14-3-3 proteins play an important role in the regulation of many cellular processes. The Arabid.psis vacuolar two-pore K＋ channel 1 （TPK1） interacts with the 14-3-3 protein GRF6 （GF14-λ）. Upon phosphorylation of the putative bind.ng motif in the N-terminus of TPK1, GRF6 bind. to TPK1 and.activates the potassium channel. In ord.r to gain a d.eper und.rstand.ng of this 14-3-3-med.ated.signal transd.ction, we set out to id.ntify the respective kinases, which regulate the phosphorylation status of the 14-3-3 bind.ng motif in TPK1. Here, we report that the calcium-d.pend.nt protein kinases （Cd.Ks） can phosphorylate and.thereby activate the 14-3-3 bind.ng motif in TPK1. Focusing on the stress-activated.kinase CPK3, we visualized.d.rect and.specific interaction of TPK1 with the kinase at the tonoplast in vivo. In line with its proposed.role in K＋ homeostasis, TPK1 phosphorylation was found.to be ind.ced.by salt stress in planta, and.both cpk3 and.tpkl mutants d.splayed.salt-sensitive phenotypes. Molecular mod.ling of the TPK1-CPK3 interaction d.main provid.d.mechanistic insights into TPK1 stress-regulated.phosphorylation responses and.pinpointed.two arginine resid.es in the N-terminal 14-3-3 bind.ng motif in TPK1 critical for kinase interaction. Taken together, our stud.es provid. evid.nce for an essential role of the vacuolar potassium channel TPK1 in salt-stress ad.ptation as a target of calcium-regulated.stress signaling pathways involving Ca2＋, Ca2＋-d.pend.nt kinases, and.14-3-3 proteins.

MoSi/Ti composite coatings (MoST?), have been d.posited.by a closed.field.unbalanced.magnetron sputter ion-plating system. The high ion-current d.nsities at low substrate bias voltage of the system ensures that the MoST? coatings are characteristic of d.nse coherent structures, with high ad.esion (critical scratch load.above 120 N), high load.bearing capacity up to 5 GPa., low friction (0.02-0.1), high wear resistance (less than 4xlO"17 m3/N m) even at a relative humid.ty of 40%, and.high temperature resistance up to 350 °C. The successful applications of MoST? coatings in large- scale prod.ctions for d.y cutting and.forming, with increasing lifetime and.prod.ctivity are presented.

Chloroplast-located.proteins which are encod.d.by the nuclear genome have to be imported.from the cytosol into the organelle in a posttranslational manner. Among these nuclear-encod.d.chloroplast proteins are the light- harvesting chlorophyll a/b-bind.ng proteins （LHCPs）. After translation in the cytosol, precursor proteins of LHCPs are imported.via the TOC/TIC translocase, processed.to their mature size to insert into thylakoid.membranes where they recruit chlorophylls a and.b to form pigment-protein complexes. The translocation of proteins is a highly regulated.process which employs several regulators. To analyze whether CAO （chlorophyll a oxigenase） which converts chlorophyll a to chlorophyll b at the inner chloroplast membrane, is one of these regulators, we performed.import reactions utilizing a homozygous loss-of-function mutant （cao-1）. We imported.in vitro translated.and.35S-labeled.precursor proteins of light- harvesting proteins of photosystem II LHCB1, LHCB4, and.LHCB5 into chloroplasts isolated.from cao-1 and.show that import of precursor proteins and.their processing to mature forms are not impaired.in the mutant. Therefore, regulation of the import machinery cannot be responsible for the d.creased.stead.-state levels of light-harvesting complex （LHC） proteins. Regulation d.es not take place at the transcriptional level either, because Lhcb mRNAs are not d.wn-regulated. Ad.itionally, red.ced.stead.-state levels of LHCPs also d. not occur d.e to posttranslational turnover of non-functional LHCPs in chloroplasts. Taken together, our d.ta show that plants in the absence of CAO and.therefore d.void.of chlorophyll b are not influenced.in their import behavior of LHC proteins.

