Comprehensive molecular characterization to predict immunotherapy response in advanced biliary tract cancer: a phase II trial of pembrolizumab

Background:Immune checkpoint inhibitors (ICIs) are effective in a subset of patients with metastatic solid tumors. However, the patients who would benefit most from ICIs in biliary tract cancer (BTC) are still controversial.Materials and methods:We molecularly characterized tissues and blood from 32 patients with metastatic BTC treated with the ICI pembrolizumab as second-line therapy.Results:All patients had microsatellite stable (MSS) type tumors. Three of the 32 patients achieved partial response (PR), with an objective response rate (ORR) of 9.4% (95% confidence interval [CI], 2.0-25.2) and nine showed stable disease (SD), exhibiting a disease control rate (DCR) of 37.5% (95% CI, 21.1-56.3). For the 31 patients who had access to PD-1 ligand 1 (PD-L1) combined positive score (CPS) testing (cut-off value ≥1%), the ORR was not different between those who had PD-L1-positive (PD-L1+; 1/11, 9.1%) and PDL1-(2/20, 10.0%) tumors ( = 1.000). The tumor mutational burden (TMB) of PD-L1+ BTC was comparable to that of PD-L1-BTC ( = 0.630). TMB and any exonic somatic mutations were also not predictive of pembrolizumab response. Molecular analysis of blood and tumor samples demonstrated a relatively high natural killer (NK) cell proportion in the peripheral blood before pembrolizumab treatment in patients who achieved tumor response. Moreover, the tumors of these patients presented high enrichment scores for NK cells, antitumor cytokines, and Th1 signatures, and a low enrichment score for cancer-associated fibroblasts.Conclusions:This study shows the molecular characteristics associated with the efficacy of pembrolizumab in BTC of the MSS type.

Designing a Biomimetic Ornithopter Capable of Sustained and Controlled Flight

We describe the design of four ornithopters ranging in wing span from 10 cm to 40 cm, and in weight from 5 g to 45 g. The controllability and power supply are two major considerations, so we compare the efficiency and characteristics between different types of subsystems such as gearbox and tail shape. Our current omithopter is radio-controlled with inbuilt visual sensing and capable of takeoff and landing. We also concentrate on its wing efficiency based on design inspired by a real insect wing and consider that aspects of insect flight such as delayed stall and wake capture are essential at such small size. Most importantly, the advance ratio, controlled either by enlarging the wing beat amplitude or raising the wing beat frequency, is the most significant factor in an ornithopter which mimics an insect.

Thermoresponsive Al^(3+)-crosslinked poly(N-isopropylacrylamide)/alginate composite for green recovery of lithium from Li-spiked seawater

With the rapid increase in the demand for lithium as an energy-critical element,the recovery of Li+ions from seawater is a worldwide challenging ***,we propose a new facile and fast selective recovery approach of Li+using an Al^(3+)-crosslinked poly(N-isopropylacrylamide)(PNIPAAm)/alginate(Alg)(PNP/Alg(Al)) *** in situ TEM images indicate that Alg-Al^(3+)coordination is reorganized via the rearrangement of PNIPAAm and Alg networks,as the temperature *** reorganization eventually leads to the formation of polycrystalline *** in situ FTIR results exhibit that PNP/Alg(Al) composite has peculiar phase transitions,which includes a retrogressive phase change from hydrophobic to *** synergetic effect of the strong repulsion force of Al^(3+) ions and the attractive force of negatively charged polymeric chains enables the efficient adsorption of Li+ions with a low affinity from Li-spiked seawater.7.3% of Li+ions are recovered from Li-spiked seawater although the concentration of Li-spiked seawater is very *** addition,Li+ions can be extracted from PNP/Alg(Al) composite with the use of a small thermal *** proposed thermoresponsive IPN gel provides a strong potential in practical applications for Li+recovery as an innovative energy-material strategy.

Muscovite nanoparticles mitigate neuropathic pain by modulating the inflammatory response and neuroglial activation in the spinal cord

Despite numerous efforts to overcome neuropathic pain,various pharmacological drugs often fail to meet the needs and have many side *** is an aluminosilicate mineral that has been reported to have an anti-inflammatory effect,but the efficacy of muscovite for neuropathic pain has not been ***,we assessed whether muscovite nanoparticles can reduce the symptoms of pain by controlling the inflammatory process observed in neuropathic *** analgesic effects of muscovite nanoparticles were explored using partial sciatic nerve ligation model of neuropathic pain,in which one-third to onehalf of the nerve trifurcation of the sciatic nerve was tightly tied to the dorsal *** nanoparticles(4 mg/100μL)was given intramuscularly to evaluate its effects on neuropathic pain(3 days per week for 4 weeks).The results showed that the muscovite nanoparticle injections significantly alleviated partial sciatic nerve ligation-induced mechanical and cold *** the spinal cord,the muscovite nanoparticle injections exhibited inhibitory effects on astrocyte and microglia activation and reduced the expression of pro-inflammatory cytokines,such as interleukin-1β,tumor necrosis factor-α,interleiukin-6 and monocyte chemoattractant protein-1,which were upregulated in the partial sciatic nerve ligation ***,the muscovite nanoparticle injections resulted in a decrease in activating transcription factor 3,a neuronal injury marker,in the sciatic *** results suggest that the analgesic effects of muscovite nanoparticle on partial sciatic nerve ligation-induced neuropathic pain may result from inhibiting activation of astrocytes and microglia as well as pro-inflammatory *** propose that muscovite nanoparticle is a potential anti-nociceptive candidate for neuropathic *** experimental protocols in this study were approved by the Institutional Animal Ethics Committee(IACUC)at Dongguk University,South Korea(approval No.2017-022-1)

Dear Editor,Flowering is a major developmental switch in the life cycle of flowering plants. FRIGIDA （FRI） activates FLOWERING LOCUS C （FLC） transcription, a major repressor of this switch regulating flow- ering time （Johanson et al., 2000）. FRI is highly conserved and many natural variations in FRI are found to affect flowering time. However, little is known about the molecular characteristics of FRI.

