in prepubertal or mature spermatogonia and the expression level of Eps8 in Sertoli cells increases with age. No regulation of Eps8 expression in primary immature rat Sertoli cells by Follicle stimulating hormone (FSH) was detected by Western blotting. conclusion: Eps8 seems to be involved in the growth factor-controlled regulation of cell proliferation and differentiation in the seminiferous epithelium. Eps8 is a possible marker for gonocytes and in Sertoli cells it could be involved in crosstalk with other growth factor pathways.

Thermal profile modification of an active material in a laser amplifier via optical pumping results in a change in the material’s refractive index,and causes thermal expansion and stress,eventually leading to spatial phase aberrations,or even permanent material *** this purpose,knowledge of the 3D spatio-temporal thermal profile,which can currently only be retrieved via numerical simulations,is critical for joule-class laser amplifiers to reveal potentially dangerous thermal features within the pumped active *** this investigation,a detailed,spatio-temporal numerical simulation was constructed and tested for accuracy against surface thermal measurements of various endpumped Yb^3+-doped laser-active *** measurements and simulations show an excellent agreement and the model was successfully applied to a joule-class Yb3+-based amplifier currently operating in the POLARIS laser system at the Friedrich-Schiller-University and Helmholtz-Institute Jena in Germany.

We present integrated-optic building blocks and functional photonic devices based on amorphous siliconon-insulator technology. Efficient deep-etched fiber-to-chip grating couplers, low-loss single-mode photonic wire waveguides, and compact power splitters are presented. Based on the sub-μm photonic wires, 2 × 2 Mach–Zehnder interferometers and add/drop microring resonators(MRRs) with low device footprints and high finesse up to 200 were realized and studied. compact polarization rotators and splitters with ≥10 d B polarization extinction ratio were fabricated for the polarization management on-chip. The tuning and trimming capabilities of the material platform are demonstrated with efficient microheaters and a permanent device trimming method, which enabled the realization of energy-efficient photonic circuits. Wavelength multiplexers in the form of cascaded filter banks and 4 × 4 routers based on MRR switches are presented. Fabrication imperfections were analyzed and permanently corrected by an accurate laser-trimming method, thus enabling eight-channel multiplexers with record low metrics of sub-m W static power consumption and ≤1°c temperature overhead. The high quality of the functional devices, the high tuning efficiency, and the excellent trimming capabilities demonstrate the potential to realize low-cost, densely integrated, and ultralow-power 3D-stacked photonic circuits on top of cMOS microelectronics.

Alpine Activity Patterns of Mitopus morio(Fabricius, 1779) are Induced by Variations in Temperature and Humidity at Different Scales in central Norway

Our research addresses questions about how micro-climate affects activity abundance of a common and widespread harvestman in an alpine ecosystem. Activity patterns of the Harvestman Mitopus morio(Fabricius, 1779) were studied along different alpine gradients in the central Norwegian Scandes. Within a nested design, we surveyed 18 alpine habitats with pitfall traps and microclimatological equipment along oceanic-continental, two elevational, and(fine-scaled) microtopographic gradients. Sites in the oceanic region of the Scandes showed generally higher abundance of M. morio than sites in the continental region. Furthermore, along the elevational gradient, middle-alpine sites showed higher abundances than low-alpine sites. These general patterns are best explained by higher humidity in the oceanic region and in the middlealpine belt. Focusing at a finer scale, i.e. one elevational level within each region, revealed partly opposing activity patterns within relatively short distances. While in the western middle-alpine belt these patterns were best explained by humidityrelated measures but now with higher activity abundance during drier conditions, in the drier eastern middle-alpine belt heat sums rather than humidity were found to be the best explanatoryvariables for the observed patterns. Hence, our results imply a pronounced different reaction of the two populations towards climatic variables that partly even contradict the previously described general pattern. Regardless whether these differences in activity abundance in M. morio are a form of phenotypic plasticity or adaptation, our findings stress the importance of detailed autecological knowledge combined with fine-scaled climatic measurements when aiming at predictions about possible future ecosystem structures and spatiotemporal phenomena. M. morio proves to be an ideal biogeographic model organism for understanding spatio-temporal responses of alpine ecosystems under modified climatic conditions.