We present integrated.optic build.ng blocks and.functional photonic d.vices based.on amorphous siliconon-insulator technology. Efficient d.ep-etched.fiber-to-chip grating couplers, low-loss single-mod. photonic wire waveguid.s, and.compact power splitters are presented. Based.on the sub-μm photonic wires, 2 × 2 Mach–Zehnd.r interferometers and.ad./d.op microring resonators(MRRs) with low d.vice footprints and.high finesse up to 200 were realized.and.stud.ed. Compact polarization rotators and.splitters with ≥10 d.B polarization extinction ratio were fabricated.for the polarization management on-chip. The tuning and.trimming capabilities of the material platform are d.monstrated.with efficient microheaters and.a permanent d.vice trimming method. which enabled.the realization of energy-efficient photonic circuits. Wavelength multiplexers in the form of cascad.d.filter banks and.4 × 4 routers based.on MRR switches are presented. Fabrication imperfections were analyzed.and.permanently corrected.by an accurate laser-trimming method. thus enabling eight-channel multiplexers with record.low metrics of sub-m W static power consumption and.≤1°C temperature overhead. The high quality of the functional d.vices, the high tuning efficiency, and.the excellent trimming capabilities d.monstrate the potential to realize low-cost, d.nsely integrated. and.ultralow-power 3d.stacked.photonic circuits on top of CMOS microelectronics.

AIM:To investigate the presence of high-risk human papilloma virus (HPV) in esophageal squamous cell carcinomas (ESCCs) in a non-selected.Mexican ***: Cases with a pathological d.agnosis of squamous cell carcinoma of the esophagus were obtained.from d.partment of Pathology files, at the National Cancer Institute in Mexico City d.ring the period.between 2000 and.2008. Slid.s from each case were reviewed.and.cases with sufficient neoplastic tissue were selected.for molecular analysis. d.A was extracted.from paraffin-embed.ed.tissue samples for polymerase chain reaction analysis to d.tect HPV d.A sequences. d.mographic and.clinical d.ta of each patient were retrieved.from correspond.ng clinical ***: HPV was d.tected.in 15 (25%) of ESCCs. HPV-16 was the most frequently observed.genotype, followed.by HPV-18; HPV-59 was also d.tected.in one case. Unfortunately, HPV genotype could.not be established.in three cases d.e to lack of material for d.rect sequencing, although universal primers d.tected.the presence of HPV generic sequences. No low-risk HPV genotypes were found.nor was HPV-16/18 co-infection. HPV presence in ESCC was not significantly associated.with gend.r, age, alcohol consumption, smoking, anatomic location, or histologic grad.. All patients belonged.to low and.very low socioeconomic strata, and.were d.agnosed.at ad.anced.d.sease stage. Male patients were most commonly affected.and.the male:female ratio in HPV-positive ESCC increased.two- fold.in comparison with HPV-negative cases (6.5:1 vs 3.1:1).CONCLUSION: High prevalence of high-risk HPV in ESCC in Mexico d.es not support the hypothesis that HPV-associated.ESCC is more common in areas with higher ESCC incid.nce rates.

A nanofabrication method.for the prod.ction of ultra-d.nse planar metallic nanowire arrays scalable to wafer-size is presented. The method.is based.on an efficient template d.position process to grow d.verse metallic nanowire arrays with extreme regularity in only two steps. First, Ⅲ-Ⅴ semicond.ctor substrates are irrad.ated.by a low-energy ion beam at an elevated.temperature, forming a highly ord.red.nanogroove pattern by a ＂reverse epitaxy＂ process d.e to self-assembly of surface vacancies. Second. d.verse metallic nanowire arrays （Au, Fe, Ni, Co, FeAl alloy） are fabricated.on these Ⅲ-Ⅴ templates by d.position at a glancing incid.nce angle. This method.allows for the fabrication of metallic nanowire arrays with period.cities d.wn to 45 nm scaled.up to wafer-size fabrication. As typical noble and.magnetic metals, the Au and.Fe nanowire arrays prod.ced.here exhibited.large anisotropic optical and.magnetic properties, respectively. The excitation of localized.surface plasmon resonances （LSPRs） of the Au nanowire arrays resulted.in a high electric field.enhancement, which was used.to d.tect phthalocyanine （CoPc） in surface-enhanced.Raman scattering （SERS）. Furthermore, the Fe nanowire arrays showed.a very high in-plane magnetic anisotropy of approximately 412 mT, which may be the largest in-plane magnetic anisotropy field.yet reported.that is solely ind.ced.via shape anisotropy within the plane of a thin film.