Interface properties and phase formation between surface coated SKD61 and aluminum alloys

The intermediate phase formation and surface protection effects between SKD61 die mold alloys and aluminum alloys were investigated during a simulated die-casting *** surface coatings of SKD61 alloy were carried out via Si pack cementation coatings at 900 ℃ for 10 h and the ε-FeSi phase *** the coated SKD61 alloy was dipped in the liquid aluminum alloy(ALDC12),the surface coated SKD61 alloys showed better surface properties compared with uncoated SKD61 alloys,i.e.,the intermediate phases(FeSiAl compound) were not produced for the coated SKD61 *** coating layer of ε-FeSi served as a diffusion barrier for the formation of FeSiAl compounds.

The PRR-EC complex and SWR1 chromatin remodeling complex function cooperatively to repress nighttime hypocotyl elongation by modulating PIF4 expression in Arabidopsis

The circadian clock entrained by environmental light-dark cycles enables plants to fine-tune diurnal growth and developmental ***,we show that physical interactions among evening clock components,including PSEUDO-RESPONSE REGULATOR 5(PRR5),TIMING OF CAB EXPRESSION 1(TOC1),and the Evening Complex(EC)component EARLY FLOWERING 3(ELF3),define a diurnal repressive chromatin structure specifically at the PHYTOCHROME-INTERACTING FACTOR 4(PIF4)locus in *** three clock components act interdependently as well as independently to repress nighttime hypocotyl elongation,as hypocotyl elongation rate dramatically increased specifically at nighttime in the prr5-1 toc1-21 elf3-1 mutant,concomitantly with a substantial increase in PIF4 *** repression of PIF4 by ELF3,PRR5,and TOC1 is mediated by the SWI2/SNF2-RELATED(SWR1)chromatin remodeling complex,which incorporates histone H2A.Z at thePIF4 locus,facilitating robust epigenetic suppression ofPIF4 during the ***,these findings demonstrate that the PRR-EC-SWR1 complex represses hypocotyl elongation at night through a distinctive chromatin domain covering PIF4 chromatin.

High speed continuous turning of hardened steel with newly developed low content PCBN

In this study,newly developed low content PCBN was used to evaluate the cutting performance in high speed turning of hardened *** new PCBN and commercially available PCBN tipped inserts under the same cutting condition,cutting performance with variable cutting speed,feed rate and depth of cuts on tools were measured by observation of flank and crater *** microstructures were analyzed through SEM microscope,measurement of surface roughness on workpiece was also performed. According to cutting performance results,it is shown that new PCBN shows much longer tool life in high speed continuous turning than our conventional PCBN tools,with improved wear and chipping *** result on the machinability of new PCBN for hardened steel will provide effective guidelines to manufacturing engineers,also provide useful economic machining solution for high speed continuous turning for hardened steel.

Environmentally adaptive reshaping of plant photomorphogenesis by karrikin and strigolactone signaling

Coordinated morphogenic adaptation of growing plants is critical for their survival and propagation under fluctuating *** morphogenic responses to light and warm temperatures,termed photomorphogenesis and thermomorphogenesis,respectively,have been extensively studied in recent *** photomorphogenesis,plants actively reshape their growth and developmental patterns to cope with changes in light ***,photomorphogenesis is closely associated with diverse growth hormonal ***,accumulating evidence indicates that light-directed morphogenesis is profoundly affected by two recently identified phytochemicals,karrikins(KARs)and strigolactones(SLs).KARs and SLs are structurally related butenolides acting as signaling molecules during a variety of developmental steps,including seed *** receptors and signaling mediators have been identified,and associated working mechanisms have been explored using gene-deficient mutants in various plant *** particular interest is that the KAR and SL signaling pathways play important roles in environmental responses,among which their linkages with photomorphogenesis are most comprehensively studied during seedling *** this review,we focus on how the phytochemical and light signals converge on the optimization of morphogenic *** also discuss molecular mechanisms underlying the signaling crosstalks with an aim of developing potential ways to improve crop productivity under climate changes.

Effect of alkaline metal ions on the photoluminescence properties of Eu^(3+)-doped Ca_3Al_2O_6 phosphors

Ca3–xAl2O6：x Eu^3＋（0.02≤x≤0.10） and Ca（2.92）Al2O6：0.04Eu^3＋,0.04M^＋（M^＋=Li^＋, Na^＋, and K^＋） phosphors were prepared by sol-gel method and their photoluminescence properties were investigated. The crystal structure of the annealed Ca3–xAl2O6：x Eu^3＋ and Ca（2.92）Al2O6：0.04Eu^3＋,0.04M^＋ phosphors was analyzed with X-ray diffraction（XRD）. The size and morphological characteristics of the annealed phosphors were investigated with a field emission scanning electron microscope（FE-SEM）. The photoluminescence properties were evaluated with a spectrofluorometer. The phosphors were effectively excited by the light of 396 nm and showed intense red emission at 616 nm, which originated from the 5D0→7F2 transition of Eu^3＋. Among the prepared phosphors, the Ca（2.96）Al2O6：0.04Eu^3＋ phosphor showed the strongest emission intensity and the highest color purity. The addition of alkaline metal ions（Li^＋, Na^＋, and K^＋） to Ca（2.96）Al2O6：0.04Eu^3＋ phosphor degraded the emission intensity and color purity.
