Unilateral 6-OHDA <i>th-fgfr1</i>(<i>tk.</i>) mouse model supports the role of FGFs in Park.nson’s disease and the effects of nicotine and L-DOPA on spontaneous motor impairments

In the developing and adult brain, neurotrophic growth factors support the growth and protec tion of dopaminergic neuronal systems. Recently, link. between impaired neurotrophin support of dopamine (DA) neurons has been described in Park.nson’s Disease (PD). Fibro- blast growth factor (FGF) has a unique association with DA neurons in that FGF signaling is vitally important for the development and protection of adult DA neurons. We assessed the role of substantia nigra (SN)-expressed FGFs in the nigrostriatal dopaminergic system using a transgenic mouse, th-fgfr1(tk.). In these mice, generated by expression of dominant negative FGFR1(Tk.) from the tyrosine hydroxylase (TH) gene promoter, reduced FGF signaling results in smaller and less dense adult nigrostriatal DA neurons, similar to what is observed in PD. With unilateral 6-hydroxydopamine (6-OHDA) lesions, th-fgfr1(tk.) mice exhibited extensive unilateral nigrostriatal damage with robust spontaneous (non-drug induced) asymmetrical turning and a decreased latency to remain on the accelerating rotarod. L-DOPA remains the gold standard for PD therapy despite debilitating hyperk.netic and dysk.netic side effects. The nicotinic acetylcholine system has recently been targeted as an alternative system to combat PD motor symptoms. Nicotine effectively stimulates dopaminergic transmission in the nigrostriatal pathway and mediates movement. Using unilaterally lesioned th-fgfr1(tk.) mice, long term (11 day) oral administration of nicotine increased spontaneous bidirectional turning and increased the latency before falling from the accelerating rotarod. In a separate analysis, L-DOPA treatment reversed directionality of rotation and further deepened motor discoordination, suggesting activation of hypersensitive postsynaptic DA receptors in the denervated striata. These results in a transgenic model of PD provide insights into the cellular mechanisms underlying L-DOPA and nicotinic therapies and offer further evidence of nicot

Post-operative pancreatic fistula(POPF)is a common and dangerous complication of pancreatic resection,occurring in 5-30%of *** is a significant source of morbidity and mortality,leading to prolonged hospital stays and increased healthcare costs(1).The most widely accepted definition of POPF comes from the International Study Group on Pancreatic Fistula(ISGPF).Initially created in 2005,this classification system for POPF was revised in 2016 such that POPF should be associated with a clinically relevant change in status,deeming what was originally defined as a Grade A fistula as a biochemical leak.and grade B and C fistulas as clinically relevant(CR)fistulae(Table 1)(2).

Drought is a recurring phenomenon that puts crop yields at risk.and threatens the livelihoods of many people around the ***-green is a drought adaption phenotype found in sorghum and other *** expressing this phenotype show less drought-induced senescence and maintain functional green leaves for longer when water limitation occurs during grain fill,conferring benefits in both yield per se and *** physiological causes of the phenotype are postulated to be water saving through mechanisms such as reduced canopy size or access to extra water through mechanisms such as deeper *** sorghum breeding programs,stay-green has traditionally been assessed by comparing visual scores of leaf senescence either by identifying final leaf senescence or by estimating rate of leaf *** this study,we compared measurements of canopy dynamics obtained from remote sensing on two sorghum breeding trials to stay-green values(breeding values)obtained from visual leaf senescence ratings in multienvironment breeding trials to determine which components of canopy development were most closely link.d to the stay-green ***,canopy size as estimated using preflowering canopy parameters was weak.y correlated with stay-green values for leaf senescence while postflowering canopy parameters showed a much stronger association with leaf *** study suggests that factors other than canopy size have an important role in the expression of a stay-green phenotype in grain sorghum and further that the use of UAVs with multispectral sensors provides an excellent way of measuring canopy traits of hundreds of plots grown in large field trials.

The use of thiopurines in inflammatory bowel disease(IBD) has been examined in numerous prospective, controlled trials, with a majority demonstrating a clinical benefit. We conducted this review to describe the historical and current evidence in the use of thiopurines in IBD. A systematic search was performed on MEDLINE between 1965 and 2016 to identify studies on thiopurines in IBD. The most robust evidence for thiopurines in IBD includes induction of remission in combination with anti-tumor necrosis factor(antiTNF) agents, and maintenance of remission and postoperative maintenance in Crohn's disease. Less evidence exists for thiopurine monotherapy in induction of remission, maintenance of ulcerative colitis, chemoprevention of colorectal cancer, and in preventing immunogenicity to anti-TNF. Evidence was often limited by trial design. Overall, thiopurines have demonstrated efficacy in a broad range of presentations of IBD. With more efficacious novel therapeutic agents, the positioning of thiopurines in the management of IBD will change and future studies will analyze the benefit of thiopurines alone and in conjunction with these new medications.