complex protein interaction networks constitute plant metabolic and signaling systems. Bimolecular fluorescence complementation （BiFc） is a suitable technique to investigate the formation of protein complexes and the localization of protein-protein interactions in planta. However, the generation of large plasmid collections to facilitate the exploration of complex interaction networks is often limited by the need for conventional cloning techniques. Here, we report the implementation of a GATEWAY vector system enabling large-scale combination and investigation of candidate proteins in BiFc studies. We describe a set of 12 GATEWAY-compatible BiFc vectors that efficiently permit the combination of candidate protein pairs with every possible N-or c-terminal sub-fragment of S（cFP）3A or Venus, respectively, and enable the performance of multicolor BiFc （mcBiFc）. We used proteins of the plant molybdenum metabolism, in that more than 20 potentially interacting proteins are assumed to form the cellular molybdenum network, as a case study to establish the functionality of the new vectors. Using these vectors, we report the formation of the molybdopterin synthase complex by interaction of Arabidopsis proteins cnx6 and cnx7 detected by BiFc as well as the simultaneous formation of cnx6/cnx6 and cnx6/cnx7 complexes revealed by mcBiFc. consequently, these GATEWAY-based BiFc vector systems should significantly facilitate the large-scale investigation of complex regulatory networks in plant cells.

Herein,a one-step co-pyrolysis protocol was adopted for the first time to prepare a novel pyrogenic carbon-cu^(0)/Fe_(3)O_(4)heteroatoms (FcBc) in cO_(2)ambiance to discern the roles of each component in PDS *** co-pyrolysis,cO_(2)catalyzed formation of reducing gases by biomass which facilitated reductive transformation of Fe^(3+)and cu^(2+)to cu^(0)and Fe_(3)O_(4),*** to the analysis,the resulting metal (oxide) catalyzed graphitization of biocharand decomposition of volatile substances resulting in an unprecedented surface area (1240 m^(2)/g).The resulting FcBc showed greater structural defects and less electrical *** experiments indicated that Rhodamine B (RhB) degradation by FcBc (100%) was superior to Fe_(3)O_(4)(50%) and cu^(0)/Fe_(3)O_(4)(76.4%) in persulfate (PDS) system,which maintained reasonable efficiency (75.6%-63.6%) within three *** reactive oxygen species (ROS) associated with RhB degradation was identified by an electron paramagnetic resonance and confirmed by scavenging *** degradation invoked both(sulfate and dominantly hydroxyl) radical and non-radical (singlet oxygen,^(1)O_(2)) *** FcBc,cu^(0)can continuously react with Fe^(3+)in Fe_(3)O_(4)to generate larger quantities of Fe^(2+),and both cu^(0)and Fe^(2+)activated PDS to yield sulfate radicals which was quickly converted to hydroxyl ***,cu^(0)/cu^(2+)could complex with PDS to form a metastable complex,which particularly contributed to1O_(2)*** cascade reactions by FcBc were reinforced by carbonyl group of biochar and favorable electron transfer *** work highlighted a new approach to prepare a magnetic and environment-benign heterogonous catalyst to remove organic pollutants in water.

Organellar biogenesis is mainly regulated by nucleus-encoded factors, which act on various steps of gene expression including RNA editing, processing, splicing, stabilization, and translation initiation. Among these regulatory factors, pentatricopeptide repeat （PPR） proteins form the largest family of RNA binding proteins, with hundreds of members in flowering plants. In striking contrast, the genome of the unicellular green alga chlamydomonas reinhardtii encodes only 14 such proteins. In this study, we analyzed PPR7, the smallest and most highly expressed PPR protein in c. reinhardtii. Green fluorescent protein-based localization and gel-filtration analysis revealed that PPR7 forms a part of a high-molecular-weight ribonu- cleoprotein complex in the chloroplast stroma. RIP-chip analysis of PPRT-bound RNAs demonstrated that the protein associates with a diverse set of chloroplast transcripts in vivo, i.e. rrnS, psbH, rpoc2, rbcL, atpA, cemA-atpH, tscA, and atpl-psaJ. Furthermore, the investigation of PPR7 RNAi strains revealed that deple- tion of PPR7 results in a light-sensitive phenotype, accompanied by altered levels of its target RNAs that are compatible with the defects in their maturation or stabilization. PPR7 is thus an unusual type of small multi- functional PPR protein, which interacts, probably in conjunction with other RNA binding proteins, with numerous target RNAs to promote a variety of post-transcriptional events.
